RESUMO

The exact accuracy of estimated breeding values can be calculated based on the prediction error variances obtained from the diagonal of the inverse of the left-hand side (LHS) of the mixed model equations (MME). However, inverting the LHS is not computationally feasible for large datasets, especially if genomic information is available. Thus, different algorithms have been proposed to approximate accuracies. This study aimed to: 1) compare the approximated accuracies from 2 algorithms implemented in the BLUPF90 suite of programs, 2) compare the approximated accuracies from the 2 algorithms against the exact accuracy based on the inversion of the LHS of MME, and 3) evaluate the impact of adding genotyped animals with and without phenotypes on the exact and approximated accuracies. Algorithm 1 approximates accuracies based on the diagonal of the genomic relationship matrix (G). In turn, algorithm 2 combines accuracies with and without genomic information through effective record contributions. The data were provided by the American Angus Association and included 3 datasets of growth, carcass, and marbling traits. The genotype file contained 1,235,930 animals, and the pedigree file contained 12,492,581 animals. For the genomic evaluation, a multi-trait model was applied to the datasets. To ensure the feasibility of inverting the LHS of the MME, a subset of data under single-trait models was used to compare approximated and exact accuracies. The correlations between exact and approximated accuracies from algorithms 1 and 2 of genotyped animals ranged from 0.87 to 0.90 and 0.98 to 0.99, respectively. The intercept and slope of the regression of exact on approximated accuracies from algorithm 2 ranged from 0.00 to 0.01 and 0.82 to 0.87, respectively. However, the intercept and the slope for algorithm 1 ranged from -0.10 to 0.05 and 0.98 to 1.10, respectively. In more than 80% of the traits, algorithm 2 exhibited a smaller mean square error than algorithm 1. The correlation between the approximated accuracies obtained from algorithms 1 and 2 ranged from 0.56 to 0.74, 0.38 to 0.71, and 0.71 to 0.97 in the groups of genotyped animals, genotyped animals without phenotype, and proven genotyped sires, respectively. The approximated accuracy from algorithm 2 showed a closer behavior to the exact accuracy when including genotyped animals in the analysis. According to the results, algorithm 2 is recommended for genetic evaluations since it proved more precise.


The genomic estimated breeding value (GEBV) represents an animal's genetic merit calculated using a combination of phenotypes, pedigree, and genomic information through a procedure known as single-step genomic best linear unbiased prediction (ssGBLUP). The accuracy of a GEBV reflects how closely it correlates with the true breeding value. However, calculating accuracies is not computationally feasible for large datasets with genomic information. In this context, methods for approximating accuracies have been proposed and implemented into genetic evaluations. This study aimed to compare 2 algorithms to approximate accuracies for ssGBLUP. In algorithm 1, genomic contributions are based on the diagonal of the genomic relationship matrix (G), combined with contributions from animal records and pedigrees. In turn, algorithm 2 combines accuracies with and without genomic information through effective record contributions. The data for this study were provided by the American Angus Association and included datasets of growth, carcass, and marbling traits. Genotypes were available for 1,235,930 animals, and the pedigree had 12,492,581 animals. We showed that algorithm 2 is better suited for approximating accuracies, as its approximations closely matched the exact accuracy values obtained from the inverse of the mixed model equations.

BACKGROUND: Plasmodium falciparum malaria is a public health issue mostly seen in tropical countries. Until now, there is no effective malaria vaccine against antigens specific to the blood-stage of P. falciparum infection. Because the pathogenesis of malarial disease results from blood-stage infection, it is essential to identify the most promising blood-stage vaccine candidate antigens under natural exposure to malaria infection. METHODS: A cohort of 400 pregnant women and their infants was implemented in South Benin. An active and passive protocol of malaria surveillance was established during pregnancy and infancy to precisely ascertain malaria infections during the follow-up. Twenty-eight antibody (Ab) responses specific to seven malaria candidate vaccine antigens were repeatedly quantified during pregnancy (3 time points) and infancy (6 time points) in order to study the Ab kinetics and their protective role. Abs were quantified by ELISA and logistic, linear and cox-proportional hazard model were performed to analyse the associations between Ab responses and protection against malaria in mothers and infants, taking into account socio-economic factors and for infants an environmental risk of exposure. RESULTS: The levels of IgM against MSP1, MSP2 and MSP3 showed an early protective response against the onset of symptomatic malaria infections starting from the 18th month of life, whereas no association was found for IgG responses during infancy. In women, some IgG responses tend to be associated with a protection against malaria risk along pregnancy and at delivery, among them IgG3 against GLURP-R0 and IgG2 against MSP1. CONCLUSION: The main finding suggests that IgM should be considered in vaccine designs during infanthood. Investigation of the functional role played by IgM in malaria protection needs further attention.

