RESUMO

The mussel-inspired method has been investigated to modify commercial ultrafiltration membranes to induce antifouling characteristics. Such features are essential to improve the feasibility of using membrane processes in protein recovery from waste streams, wastewater treatment, and reuse. However, some issues still need to be clarified, such as the influence of membrane pore size and the polymer concentration used in modifying the solution. The aim of the present work is to study a one-step deposition of dopamine (DA) and polyethyleneimine (PEI) on ultrafiltration membrane surfaces. The effects of different membrane molecular weight cut-offs (MWCO, 20, 30, and 50 kDa) and DA/PEI concentrations on membrane performance were assessed by surface characterization (FTIR, AFM, zeta potential, contact angle, protein adsorption) and permeation of protein solution. Results indicate that larger MWCO membranes (50 kDa) are most benefited by modification using DA and PEI. Moreover, PEI is primarily responsible for improving membrane performance in protein solution filtration. The membrane modified with 0.5:4.0 mg mL-1 (DA: PEI) presented a better performance in protein solution filtration, with only 15% of permeate flux drop after 2 h of filtration. The modified membrane can thus be potentially applied to the recovery of proteins from waste streams.

We studied different pre-treatments of poultry litter aiming to add economic value to this residue. Strategies were applied to extract ammonium nitrogen with the aim of allowing its further use as fertilizer, and to promote the hydrolysis and solubilization of lignocellulosic components with the aim of facilitating its subsequent conversion to biogas. Ammonia extraction was performed by solubilization in water in a one-step process and by successive extraction steps (3 times 60 min). Successive extractions presented greater removal of total ammonia nitrogen than did one-step extraction, solubilizing about 36% of the ammonia in water. In parallel pre-treatment using ultrasound was performed to increase carbon bioavailability for anaerobic digestion. Using this tool, 24.7 g kg-1 of total organic carbon and 13.0 g kg-1 of total reducing sugars were solubilized, employing 10% dry mass sample amount, 100% amplitude ultrasound at frequency of 20 kHz amplitude and 2.5 min of treatment (energy input of 299 ± 7 kJ L-1; 3,822 ± 95 kJ kg-1). Anaerobic digestion of ultrassound pre-treated biomass was evaluated using a biological biogas production assay, and an increase of 10% of biogas production was obtained compared to untreated samples (147.9 and 163.0 mL g-1 for crude and pre-treated PL, respectively). The findings suggest that these are environmentally friendly and sustainable strategies to add economic value to poultry litter, reducing the environmental impacts of improper disposal.

The objective of this study is to evaluate the batch enzymatic production of biodiesel in solvent-free system under ultrasound using as substrates ethanol, soybean oil and macauba fruit oil. For this purpose, a Plackett & Burman experimental design was carried out for soybean oil while a 24-1 design was conducted for macauba oil in order to maximize the biodiesel conversion for each system. Good conversions to fatty acid ethyl esters (FAEE), 88% for soybean oil and 75.2% for macauba oil, was obtained thus demonstrating the potential use of ultrasound for this reaction system.

Membrane performance is decreased by fouling, reducing permeate flux and membrane lifespan. This paper assesses ultrafiltration of a model textile effluent assisted by permanent magnetic field as an alternative to improve the water permeability recovery. Ultrafiltration was performed in a tangential module and model solutions, composed of carboxymethylcellulose (CMC) and sodium sulphate (Na2SO4). The feed was permeated through 30 kDa polysulphone membrane with and without the presence of a permanent magnetic field of 0.41 T, perpendicular to the membrane surface. Magnetic induction (MI) of feed solution was also investigated by recirculation of the feed stream through the magnetic field for 3 h. The increase in feed concentration did not affect permeate flux, while the presence of salt resulted in a severe flux decline. Effective water permeability recovery was obtained when the magnetic field was applied in the ultrafiltration process, although the MI of the CMC and Na2SO4 solutions also caused some enhancement in permeability recovery. Scanning electron microscopy images showed differences between the assays done with and without the presence of magnetic field. The magnetic field application in ultrafiltration of CMC and Na2SO4 solutions has proved to be an attractive alternative for improving the permeability recovery.

