While normal human eosinophils are destroyed in vitro by virulent Entamoeba histolytica, notwithstanding the presence of antibodies and complement, activated eosinophils promptly destroy the parasite although dying also at the end of the process. To study the possible in vivo participation of eosinophils in invasive amebiasis, we compared the induction of experimental amebic abscess of the liver (AAL) in gerbils (Meriones unguiculatus) previously made eosinophilic through Toxocara canis antigen injection and in normal control gerbils. After intraportal inoculation of 10(5) ameba trophozoites (6 and 24 hr), the ratio of gerbils with AAL, as well as the number and size of the microabscesses was comparable in eosinophilic and control gerbils. However, at 96 hr the number and size of the microabscesses were significantly smaller (p < 0.05) in eosinophilic gerbils. On the other hand the actuarial AAL survival curve up to 45 days post-amebic inoculation was significantly (p < 0.05) shifted to the right in controls. These results suggest that antigen-induced eosinophilia may exert a protective effect against AAL in gerbils.

The increase of HLA-DR3 and complotype SCO1 previously found in Mexican mestizo adults with E. histolytica amoebic abscess of the liver, was also found in Mexican mestizo children of either sex with the same disease, when compared to the healthy control population (adults and/or children) of the same ethnic and socioeconomic background. This HLA and complotype pattern was not found in Mexican Mestizo patients with amoebic rectocolitis. No linkage disequilibrium was found between these and the other MHC determinants tested in this survey. Thus, HLA-DR3 and SCO1 may constitute primary, independent risk factors, not for any kind of amoebic tissue invasion (i.e. amoebic rectocolitis), but specifically for amoebic liver abscess, irrespective of age or sex. The possibility of linkage disequilibrium with other factors (i.e. the TNF family) within or close to the MHC that were not tested in this study, is discussed. Children with amoebic liver abscess revealed a significant increase in HLA-DR5, and the absence of HLA-DR6 when compared to adults with amoebic liver abscess, suggesting that at least in this ethnic group these class II HLA traits may contribute to some of the peculiarities of pediatric amoebic liver abscess as opposed to the adult version of this disease. HLA-DR3, SCO1, but also HLA-DR5 and HLA-DR6 have all been associated with certain forms of immune-dysfunction, and may thus contribute to some of the clinical and immunological features of this parasitic disease.

RESUMO

Mexican mestizo pediatric patients with ALA revealed a significantly increased frequency of HLA-DR3 alone, or in its haplotype form HLA-A2, DR3, which confirms our previous observation in adult patients with ALA in the same ethnic group. However, the relative increase in these HLA specificities in pediatric ALA patients when compared to adult patients was not statistically significant, and calls for an enlargement of the population studied.

The incidence of incongruent parenthood was determined by confronting the declared familial relationship of 98 father-mother-son/daughter trinomials (drawn from the Centro Médico Nacional-IMSS (CMN-IMSS) Kidney Transplant Program) with their HLA-A and B antigens. Fourteen (14.3%) discrepant cases were found: three each with the putative father (3.1%) or with the putative mother (3.1%), two with either father or mother (though impossible to determine precisely with whom) (2.0%), and six with both parents (6.1%). Several possible explanations of these findings were considered, such as out-of-wedlock pregnancies, complex known or unknown, declared or undeclared social circumstances frequently related to the population studied (i.e. imminent kidney transplantation) and technical pitfalls. These data underscore the level of possible "noise" and imprecision in population genetics; genetic-epidemiology and transplantation programs.

Even though eosinophiles are not characteristic of amebiasis, polymorphonuclear eosinophilic cells are regularly found in the inflammatory lesion which occurs in the early phases of amebic invasion. To acquire better knowledge of the intervention of these cells when confronted with E. histolytica, testicular lesions were produced by means of direct injection of amebas in rats with eosinophilia. Eosinophilia (greater than or equal to 4%) was induced in male Sprague Dawley rats by an intravenous injection of Sephadex G200 suspension. Later, both the eosinophilic and normal (control) rats were challenged with an intratesticular injection (on the left side) of 2 x 10(6) amebas of the virulent strain HM1-IMSS. The other testicle was injected with BFS-A to serve as control. The testicles were removed 5 hours later and evaluated histologically. The size and celullarity of the testicular lesions produced by the ameba on both eosinophilic and controls were similar, whereas lesions were not found in testicles injected with BSF-A. Within the limits of our experimental model, the results suggest that eosinophils do not contribute substantially to the genesis of tissue lesions produced by E. histolytica.

Las poblaciones de linfocitos T. (activos y totales) y B en 21 pacientes con psoriasis activa no fueron diferentes a las encontradas en 25 sujetos sanos. Es posible que las alteraciones en estos aspectos descritas ocasionalmente en la literatura se deban a diferencias metodologicas, o a la inclusion de pacientes con grados intensos de actividad epidermica, tratamiento con esteroides y presencia de artropatias en algunas series estudiadas