Background: Hamstrings injuries are a major concern in football (soccer), affecting both recreational players and professional athletes. Although being a recognized issue within the football community, its incidence has been increasing over the last years and still poses a challenge to all practitioners involved. Study objectives and rationale: The goal of this narrative review is to outline hamstrings injuries epidemiology and mechanisms of injury, identify and discuss its risk factors, provide an approach to a proper early diagnosis, evaluate the efficacy of current treatment options and return to sports, and present the best strategies for hamstrings injury prevention. These guidelines will help the sports medicine staff team on how to better manage their players with or at risk of hamstrings injuries. Conclusion: Despite several breakthroughs in research of hamstrings injuries, there is still heterogeneity across studies and lack of consensus in regards to classification, diagnosis, treatment and prevention. Hamstrings injuries compromise the athlete's performance with time loss due to injury, shortens their highest-level career longevity with higher risk of reinjury rates, and is a defying problem for clubs to balance financial losses due to having their players off the pitch. Further research is warranted to keep moving forward with evidence on treating and preventing hamstrings injuries to mitigate its high incidence and keep the players safe.

ABSTRACT Objective: To determine the preoperative radiographic method for measuring the Cobb angle that is closest to the postoperative result in patients with scoliotic deformity. Methods: Retrospective cohort study of radiographic spinal evaluation (preoperative posteroanterior (PA), bending, traction, traction under anesthesia and immediate postoperative posteroanterior (PO)) of 26 patients treated surgically for scoliotic deformities during the period from January 2017 to September 2019. The final mean Cobb angle and its decrease in relation to the PA value were evaluated in the three curves in patients with idiopathic (IS) and non-idiopathic scoliosis. Results: All the mean curve values were statistically significant, except for bending in non-idiopathic scoliosis (non-IS). The mean traction under anesthesia values were closer to the PO values. Regarding the delta (decrease) of the maneuvers in relation to the PA, no statistical significance was observed in the non-IS group. The traction under anesthesia maneuver had a greater delta in all curves. Conclusions: The traction under anesthesia maneuver in patients with idiopathic scoliosis is the method with the greatest flexibility and which best predicts the postoperative result. RESUMO Objetivo: Objetivo: Determinar o método radiográfico pré-operatório para aferição do ângulo de Cobb que mais se aproxima dos resultados pós-operatórios em pacientes com deformidade escoliótica. Métodos: Estudo de coorte retrospectivo de avaliação radiográfica da coluna vertebral (posteroanterior pré-operatória [PA], inclinações, tração, tração com anestesia e posteroanterior pós-operatória imediata [PO]) de 26 pacientes com deformidades escolióticas no período de Janeiro de 2017 a Setembro de 2019 tratados com cirurgia . Avaliou-se a média final do ângulo de Cobb e a sua diminuição com relação ao PA nas três curvas em pacientes com escoliose idiopática (EI) e não idiopática. Resultados: Todas as médias das curvas têm significância estatística, exceto a inclinação na escoliose não idiopática (não EI). A tração com anestesia apresenta média de valores mais próximos ao PO. Com relação ao delta (diminuição) das manobras referentes ao PA, foi observado que não houve significância estatística nas não EI. A manobra de tração com anestesia tem delta maior em todas as curvas. Conclusões: A manobra de tração com anestesia em pacientes com escoliose idiopática configura-se como o método com maior flexibilidade e que melhor prediz o resultado pós-operatório. La maniobra de tracción con anestesia tiene un delta mayor en todas las curvas. Conclusiones: La maniobra de tracción con anestesia en pacientes con escoliosis idiopática se configura como el método con mayor flexibilidad y que mejor predice el resultado postoperatorio.Nivel de evidencia III; Estudio diagnóstico.

