AIMS: Cardiovascular manifestations are a major cause of mortality in Marfan syndrome (MFS). Animal models that mimic the syndrome and its clinical variability are instrumental for understanding the genesis and risk factors for cardiovascular disease in MFS. This study used morphological and ultrastructural analysis to the understanding of the development of cardiovascular phenotypes of the the mgΔloxPneo model for MFS. METHODS AND RESULTS: We studied 6-month-old female mice of the 129/Sv background, 6 wild type (WT) and 24 heterozygous animals from the mgΔloxPneo model. Descending thoracic aortic aneurysm and/or dissection (dTAAD) were identified in 75% of the MFS animals, defining two subgroups: MFS with (MFS+) and without (MFS-) dTAAD. Both subgroups showed increased fragmentation of elastic fibers, predominance of type I collagen surrounding the elastic fiber and fragmentation of interlaminar fibers when compared to WT. However, only MFS animals with spine tortuosity developed aortic aneurysm/dissection. The aorta of MFS+ animals were more tortuous compared to those of MFS- and WT mice, possibly causing perturbations of the luminal blood flow. This was evidenced by the detection of diminished aorta-blood flow in MFS+. Accordingly, only MFS+ animals presented a process of concentric cardiac hypertrophy and a significantly decreased ratio of left and right ventricle lumen area. CONCLUSIONS: We show that mgΔloxPneo model mimics the vascular disease observed in MFS patients. Furthermore, the study indicates role of thoracic spine deformity in the development of aorta diseases. We suggest that degradation of support structures of the aortic wall; deficiency in the sustenance of the thoracic vertebrae; and their compression over the adjacent aorta resulting in disturbed blood flow is a triad of factors involved in the genesis of dissection/aneurysm of thoracic aorta.

RESUMO

This study focused on the isolation and characterization of parvovirus in an infected dog in midwestern Brazil. Non-enveloped icosahedral parvovirus-like particles were isolated in CRFK cells and were allocated to a clade comprised of strains of CPV-2c, based on genome analysis. This is the first isolate of CPV-2c genomically characterized in Brazil.

Runting-stunting syndrome (RSS) is one of the diseases associated with many detected viruses. In Brazil, there were reports of several enteric disease outbreaks in chickens in which avian nephritis virus (ANV) was detected; however, the role of ANV in the outbreaks and whether the virus was a causative agent of these cases of enteric diseases were not determined. The aim of this study was to isolate ANV in specific pathogen-free (SPF) chicken embryonated eggs (CEE) from the enteric contents of chickens showing signs of RSS. For this purpose, 22 samples of chicken enteric contents that were positive only for ANV were inoculated into 7 and 14-day-old SPF-CEE via the yolk sac route and incubated for 5 d, with a total of 3 passages. Virus isolation was confirmed by the presence of embryo injuries, detection of viral RNA by RT-PCR, and visualization of viral particles using electron microscopy. Therefore, the 7-day-old inoculated embryos showed dwarfism, gelatinous consistency, hemorrhage, and edema in the embryos, whereas the 14-day-old did not show any alteration. Viral RNA was detected in the embryos of both ages of inoculation, and the same viral particles were visualized. The embryos from the mock group showed no alteration and were negative for all the tests. The viral cDNA was sequenced, and the molecular and phylogenetic analyses showed that the Brazilian isolates are more related with the ANV-1 serotype group; the sequences of these isolates showed a high percentage of nucleotide (86.4 to 94.9%) and amino acid (92.3 to 98.7%) similarity with other sequences from China, Japan, Australia, and the United States that belong to this serotype previously classified group. In this study, we isolated 8 samples of ANV in SPF-CEE from enteric content samples from chickens with RSS. In doing so, we showed the pathological injuries to the embryo caused by the virus and the molecular characterization of a part of the ORF 1b gene of the virus.

Iridoviruses of the Ranavirus genus have been implicated in the decline in amphibians worldwide, capable of affecting animals both in the wild and in captivity. This study aimed to detect iridovirus-like particles from three frog farms in southeastern Brazil using primary polyclonal antibodies, transmission electron microscopy (TEM) and histologic findings. The target organs were liver and kidneys. Sixty adults and sixty tadpoles of bullfrogs (Lithobates catesbeianus) were used in the study. TEM revealed the presence of iridovirus-like particles in hepatic tissue using the negative staining technique. Positive results were also observed by immunoelectron microscopy and immunocytochemistry (ICC). The histological analysis of the samples showed liver hemorrhage and corpuscles inclusion in hepatocytes as well as glomerulotubular degeneration and necrosis in the kidneys. The methods used in this study were highly efficient to detect the presence of iridovirus-like particles and possible infection of ranavirus.


Os iridovirus do gênero Ranavirus têm sido implicados no declínio dos anfíbios em todo o mundo afetando animais de vida livre e aqueles em cativeiro. O objetivo deste estudo foi detectar a presença de partículas semelhantes ao iridovírus em três ranários na região sudeste do Brasil, utilizando anticorpos policlonais primários, microscopia eletrônica de transmissão (MET) e achados histológicos. Os órgãos alvo foram o fígado e os rins. Sessenta rãs-touro adultas (Lithobates catesbeianus) e sessenta girinos da mesma espécie foram usados para o estudo. A MET revelou a presença de partículas semelhantes ao iridovírus em tecido hepático utilizando a técnica de contrastação negativa. Os resultados positivos foram também observados por imunomicroscopia eletrônica e imunocitoquímica. As análises histológicas nas mesmas amostras evidenciaram hemorragia no fígado e corpúsculos de inclusão em hepatócitos, e degeneração glomerulotubular e necrose nos rins. Os métodos usados neste estudo foram altamente eficientes na detecção de partículas semelhantes ao iridovírus e possivel infecção por ranavirus.

