RESUMO

The objective of this study was to evaluate the effect of exogenous insulin administration on follicular growth, estrus presentation and conception rate during a protocol of ovulation synchronization. Dairy cows were subjected to the Heatsynch protocol, with the insertion of an intravaginal device containing 1.9 g of progesterone (CIDR) and an intramuscular injection containing 2.5 mg of GnRH on day 0. On day 7, the CIDR was removed and subjects were given 12.5 mg of dinoprost. Also on day 7, Insulin Group (IG, n = 21) animals received a subcutaneous injection containing 0.25 IU / kg of recombinant human insulin and the control group (CG, n = 25) received a 0.9% NaCl injection. On day 8, an injection of 0.5 mg of estradiol cypionate was administered to all cows. Animals were inseminated 12 hours after estrus presentation or at day 10 at fixed time. Follicular development was evaluated on days 7, 9, and 10 using transrectal ultrasonography, estrus presentation was observed between days 8 and 10, and conception rate was evaluated 30 days after AI. There were no differences in growth rate, follicular diameter, estrus presentation, and conception rate. Therefore, application of a single dose of insulin does not promoted an increase in follicular size, estrus presentation and conception rate in dairy cows.(AU)


O objetivo deste estudo foi avaliar o efeito da administração exógena de insulina sobre crescimento folicular, apresentação de cio e taxa de concepção durante um protocolo de sincronização da ovulação. As vacas holandês lactantes foram submetidas ao protocolo Heatsynch, com inserção do dispositivo intravaginal contendo 1,9 g de progesterona (CIDR) no dia 0 e uma injeção intramuscular de 2,5 mg de GnRH. No dia 7, o CIDR foi removido e foi aplicado 12,5 mg de dinoprost. Ainda no dia 7, os animais do Grupo Insulina (IG, n = 21) receberam uma injeção subcutânea de 0,25 UI / kg de insulina humana recombinante e o grupo controle (CG, n = 25) recebeu uma injeção de NaCl 0,9%. Background: Reproductive efficiency is one of the most important factors affecting the performance dairy cattle, and fixedtime artificial insemination (TAI) is an important biotechnology factor employed in bovine livestock. The achievement of satisfactory results in TAI protocols depends of several factors, such as health and body condition, milk yield, environmental temperatures, ovulation time, semen manipulation, as well as semen quality. The goal of this study was to evaluate the conception rate of heifers and multiparous cows based on different Holstein bulls used for artificial insemination (AI). The effects of the type of management AI, animal category, and season of the year were also examined.Materials, Methods & Results: A three-year database on the reproductive management of a dairy farm was formed. All the cows and heifers were of the Holstein breed, in semi-intensive nutritional management, milked twice a day, and received a total mix ration. After 45 days in milk, when cows were observed twice daily with estrus signs, posterior artificial insemination occurred 12 h after estrus visualization was made. However, cows that were not observed in estrus were submitted to a timed artificial insemination (TAI). In this study, these fixed factors were considered: bulls utilized for the AI (totalizing 10 animals with at least 100 inseminations in each season, denoted by the letter B and a number - B1, B2, B3, B4, B5, B6, B7, B8, B9, B10); the season of the insemination (spring/summer being from November to April and autumn/winter from May to October), category animal (heifers, animals that never have been calved, or multiparous, animals that have already given birth more than once), and the type of AI (which included TAI using a protocol for synchronization, but cows having not necessarily shown estrus signs at the AI; ETAI: using protocol of synchronization, cows showing estrus signs at the AI;[...](AU)

The objective, with this study, was to investigate the consequences of subclinical hypomagnesemia on the metabolic parameters of dairy cows. Blood samples were collected from 12 animals every two days, between -22 pre and 22 days postpartum, for blood analysis and determining the metabolic profile. The animals were grouped according to magnesium blood concentrations: Hypomagnesemia group (n=5), with blood levels below 1.8 mg/dL in at least two consecutive days, and Control group (n=7), with blood levels above 1.8 mg/dL during the period. The hypomagnesemia group had higher levels of glucagon on days 10, 18 and 20 as well as glucagon/insulin ratio was higher on 6, 8, 10, 12 and 16 days after calving. The blood concentrations of aspartate aminotransferase in the hypomagnesemia group were higher during days 0, 6, 8, 10, 12 and 14 after calving. The results indicate that the low blood levels of magnesium during peripartum may be associated with elevated levels of aspartate amino transferase and glucagon in the blood. In general, subclinical hypomagnesemia does not alter the levels of indicators of energy metabolism, but the results show that cows with hypomagnesemia have a higher rate of glucagon/insulin, demonstrating a greater challenge to maintain glucose blood concentration.(AU)

Several programs are available to inseminate dairy cows, with variation in the use of different types of hormones, time of use and range of duration period, as well as the time of AI. The objective of this study was to evaluate the efficiency of follicular wave emergence induction of estradiol benzoate in comparison to GnRH in a Heatsynch protocol and its influence on reproductive parameters. Materials, Methods & Results: In this study were used primiparous and multiparous Holstein cows (n = 325). Animals were divided randomly into two different treatments according with the hormone used to induce follicular wave emergence (D0). The GnRH group (n = 167), was given 25 mg, while in the EB group (n = 158) was used 2 mg. At same day, were inserted, a controlled internal drug-releasing containing 1.9 g of progesterone CIDR®. On day 7, CIDR was removed and cows were given 25 mg luteinizing hormone followed by an injection of 1 mg Inducer Ovulation at day 8. The benefits associated to Heatsynch are the lower cost of the protocol and the increase in the number of cows detected in estrus. Furthermore, it has been suggested that protocols combining AI after estrus detection followed by TAI could increase overall conception rates, as well as increased AI submission rates. The aim of this study was evaluate reproductive responses of dairy cows submitted to a Heatsynch protocol according the estrus behavior and controlled internal device release (CIDR) uses.Material, Methods & Results: Holstein lactating cows (n = 562) were enrolled in protocol using (CIDR) (1.9 g of progesterone) insertion at (day 0) D0 either a new one, a previous used once (7 days - second use) or twice used (14 days - third use). Based on the detection of estrus and the moment of AI after the CIDR removal, cows were divided into 3 groups: cows showing signs of estrus and therefore AI 36 to 60 h after CIDR removal (EPTAI group; n = 241); cows showing estrus and AI 72 h after CIDR removal (ETAI group; n = 207); and cows that did not show any sign of estrus after CIDR removal and were TAI 72 h later (NoETAI group; n = 335).