The ultimate goal of genetic selection is to improve genetic progress by increasing favorable alleles in the population. However, with selection, homozygosity, and potentially harmful recessive alleles can accumulate, deteriorating genetic variability and hampering continued genetic progress. Such potential adverse side effects of selection are of particular interest in populations with a small effective population size like the Romosinuano beef cattle in Mexico. The objective of this study was to evaluate the genetic background and inbreeding depression in Mexican Romosinuano cattle using pedigree and genomic information. Inbreeding was estimated using pedigree (FPED) and genomic information based on the genomic relationship matrix (FGRM) and runs of homozygosity (FROH) of different length classes. Linkage disequilibrium (LD) was evaluated using the correlation between pairs of loci, and the effective population size (Ne) was calculated based on LD and pedigree information. The pedigree file consisted of 4875 animals born between 1950 and 2019, of which 71 had genotypes. LD decreased with the increase in distance between markers, and Ne estimated using genomic information decreased from 610 to 72 animals (from 109 to 1 generation ago), the Ne estimated using pedigree information was 86.44. The reduction in effective population size implies the existence of genetic bottlenecks and the decline of genetic diversity due to the intensive use of few individuals as parents of the next generations. The number of runs of homozygosity per animal ranged between 18 and 102 segments with an average of 55. The shortest and longest segments were 1.0 and 36.0 Mb long, respectively, reflecting ancient and recent inbreeding. The average inbreeding was 2.98 ± 2.81, 2.98 ± 4.01, and 7.28 ± 3.68% for FPED, FGRM, and FROH, respectively. The correlation between FPED and FGRM was -0.25, and the correlations among FPED and FROH of different length classes were low (from 0.16 to 0.31). The correlations between FGRM and FROH of different length classes were moderate (from 0.44 to 0.58), indicating better agreement. A 1% increase in population inbreeding decreased birth weight by 0.103 kg and weaning weight by 0.685 kg. A strategy such as optimum genetic contributions to maximize selection response and manage the long-term genetic variability and inbreeding could lead to more sustainable breeding programs for the Mexican Romosinuano beef cattle breed.

Saturated solutions of calcium l-lactate in water or in deuterium oxide continuously dissolve calcium l-lactate by addition of solid sodium d-gluconate and become strongly supersaturated in calcium d-gluconate due to no or slow precipitation. The quantification of total dissolved calcium allied with the calcium complexes equilibrium constants allowed an ion speciation, which shows an initial non-thermal and spontaneous supersaturation of more than a factor of 50 at 25 °C only slowly decreasing after initiation of precipitation of calcium d-gluconate after a lag phase of several hours. A mathematical model is proposed, based on numerical solution of coupled differential equations of dynamics of l-lactate and d-gluconate exchange during the lag phase for precipitation and during precipitation. A slow exchange of l-lactate coordinated to calcium with d-gluconate is indicated with a time constant of 0.20 h-1 in water and of 0.15 h-1 in deuterium oxide and a kinetic deuterium/hydrogen isotope effect of 1.25. Such spontaneous non-thermal supersaturation and slow ligand exchange with a pseudo first order equilibration process with a half-life of 3.5 h in water for calcium hydroxycarboxylates can help to understand the higher calcium bioavailability from calcium hydroxycarboxylates compared to simple salts.

Human leukocyte antigen-C (HLA-C) is a classical HLA class I molecule that binds and presents peptides to cytotoxic T lymphocytes in the cell surface. HLA-C has a dual function because it also interacts with Killer-cell immunoglobulin-like receptors (KIR) receptors expressed in natural killer and T cells, modulating their activity. The structure and diversity of the HLA-C regulatory regions, as well as the relationship among variants along the HLA-C locus, are poorly addressed, and few population-based studies explored the HLA-C variability in the entire gene in different population samples. Here we present a molecular and bioinformatics method to evaluate the entire HLA-C diversity, including regulatory sequences. Then, we applied this method to survey the HLA-C diversity in two population samples with different demographic histories, one highly admixed from Brazil with major European contribution, and one from Benin with major African contribution. The HLA-C promoter and 3'UTR were very polymorphic with the presence of few, but highly divergent haplotypes. These segments also present conserved sequences that are shared among different primate species. Nucleotide diversity was higher in other segments rather than exons 2 and 3, particularly around exon 5 and the second half of the 3'UTR region. We detected evidence of balancing selection on the entire HLA-C locus and positive selection in the HLA-C leader peptide, for both populations. HLA-C motifs previously associated with KIR interaction and expression regulation are similar between both populations. Each allele group is associated with specific regulatory sequences, reflecting the high linkage disequilibrium along the entire HLA-C locus in both populations.

