RESUMO

One lung ventilation (OLV) often results in trauma to the unventilated contralateral lung. This study aims to evaluate the effects of different OLV regimens on the injury of the unventilated contralateral lung to identify the best conditions for OLV. Forty rabbits were divided into five groups: a sham group, OLV group I (fraction of inspired oxygen (FIO2) 1.0, tidal volume (VT) 8mL/kg, respiratory rate (R) 40 breaths/min and inspiratory/expiratory ratio (I:E) 1:2), OLV group II (FIO2=1.0, VT 8mL/kg, R 40 breaths/min, I:E 1:2, and positive end-expiratory pressure (PEEP) 5 cm H2O), OLV group III (FIO2 1.0, VT 6mL/kg, R 40 breaths/min, I:E 1:2 and PEEP 5 cm H2O) and OLV group IV (FIO2 0.8, VT 6mL/kg, R 40 breaths/min, I:E 1:2 and PEEP 5 cm H2O). Animals from all OLV groups received two-lung ventilation (TLV) to establish a baseline, followed by one of the indicated OLV regimens. The rabbits in the sham group were intubated through trachea and ventilated with fresh air. Arterial blood gas samples were collected, lung injury parameters were evaluated, and the concentrations of TNF-α and IL-8 in bronchoalveolar lavage fluid (BALF) and pulmonary surfactant protein A (SPA) in the unventilated lung were also measured. In OLV group I, the unventilated left lung had higher TNF-α, IL-8 and lung injury score but lower SPA than the ventilated right lung. In OLV groups I to III, the concentrations of TNF-α, IL-8 and lung injury score in the left lung decreased but SPA increased. No differences in these parameters between OLV groups III and IV were observed. Strategic ventilation designed for OLV groups III and IV reduced OLV-induced injury of the non-ventilated contralateral lung in rabbits.(AU)


Ventilação pulmonar unilateral (OLV) frequentemente resulta em trauma no pulmão contralateral não ventilado. Este estudo visa avaliar os efeitos de diferentes regimes de OLV sobre a lesão do pulmão contralateral não ventilado para identificar as melhores condições para OLV. Quarenta coelhos foram divididos em cinco grupos: um grupo falso, OLV grupo I (fração de oxigênio inspirado (FIO2) 1.0, volume corrente (VT) 8mL/kg, frequência respiratória (R) 40 respirações/min e relação inspiração/expiração (I:E) 1:2), OLV grupo II (FIO2=1.0, VT 8mL/kg, R 40 respirações/min, I:E 1:2, e pressão positiva expiratória final (PEEP) 5 cm H2O), OLV grupo III (FIO2 1.0, VT 6mL/kg, R 40 respirações/min, I:E 1:2 e PEEP 5 cm H2O) e OLV grupo IV (FIO2 0.8, VT 6mL/kg, R 40 respirações/min, I:E 1:2 e PEEP 5 cm H2O). Os animais de todos os grupos OLV receberam ventilação nos dois pulmões (TLV) para estabelecer uma linha de base, seguida por um dos regimes OLV indicados. Os coelhos do grupo falso foram intubados através da traqueia e ventilados com ar fresco. Amostras de gases no sangue arterial foram coletadas, parâmetros de lesão pulmonar foram avaliados e as concentrações de TNF-α e IL-8 no fluido de lavagem bronco alveolar (BALF) e proteína A do surfactante pulmonar (SPA) no pulmão não ventilado também foram medidas. No grupo OLV I, o pulmão esquerdo não ventilado tinha maior índice de TNF-α, IL-8 e lesão pulmonar, mas menor SPA do que o pulmão direito ventilado. Nos grupos OLV I a III, as concentrações de TNF-α, IL-8 e a pontuação de lesão pulmonar no pulmão esquerdo diminuíram, mas o SPA aumentou. Não foram observadas diferenças nestes parâmetros entre os grupos OLV III e IV. A ventilação estratégica projetada para os grupos OLV III e IV reduziu a lesão induzida por OLV do pulmão contralateral não ventilado em coelhos.(AU)

