This study aimed to identify the Staphylococcus species responsible for bovine mastitis in dairy herds in northern Brazil, to investigate the capacity of biofilm production, and to analyze the association of biofilm production with multiresistance and intensity of California Mastitis Tests (CMT) reactions that can make treatment more difficult and cause misdiagnoses, respectively. Milk samples were collected from 23 dairy farms located in five municipalities in the state of Acre. A total of 339 crossbred cows were tested by CMT, with 109 animals (229 udder ceilings) reacting to the test. After bacterial isolation in blood agar, the catalase-positive and gram-positive cocci were submitted for identification by MALDI-TOF MS. Of 103 strains identified as staphylococci, Staphylococcus chromogenes (58.3%) and Staphylococcus aureus (19.4%) were the most prevalent species. Biofilm production was quantitatively evaluated using a microplate adherence test. Among the Staphylococcus strains, 71.8% were biofilm producers. Most strains of S. chromogenes (68.3%) had the capacity to produce biofilms, ranging from weak (43.3%), moderate (13.3%), and strong (11.7%) producers. Among S. aureus strains, 50% were non-biofilm producers, and none were strong producers. Our data showed an association between biofilm production capacity and multidrug resistance. In addition, there was a reduction in the response to the CMT test, which can mask the diagnosis.


O presente estudo teve como objetivos identificar as espécies de estafilococos envolvidas com mastite em rebanhos leiteiros no norte do Brasil, investigar a capacidade de produção de biofilme e analisar a associação da produção de biofilme com multirresistência e intensidade de reações de CMT que podem dificultar o tratamento e causar erros de diagnósticos, respectivamente. Amostras de leite foram coletadas em 23 propriedades leiteiras, situadas em cinco municípios do estado do Acre. Um total de 339 vacas mestiças foram testadas pelo CMT, sendo detectados 109 animais (229 tetos) reagentes ao teste. Após isolamento em ágar sangue, os cocos Gram positivos produtores de catalase foram submetidos à identificação por MALDI TOF MS. Um total de 103 estirpes foram identificadas como estafilococos, sendo mais prevalentes as espécies Staphylococcus chromogenes (58,3%) e S. aureus (19,4%). A produção de biofilme foi avaliada quantitativamente pelo teste de aderência em microplaca. Dentre todas as amostras de estafilococos, 71,8% foram produtoras de biofilme. A maioria das amostras de S. chromogenes (68,3%) apresentou capacidade de produzir biofilme, variando de fraco (43,3%), moderado (13,3%) a forte (11,7%) produtor. Entre as amostras de S. aureus, 50% não foram produtoras e nenhuma foi produtora forte. Essa capacidade de produção de biofilme mostrou-se associada à multirresistência a antimicrobianos, além de diminuir a intensidade no teste CMT, podendo mascarar o diagnóstico.

Chronic mild stress (CMS) is a widely accepted animal model relevant to depression that among other consequences, is chiefly known to induce anhedonia, often assessed as decreased preference for sucrose solution. CMS is also known to affect cognition, particularly memory tasks. In this study we have employed the multiple-trial inhibitory avoidance memory task (MTIA) to assess CMS effects on memory acquisition and retrieval. MTIA consists of repeated exposures to the unconditioned stimulus until a learning criterion is reached. Wistar rats underwent CMS for 5 weeks, and sucrose consumption was assessed once a week. At the end of CMS, animals were evaluated in the MTIA task. Overall decreased sucrose solution preference was highly variable. Further analyses showed that a subset of animals expressed resilience while another subset was sensitive to stress. CMS did not affect the number of acquisition sessions before reaching criterion or retrieval latency of MTIA task in neither sensitive nor resilient groups. Although tasks that assess learning ability in animal models relevant to depression indicate cognitive deficits, the ability to learn the association between compartment crossing and the aversive electric foot shock, which is strongly dependent on emotional aspects, was intact.

Sleep deprivation impairs performance in emotional memory tasks, however this effect on memory is not completely understood. Possible mechanisms may involve an alteration in neurotransmission systems, as shown by the fact that many drugs that modulate neural pathways can prevent memory impairment by sleep loss. Gastrin releasing peptide (GRP) is a neuropeptide that emerged as a regulatory molecule of emotional memory through the modulation of other neurotransmission systems. Thus, the present study addressed the effect of intraperitoneal (IP) administration of bombesin (BB) (2.5, 5.0 and 10.0µg/kg), a GRP agonist, on the performance of Wistar rats in a multiple trail inhibitory avoidance (MTIA) task, after sleep deprivation, using the modified multiple platforms method (MMPM). Sleep deprived animals exhibited acquisition and retention impairment that was not prevented by BB injection. In addition, non-sleep deprived animals treated with BB before and after the training session, but not before the test, have shown a retention deficit. In summary, BB did not improve the memory impairment by sleep loss and, under normal conditions, produced a memory consolidation deficit.

The replacement of the calcified cartilage by bone tissue during the endochondral ossification of the mandibular condyle is dependent of the resorbing activity of osteoclats. After partial resorption, calcified cartilage septa are covered by a primary bone matrix secreted by osteoblasts. Osteoadherin (OSAD) is a small proteoglycan present in bone matrix but absent in cartilage during the endochondral ossification. The aim of this study was to analyze the effect of alendronate, a drug known to inhibit bone resorption by osteoclasts, on the endochondral ossification of the mandibular condyle of young rats, by evaluating the distribution of osteoclasts and the presence of OSAD in the bone matrix deposited. Wistar newborn rats (n=45) received daily injections of alendronate (n=27) or sterile saline solution as control (n=18) from the day of birth until the ages of 4, 14 and 30 days. At the days mentioned, the mandibular condyles were collected and processed for transmission electron microscopy analysis. Specimens were also submitted to tartrate resistant acid phosphatase (TRAP) histochemistry and ultrastructural immunodetection of OSAD. Alendronate treatment did not impede the recruitment and fusion of osteoclasts at the ossification zone during condyle growth, but they presented inactivated phenotype. The trabeculae at the ossification area consisted of cartilage matrix covered by a layer of primary bone matrix that was immunopositive to OSAD at all time points studied. Apparently, alendronate impeded the removal of calcified cartilage and maturation of bone trabeculae in the mandibular ramus, while in controls they occurred normally. These findings highlight for giving attention to the potential side-effects of bisphosphonates administered to young patients once it may represent a risk of disturbing maxillofacial development.

Essential hypertension is a complex and multifactorial trait; genetic and environmental factors interact to produce the final phenotype. Studies have demonstrated association of hypertension with varied gene polymorphisms. However, demonstration of common genetic causes in the general population remains elusive. We investigated a possible association between hypertension and haptoglobin, angiotensin I-converting enzyme (ACE), glutathione S-transferases GSTM1 and GSTT1, MnSOD (Val9Ala), CAT (-21A/T), and GPX1 (Pro198Leu) gene polymorphisms in an urban Brazilian population group from Brasília. Although ACE has been reported to be one of the main polymorphisms associated with hypertension, we found no association with ACE's specific genotypes. However, a possible association with Hp1-1 and MnSOD Val/Ala genotypes suggests that, at least for the Brazilian population, polymorphisms related to oxidative stress should be more deeply investigated.