ABSTRACT Objective To compare the Schanz screw insertion angle and the loss of the regional kyphosis correction in thoracolumbar burst fractures following posterior short instrumentation surgery. Methods Patients with a thoracolumbar burst fracture between levels T11-L2 were divided into two groups (parallel and divergent) according to the angle formed between the Schanz screw and the vertebral plateau. Regional kyphosis was evaluated in preoperative, immediate postoperative and last follow-up radiographs. Results Of the 58 patients evaluated, 31 had a parallel assembly and 27 had a divergent assembly. When we analyzed the angle of kyphosis, no statistical difference was observed between the pre- and postoperative radiographs. However, a statistical difference in the last follow-up radiographs and in the final loss of the kyphosis correction was confirmed. Conclusion The insertion of Schanz screws with a divergent assembly presents better radiographic results with less loss of kyphosis correction angle when compared with the parallel assembly technique. RESUMO Objetivo Comparar o ângulo de inserção do pino de Schanz e os resultados da perda de correção da cifose regional nas fraturas toracolombares do tipo explosão após tratamento cirúrgico com instrumentação curta por via posterior. Métodos Os pacientes com fratura toracolombar do tipo explosão entre os níveis de T11-L2 foram divididos em dois grupos (paralelo e divergente) de acordo com o ângulo formado entre o pino de Schanz e o platô vertebral. Foi avaliada a cifose regional nas radiografias pré-operatória, pós-operatória imediata e do último acompanhamento. Resultados Dos 58 pacientes avaliados, 31 apresentaram uma montagem paralela e 27 uma montagem divergente. Ao analisarmos o ângulo da cifose, não se observou diferença estatística nas radiografias pré- e pós-operatória imediata. Porém, verificou-se uma diferença estatística nas radiografias do último acompanhamento e na perda final de correção da cifose. Conclusões A inserção do pino de Schanz com uma montagem divergente apresenta melhores resultados radiográficos com menor perda do ângulo de correção da cifose quando comparada com a técnica de montagem paralela. RESUMEN Objetivo Comparar el ángulo de inserción del tornillo de Schanz y los resultados de la pérdida de corrección de la cifosis regional en las fracturas toracolumbares del tipo explosión después del tratamiento quirúrgico con instrumentación corta por vía posterior. Métodos Los pacientes con fractura toracolumbar del tipo explosión entre los niveles de T11-L2 fueron divididos en dos grupos (paralelo y divergente) de acuerdo con el ángulo formado entre el tornillo de Schanz y la meseta vertebral. Fue evaluada la cifosis regional en las radiografías preoperatoria, posoperatoria inmediata y del último acompañamiento. Resultados De los 58 pacientes evaluados, 31 presentaron un montaje paralelo y 27 un montaje divergente. Al analizar el ángulo de la cifosis, no se observó diferencia estadística en las radiografías pre y postoperatoria inmediata. Sin embargo, se verificó una diferencia estadística en las radiografías del último acompañamiento y en la pérdida final de corrección de la cifosis. Conclusiones La inserción del tornillo de Schanz con un montaje divergente presenta mejores resultados radiográficos con menor pérdida del ángulo de corrección de la cifosis cuando comparada con la técnica de montaje paralelo. ABSTRACT Objective To correlate magnetic resonance imaging (MRI) findings with the microbiological and anatomopathological diagnosis of spinal infection. Methods A retrospective, cohort review of online medical records (laboratory, anatomopathology and diagnostic imaging sector) of patients diagnosed with spondylodiscitis, who underwent a full spine MR scan between January 2014 and July 2018 at the Department of Orthopedics and Traumatology of the Universidade Federal de São Paulo. Results Staphylococcus aureus was the most commonly found etiological agent (57%). Blood culture was positive in 76% of cases and 82% of the patients who underwent biopsy had a spondylodiscitis diagnosis. Pain was the most prevalent clinical symptom and the lumbosacral spine was the most frequent site of infection. T1 hyposignal, T2/STIR hypersignal, and terminal plate destruction were verified in almost all MR scans. Conclusions No direct correlation was found between MR findings and any specific etiological agent. Blood culture and biopsy are important diagnostic tools that should be used for accurate diagnosis of the infectious agent . Level of evidence IV; Diagnostic Study.