Here, we conducted a case-control study to investigate the association between IL-10 gene polymorphisms and development of CAD in a Chinese population. A total of 220 patients with CAD and 236 control subjects who visited the Zhengzhou People's Hospital between May 2012 and June 2014 were selected for this study. The IL-10-1082A/G and -592A/C polymorphisms were genotyped by polymerase chain reaction coupled with restriction fragment length polymorphism. The chi-squared test revealed a significant difference in the distributions of the IL-10-1082A/G genotypes (χ2 = 6.32, P = 0.04). This data was then statistically analyzed by logistic regression analysis; we observed revealed that the CC genotype of IL-10-1082A/G had a higher risk of CAD in comparison to the AA genotype (OR = 2.09, 95%CI = 1.11-3.97). Moreover, the C allele of IL-10-1082A/G had a 1.39 fold risk of CAD when compared with (OR = 1.39, 95%CI = 1.06-1.82). We did not observe any significant correlations between the IL-10-592A/C genetic variation and susceptibility to CAD. In conclusion, our study suggests that the IL-10-1082A/G genetic variation could influence the development of CAD in a Chinese population.

The aim of this study was to investigate the role played by the IL-6 rs1800795 (-174G/C) and rs1800796 (-572G>C) polymorphisms in the susceptibility to allergic rhinitis in a Chinese population. A total of 265 patients with allergic rhinitis and 265 controls from our hospital were enrolled in this study. The IL-6 rs1800795 and rs1800796 polymorphisms were genotyped by polymerase chain reaction coupled with restriction fragment length polymorphism. The results of the χ(2) statistical analysis revealed significant differences in the allele frequencies of IL-6 rs1800795 between patients with allergic rhinitis and controls (χ(2) = 4.52, P = 0.03). Multivariate logistic regression analyses revealed that individuals with the C allele of IL-6 rs1800795 were susceptible to increased risk of allergic rhinitis, compared to those expressing the G allele (adjusted OR = 1.31; 95%CI = 1.01-1.68). In conclusion, the results of our study indicated that the IL-6 rs1800795 polymorphism was associated with an increased risk of allergic rhinitis.

Our study aimed to investigate the association between multidrug resistance (MDR1) gene polymorphisms and the response to imatinib (IM) in chronic myeloid leukemia (CML). An electronic databases in PubMed, Cochrane Library, Wanfang, China National Knowledge Infrastructure, and VIP were searched using combinations of keywords relating to MDR1 polymorphisms and the response to IM in CML. Studies retrieved from database searches were screened using stringent inclusion and exclusion criteria. The Comprehensive Meta-analysis 2.0 software was utilized for all statistical analyses. In total, 186 studies were initially retrieved, and 10 studies, involving 987 CML patients, were eventually included in this meta-analysis. Results of our study revealed no significant associations between MDR1 rs1045642, rs1128503, and rs2032582 polymorphisms and major molecular response and complete molecular response in CML patients. Significant differences were observed in the genotype frequencies of MDR1 rs1128503 under homozygous, heterozygous, and recessive models, between CML patients sensitive and resistant to IM. A significant difference in genotype frequencies of MDR1 rs2032582 was also observed under allele, homozygous, heterozygous, and recessive models between CML patients sensitive and resistant to IM. In conclusion, based on our meta-analysis, the MDR1 polymorphisms, rs1045642, rs1128503, and rs2032582, are not directly correlated with the curative effect of IM treatment of CML patients.

RESUMO

Pteroceltis tatarinowii (Ulmaceae) is a scientifically and economically important temperate deciduous tree that is endemic to China. In the present study, 12 P. tatarinowii polymorphic microsatellite loci were developed using the tailed primer-M13-simple sequence repeats (TP-M13-SSR) biotin-capture method. The number of alleles per locus ranged from 2 to 10, with an average of 6.58. The observed and expected heterozygosity ranged from 0.208 to 0.958 and from 0.198 to 0.858, with average values of 0.703 and 0.710, respectively. The markers isolated in this study represent a favorable tool for further analyses of the population genetic structure and evolutionary history of this relic tree.

