We investigated the egg production, changes in luteinizing hormone (LH), follicle-stimulating hormone (FSH), gonadal hormones, and their mRNA levels in the hypothalamic-pituitary-gonadal axis of White King pigeons submitted to different photoperiods. The treatments consisted of three photoperiods (8 h light (L):16 h dark (D), 12L:12D, and 16L:8D), with three replicates of twelve pairs of adult pigeons. The birds were exposed the photoperiods for 45 days. Egg production performance was recorded daily. Six pigeon pairs per replicate were selected for plasma collection, and six pigeon pairs per replicate for the resection of the hypothalamic-pituitary-gonadal (HPG) axis. Egg production was significantly improved by long-day lighting (16L:8D), while no differences in egg shape index were detected. Higher average egg weight was obtained in 16L:8D group, whereas broken egg percentage was higher in the 8L:16D group. Female LH level was significantly higher in long-day lighting, and the FSH level significantly lower in short-day lighting. The females in the 16L:8D group had higher estrogen level. The photoperiods had a minor effect on plasma LH and testosterone in males, whereas the FSH level was significantly higher in the 16L:8D group. The level of LH mRNA expression was higher in both females and males of the 16L:8D group. Similar trends in FSH mRNA expression observed in both females and males. The 16L:8D photoperiod not only improved egg production, but also stimulated plasma LH, FSH, gonadal hormones, and promoted LH and FSH mRNA expression in pigeons.(AU)

DNA methylation is an important epigenetic modification in eukaryotes, which plays a significant role in regulating gene expression. When the host is invaded by the influenza virus, gene expression is regulated via changes in DNA methylation levels or patterns, leading to the activation or suppression of relevant signaling pathways or networks, triggering a series of immune responses against viral invasion. Here, we investigated the changes in genomic DNA methylation in the immune organs of chicken infected with H5N1 influenza virus. Genome-wide DNA methylation levels in the spleen, thymus, and bursa of Fabricius of specific pathogen-free (SPF) chicken infected with the Guangdong (G-H5N1) and Anhui (A-H5N1) H5N1 strains, and water (control) were analyzed by fluorescence-labeled methylation-sensitive amplified polymorphism (F-MSAP). The results indicated that total DNA methylation levels did not differ between spleen genomic DNA in chicken treated with different viral strains and the control (P > 0.05). However, the total DNA methylation levels were significantly upregulated in the thymus (P < 0.01) and bursa (P < 0.05) of chicken in the A-H5N1 group compared to those in the G-H5N1 and control groups. These results provide a basis for the screening of avian influenza-resistance genes or methylation markers, analyzing the epigenetic regulation mechanisms of avian influenza, and performing selective breeding for disease resistance.

Our objective is to evaluate the accuracy of three algorithms in differentiating the origins of outflow tract ventricular arrhythmias (OTVAs). This study involved 110 consecutive patients with OTVAs for whom a standard 12-lead surface electrocardiogram (ECG) showed typical left bundle branch block morphology with an inferior axis. All the ECG tracings were retrospectively analyzed using the following three recently published ECG algorithms: 1) the transitional zone (TZ) index, 2) the V2 transition ratio, and 3) V2 R wave duration and R/S wave amplitude indices. Considering all patients, the V2 transition ratio had the highest sensitivity (92.3%), while the R wave duration and R/S wave amplitude indices in V2 had the highest specificity (93.9%). The latter finding had a maximal area under the ROC curve of 0.925. In patients with left ventricular (LV) rotation, the V2 transition ratio had the highest sensitivity (94.1%), while the R wave duration and R/S wave amplitude indices in V2 had the highest specificity (87.5%). The former finding had a maximal area under the ROC curve of 0.892. All three published ECG algorithms are effective in differentiating the origin of OTVAs, while the V2 transition ratio, and the V2 R wave duration and R/S wave amplitude indices are the most sensitive and specific algorithms, respectively. Amongst all of the patients, the V2 R wave duration and R/S wave amplitude algorithm had the maximal area under the ROC curve, but in patients with LV rotation the V2 transition ratio algorithm had the maximum area under the ROC curve.

In this case-control study, we attempted to investigate the role of three common single nucleotide polymorphisms (SNPs; -1082G/A rs1800896, -819T/C rs1800871, and -592A/C rs1800872) in the IL-10 gene in the development of abdominal aortic aneurysm in a Chinese population. Three hundred and eighty-one patients with abdominal aortic aneurysm and age- and gender-matched healthy controls (N = 381) were collected between March 2012 and March 2014. The IL-10 -1082G/A, -819T/C, and -592A/C polymorphisms were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Logistic regression analyses revealed that the AA genotype of IL-10 -1082G/A was associated with an increased risk of abdominal aortic aneurysm compared to the GG genotype in a codominant model [odds ratio (OR) = 1.64, 95% confidence interval (CI) = 1.04-2.60]. Moreover, the GA+AA genotype was correlated with an elevated risk of abdominal aortic aneurysm compared to the GG genotype in a dominant model (OR = 1.34, 95%CI = 1.01-1.79). In conclusion, the results of our study suggested that the A allele of IL-10 -1082G/A is significantly associated with the development of abdominal aortic aneurysm compared to the wide-type genotype.

We investigated the relationships between single nucleotide polymorphisms (SNPs) of the interleukin (IL)-6 gene 174 G>C (rs1800795), 572 G>C (rs1800796), and 597 G/A (rs1800797) and coronary artery disease (CAD) risk in a Chinese population. This case-control study recruited 296 CAD patients and 327 controls between January 2009 and May 2012. Genotyping of IL-6 174 G>C (rs1800795), 572 G>C (rs1800796), and 597 G/A (rs1800797) was performed on a 384-well plate format using the Sequenom MassARRAY platform. CAD patients were more likely to be older and male, with a higher body mass index, diabetes, and hypertension, and presented higher triglycerides, and lower total cholesterol, low-density lipoprotein-cholesterol, and high-density lipoprotein-cholesterol. We found that the IL-6 174CC genotype was associated with a significantly increased risk of CAD compared to the wild-type GG genotype in a codominant model [odds ratio (95% confidence interval) = 1.94 (1.13-3.37)], whereas IL-6 174 G>C polymorphisms presented an increased risk of CAD in dominant and recessive models. However, we did not find that the IL-6 572 CC and 597 AA genotypes were correlated with an increased risk of CAD. IL-6 174 G>C rs1800795 was associated with CAD risk in a Chinese population. Further large-scale studies are required to determine whether IL-6 SNPs interact with environmental factors in the development of CAD.