RESUMO Objetivo Correlacionar os achados de ressonância magnética (RNM) com o diagnóstico microbiológico e anatomopatológico de infecção na coluna vertebral. Métodos Estudo de coorte retrospectivo de revisão de prontuários online (laboratório, anatomopatológico e setor de diagnóstico por imagem) de pacientes com diagnóstico de espondilodiscite, submetidos ao exame de RNM da coluna vertebral e acompanhados pelo Departamento de Ortopedia e Traumatologia da Universidade Federal de São Paulo, entre janeiro de 2014 e julho de 2018. Resultados O agente etiológico mais comum encontrado foi o S. aureus (57%). A hemocultura mostrou-se positiva em 76% dos casos e 82% dos pacientes submetidos à biópsia apresentaram diagnóstico de espondilodiscite. A dor foi o achado clínico mais prevalente e a coluna lombossacra foi o sítio mais frequente de infecção. No exame de RNM, a presença de hipossinal em T1, hipersinal em T2/STIR e destruição das placas terminais foram identificados em quase todos os casos. Conclusões Não houve correlação direta dos achados na RNM com um agente etiológico específico na espondilodiscite. A hemocultura e a biópsia são ferramentas diagnósticas importantes que devem ser utilizadas para o diagnóstico preciso do agente infeccioso. Nível de evidência IV; Estudo diagnóstico.

Métodos Un estudio de cohorte retrospectivo de revisión de prontuarios en línea (laboratorio, anatomopatológico y sector de diagnóstico por imagen) de pacientes con diagnóstico de espondilodiscitis, sometidos al examen de RNM de la columna vertebral y acompañados por el Departamento de Ortopedia y Traumatología de la Universidad Federal de São Paulo, entre enero de 2014 y julio de 2018. Resultados El agente etiológico más común encontrado fue el S. aureus (57%). El hemocultivo se mostró positivo en 76% de los casos y 82% de los pacientes sometidos a biopsia presentaron diagnóstico de espondilodiscitis. El dolor fue el hallazgo clínico más prevalente y la columna lumbosacra fue el sitio más frecuente de infección. En el examen de RNM, la presencia de hiposeñal en T1, hiperseñal en T2/STIR y destrucción de las placas terminales fueron identificadas en casi todos los casos. Conclusiones No hubo correlación directa de los hallazgos de la RNM con un agente etiológico específico en la espondilodiscitis. El hemocultivo y la biopsia son herramientas diagnósticas importantes, que deben ser utilizadas para el diagnóstico preciso del agente infeccioso. Nivel de evidencia IV; Estudio Diagnóstico.