Lipases produced by a newly isolated Sporidiobolus pararoseus strain have potential catalytic ability for esterification reactions. After production, the enzymatic extracts (conventional crude and precipitated, 'CC' and 'CP', and industrial crude and precipitated, 'IC' e 'IP') were partially characterized. The enzymes presented, in general, higher specificity for short chain alcohols and fatty acids. The precipitated extract showed a good thermal stability, higher than that for crude enzymatic extracts. The 'CC' and 'CP' enzymes presented high activities after exposure to pH 6.5 and 40 ºC. On the other hand, the 'IC' and 'IP' extracts kept their activities in a wide range of pH memory but presented preference for higher reaction temperatures. Preliminary studies of application of the crude lipase extract in the enzymatic production of geranyl propionate using geraniol and propionic acid as substrates in solvent-free system led to a reaction conversion of 42 ± 1.5%.

An experimental design was carried out to evaluate the effect of the concentrations of sodium alginate, glutaraldehyde and activated coal on the immobilization of inulinase from Kluyveromyces marxianus NRRL Y-7571. The experimental condition of 20 g/L of sodium alginate, 50 mL/L of glutaraldehyde and 30 g/L of activated coal led to the highest specific activity (2,063.5 U/mg of protein), corresponding to an enhancement of about 26 times compared to the activity of the free enzyme (79.1 U/mg of protein). The effect of pH and temperature on the immobilized enzyme activity was also evaluated, showing optimal activities at pH of 5.5 and 55 °C. The study of storage of immobilized inulinase in different temperatures showed that the extract kept its initial activity after 43 days of storage at 40 and 50 °C and after 138 days of storage either at 4 or 25 °C.

The lipase produced by a newly isolate Sporidiobolus pararoseus strain has potential catalysis ability for esterification reactions. In order to improve its synthetic activity, this work aimed at optimizing 'synthetic lipase' production by submerged fermentation of a conventional media based on peptone, yeast extract, NaCl and olive oil using experimental design technique. According to the results obtained in the first experimental design (2(4-1)), yeast extract and NaCl concentrations were tested to further optimization by response surface methodology. The maximum 'synthetic lipase' activity obtained was 26.9 U/mL in the optimized media (5.0, 6.8, 7.0 and 1.0% (wt/v) of peptone, yeast extract, NaCl and olive oil, respectively), representing a 6.36-fold increase compared to the initial medium. The time course of 'synthetic lipase' production in the optimized condition was evaluated in terms of synthetic activity, protease activity, biomass and total carbon and the maximum synthetic activity was observed during the stationary phase of growth.

The adulteration of the product Ilex paraguariensis with other Ilex species is a mAjor problem for maté tea producers. In this work, three species of Ilex were evaluated for their volatile composition by headspace solid phase microextraction coupled to gas chromatography and mass spectrum detector (HS-SPME/GC-MS). The adulterating species I. dumnosa and I. theizans Mart. ex Reissek presented a different profile of volatile organic compounds when compared to I. paraguariensis. Aldehydes methyl-butanal, pentanal, hexanal, heptanal and nonanal were detected only in the adulterating species. This result suggests that such compounds are potential chemical markers for identification of adulteration and quality analysis of products based on Ilex paraguariensis.


A adulteração do produto Ilex paraguariensis com outras espécies de Ilex é um dos principais problemas dos produtores de erva-mate. Neste trabalho, três espécies de Ilex foram avaliadas quanto à sua composição volátil por microextração em fase sólida acoplada à cromatografia gasosa e detector de espectro de massas (HS-SPME/GC-MS). As espécies adulterantes I. dumnosa e I. theizans Mart. ex Reissek apresentaram um perfil diferente de compostos orgânicos voláteis, quando comparadas com a I. paraguariensis. Os aldeídos metil-butanal, pentanal, hexanal, heptanal e nonanal foram detectados apenas nas espécies adulterantes. Esse resultado sugere que esses compostos químicos são marcadores potenciais para a identificação de adulteração e análise da qualidade dos produtos à base de Ilex paraguariensis.