We investigated local changes in BMP-2/4 expression in rat spinal cords 1 week following injury to study the damage effects of BMP-2/4 in spinal cord injury (SCI). Sprague Dawley rats (45, 4 months old) were randomized into three groups comprising 15 rats each: a SHAM group, an SCI without noggin group (SCIO), and an SCI with noggin group (SCID). The SCIO and SCID groups were subjected to spinal cord hemisection, and motor activity was assessed using the BBB score. Expression of BMP-2/4 in each injured spinal cord section was examined by hematoxylin and eosin staining, immunohistochemistry, and western blot. There were no significant differences in BBB scores among the three groups (P > 0.05). Following hemisection, the BBB score in the SHAM group was significantly higher than in the other two groups on the 1st day after modeling (P < 0.05), and the BBB scores in the SCIO and SCID groups were not significantly different (P > 0.05). Seven days after modeling, the BBB score in the SHAM group was significantly higher than in the other two groups (P < 0.05), and the BBB score in the SCID group was obviously higher than in the SCIO group (P < 0.05). The expression of BMP-2/4 was highest in the SCIO group and lowest in the SHAM group (P < 0.05). SCI can cause severe impairment of motor activity in rats. Seven days after SCI, the local expression of BMP-2/4 had obviously increased; noggin can effectively inhibit the expression of BMP-2/4 and reduce impairment.

We collected data regarding 340 disease resistance quantitative trait loci (QTLs) from the maize genomic database (MaizeGDB). We constructed an integrated linkage map and analyzed this map by using the BioMercator 2.1 software with IBM2 2008 Neighbors genetic linkage map as a reference. We used a meta-analysis method to identify five "consensus" synthetic resistance QTLs located on maize chromosomes 1, 3, 6, and 10, with map intervals of 5.14, 9.00, 28.50, 1.73, and 33.34 cM, respectively. The gene and marker sequences within the five "consensus" QTLs were downloaded from the MaizeGDB website. We identified eight resistance gene analogs (RGAs), through comparison of these sequences with the resistance genes of other members of Poaceae by using the online BLASTx software. On the basis of comparative mapping between the maize genetic map and the rice physical map, 54 rice and 44 maize resistance genes were projected onto the maize IBM2 2008 Neighbors genetic linkage map by using a synteny conservation approach. Additionally, 15 resistance genes in the "consensus" QTL regions were found in two "consensus" QTLs on chromosomes 3 and 6; these resistance genes included rp3, scmv2, wsm2, RG3, RG16, RG36, RG51, RG53, scmv1, mdm1, RG5, RG8, RG10, RG14, and RG29. Our results provide valuable information for fine-mapping QTL, gene cloning, and molecular breeding for resistance in maize.

The ether extract of the Chilean liverwort Lepicolea ochroleuca yielded three sesquiterpenoids, ent-4 beta-Hydroxy-10 alpha-methoxyaromadendrane, ent-3 beta-Hydroxyspathulenol, and 1,10-Dioxotayloriane, as minor components. The major components were ledol and 13-epi-neoverrucosan-5 beta-ol, four other minor fusicoccanoids were identified.

We have validated a method to measure bile salt binding by Maalox (aluminum hydroxide and magnesium hydroxide), Carafate (sucralfate), and Questran (cholestyramine) in vitro. The method used in this study involves a correction for adherent water volume and thus provides a correct measure of bile salt binding. With this approach, we described the binding properties of Maalox, Carafate, and Questran. The bile salt binding capacities of Carafate and Maalox are limited and do not have physiological or pharmacological significance. On the other hand, we found that Questran has substantial bile salt binding capacity. At the recommended dosage, Questran could deplete the total bile salt pool. We also found that Carafate, although not used as an antacid, has buffering capacity (maintaining a pH of solution in the range 4.2-4.8) which might contribute to its effectiveness as an ulcer treatment drug.