RESUMO Introdução: O uso de próteses totais tem impacto na qualidade de vida relacionada à saúde bucal e os idosos são uma parcela da população a ser pesquisada sob essa perspectiva. Objetivo: Avaliar a autopercepção do impacto do uso de próteses totais na qualidade de vida relacionada à saúde bucal de idosos institucionalizados. Métodos: Participaram 20 idosos do Lar dos Idosos Recanto do Tarumã, Curitiba, Paraná, Brasil. Foram avaliados aspectos demográficos, econômicos, de hábitos e clínicos, juntamente com aplicação do questionário Geriatric Oral Health Assessment Index. Os dados foram submetidos à análise descritiva e bivariada (Teste do Qui-Quadrado), com significância de 5 porcento. Resultados: A média de idade foi de 75,2 anos (DP= 8,8) e o tempo médio de uso de prótese foi de 27,9 anos (DP= 18,5). Renda inferior a um salário mínimo foi indicada por 80,0 porcento, e visitas ao cirurgião-dentista pela última vez há mais de um ano, por 70,0 porcento. Os piores relatos foram referentes à mastigação, representados por problemas para mastigar alimentos e desconforto ao comer, com 25 porcento de respostas positivas em cada questão; a soma das frequências das respostas "às vezes" e "sempre" quanto à insatisfação ou à infelicidade com a aparência da boca foi reportada 60 porcento dos pesquisados. A retenção insatisfatória da prótese inferior se deu em 50,0 porcento casos, enquanto os problemas com oclusão estiveram presentes também em metade da amostra. Os defeitos estiveram presentes em 70,0 porcento das próteses superiores e 45,0 porcento das inferiores. Não foi encontrada associação significativa (p> 0,05) entre as variáveis independentes e o desfecho. Conclusões: Os idosos avaliados, independentemente das condições das próteses totais, relataram qualidade de vida relacionada à saúde bucal satisfatória(AU)


RESUMEN Introducción: El uso de prótesis totales tiene un impacto en la calidad de vida relacionada con la salud bucal, y los ancianos son parte de la población que se investigará desde esta perspectiva. Objetivo: Evaluar la autopercepción del impacto del uso de prótesis totales en la calidad de vida relacionada con la salud bucal de ancianos institucionalizados. Métodos: Participaron 20 ancianos de "Lar dos Idosos Recanto do Tarumã", Curitiba, Paraná, Brasil. Los aspectos demográficos, económicos, de hábitos y clínicos se evaluaron junto con la aplicación del cuestionario Geriatric Oral Health Assessment Index. Los datos fueron sometidos a análisis descriptivo y bivariado (prueba de chi cuadrado), con significación del 5 por ciento. Resultados: La edad media fue de 75,2 años (DE= 8,8) y el tiempo medio de uso de la prótesis fue de 27,9 años (DE= 18,5). Los ingresos menores a un salario mínimo se indicaron en el 80,0 por ciento, y las visitas al cirujano dental la última vez, hace más de un año, en el 70, 0 por ciento. Los peores informes estaban relacionados con la masticación, representados por problemas para masticar los alimentos y la incomodidad al comer, con el 25 por ciento de respuestas positivas en cada pregunta. La suma de las frecuencias de las respuestas "a veces" y "siempre" sobre la insatisfacción o la infelicidad con la apariencia de la boca se informó en el 60 por ciento de los encuestados. ABSTRACT Introduction: The use of complete dentures has an impact on oral health-related quality of life. Elderly people are the population sector to be researched into from this perspective. Objective: Evaluate the self-perception of the impact of the use of complete dentures on the oral health-related quality of life of institutionalized elderly people. Methods: The study sample was 20 elderly people from Lar dos Idosos Recanto do Tarumã, Curitiba, Paraná, Brazil. Evaluation of demographic and economic details, habits and clinical aspects was based on the information collected with the Geriatric Oral Health Assessment Index questionnaire. The data were subjected to descriptive and bivariate analysis (Chi-squared test), with a significance of 5 percent. Results: Mean age was 75.2 (SD = 8.8) years and mean time of denture use was 27.9 (SD = 18.5) years. Income was below minimum wage in 80.0 percent of the participants, whereas the last visit to the dentist had occurred more than a year ago in 70.0 percent. The worst reports had to do with chewing, represented by problems to chew food and discomfort when eating, with 25 percent positive answers to each question. The sum of the frequencies of the answers "sometimes" and "always" about dissatisfaction with or unhappiness about the appearance of the mouth was reported by 60 percent of the respondents. Unsatisfactory retention of the lower denture occurred in 50.0 percent of the cases, whereas occlusion problems were present in half of the sample. Defects were present in 70.0 percent of the upper dentures and 45.0 percent of the lower dentures. No significant association (p> 0.05) was found between independent variables and outcome. Conclusions: The elderly people evaluated, regardless of the conditions of their complete dentures, reported satisfactory oral health-related quality of life(AU)