We have identified previously a panel of markers (C1orf24, ITM1 and PVALB) that can help to discriminate benign from malignant thyroid lesions. C1orf24 and ITM1 are specifically helpful for detecting a wide range of thyroid carcinomas, and PVALB is particularly valuable for detecting the benign Hürthle cell adenoma. Although these markers may ultimately help patient care, the current understanding of their biological functions remains largely unknown. In this article, we investigated whether PVALB is critical for the acquisition of Hürthle cell features and explored the molecular mechanism underlying the phenotypic changes. Through ectopic expression of PVALB in thyroid carcinoma cell lines (FTC-133 and WRO), we demonstrated that PVALB sequesters free cytoplasmic Ca(2+), which ultimately lowers calcium levels and precludes endoplasmic reticulum (ER) Ca(2+) refilling. These results were accompanied by induced expression of PERK, an ER stress marker. Additionally, forced expression of PVALB reduces Ca(2+) inflow in the mitochondria, which can in turn cause changes in mitochondria morphology, increase mitochondria number and alter subcellular localization. These findings share striking similarity to those observed in Hürthle cell tumors. Moreover, PVALB inhibits cell growth and induces cell death, most likely through the AKT/GSK-3ß. Finally, PVALB expression coincides with Ca(2+) deposits in HCA tissues. Our data support the hypothesis that the loss of PVALB plays a role in the pathogenesis of thyroid tumors.

BACKGROUND: Diarrhea in piglets directly affects commercial swine production. The disease results from the interaction of pathogens with the host immune system and is also affected by management procedures. Several pathogenic agents such as Campylobacter spp., Clostridium perfringens, Escherichia coli, Salmonella spp., group A rotavirus (RV-A), coronaviruses (transmissible gastroenteritis virus; porcine epidemic diarrhea virus), as well as nematode and protozoan parasites, can be associated with disease cases. RESULTS: All bacterial, viral, protozoan, and parasitic agents here investigated, with the exception of Salmonella spp. as well as both coronaviruses, were detected in varying proportions in piglet fecal samples, and positive animals were equally distributed between case and control groups. A statistically significant difference between case and control groups was found only for Cystoisospora suis (p = 0.034) and Eimeria spp. (p = 0.047). When co-infections were evaluated, a statistically significant difference was found only for C. perfringens ß2 and C. suis (p = 0.014). CONCLUSIONS: The presence of pathogens in piglets alone does not determine the occurrence of diarrhea episodes. Thus, the indiscriminate use of antibiotic and anthelminthic medication should be re-evaluated. This study also reinforces the importance of laboratory diagnosis and correct interpretation of results as well as the relevance of control and prophylactic measures.

Astroviruses have been associated with enteric disorders in many animal species, including chickens. Here, we describe the isolation, propagation, and pathological characteristics of chicken astrovirus (CAstV) in specific pathogen free (SPF) chicken embryonated eggs (CEE) from chickens with diarrhea and runting-stunting syndrome. The CEE were inoculated via the yolk sac route. Viral confirmation was carried out using PCR techniques and transmission electron microscopy negative staining with ammonium molybdate. The intestinal contents were screened for CAstV, and differential diagnostic testing was performed for avian nephritis virus, avian rotavirus, avian reovirus, chicken parvovirus, infectious bronchitis virus, and fowl adenovirus Group I to detect co-infection with other infectious agents. Seven- or 14-day-old CEEs presented with hemorrhages, edema, a gelatinous aspect, deformities, and dwarfism. The supporting membranes did not show any alterations. Here, we have described the isolation of CAstV and its pathological characteristics in SPF CEE.

O objetivo deste trabalho foi avaliar, por meio de microscopia óptica e eletrônica de transmissão, as alterações na morfologia e a viabilidade do desenvolvimento de embriões bovinos fecundados com sêmen contaminado experimentalmente à Escherichia coli produtora da toxina shiga stx2 (STEC). Para tanto, oócitos foram aspirados de ovários de vacas abatidas e selecionados para maturação in vitro. Após 20-24 horas de maturação, os oócitos foram divididos em 2 grupos. Sendo o primeiro grupo o controle (n = 418), fertilizado com sêmen testado e sem nenhum tipo de contaminante e o segundo, o grupo contaminado (n = 415), fertilizado com sêmen exposto a STEC. Cada sêmen foi tratado pela técnica de gradiente descontínuo de Percoll. Após o período de fecundação, os embriões foram avaliados quanto a sua morfologia e viabilidade, com o auxílio da microscopia óptica e eletrônica. Na ava liação morfológica, os oócitos fecundados com o sêmen contaminado apresentaram retração citoplasmática, falhas na divisão, assimetria de blastômeros, ooplasma granuloso, coloração castanho-escuro, formação de vacúolos, degeneração e rompimento da zona pelúcida. Essas alterações não foram observadas no grupo controle. A avaliação de todos oócitos incluídos mostrou taxas de clivagem de 70,3 e 52,8%, respectivamente, para embriões controle e contaminado (p = 0,0001). Após o 5° dia de desenvolvimento embrionário foram observadas 44,7% de mórulas no grupo controle e 22,4% no grupo contaminado, apresentando diferença significativa (p=0,0001). A presença da STEC interfere na taxa de clivagem dos embriões e também inviabiliza e provoca queda no desenvolvimento embrionário ao estádio de mórula, além de causar alterações morfológicas durante esse desenvolvimento.(AU)
