Emulsion systems have been a breakthrough in cosmetic products, providing performance and effectiveness of products that use this technological strategy for drug delivery systems. In this sense, the microemulsion of the multiple emulsion W/O/W type containing a standardized extract of Trichilia catigua with high levels of polyphenols and antioxidants has great potential for cosmetic use. The aim of this study was to evaluate the formulations safety through the analysis of toxicity, comedogenicity, and histopathology in rabbits and apply the Phase-Resolved Photoacoustic Spectroscopy method to determine the formulation percutaneous penetration through the skin. The ex vivo experiments were performed in the ears of albino New Zealand rabbits treated twice a day for 14 days. The results of histological, hematological, and blood chemistry showed that the formulations are safe. Histopathological analysis showed no tissue reaction in any of the analyzed organs (liver and kidneys), confirming the absence of toxicity. Histological analysis showed that the formulations with extract of T. catigua demonstrated mild-moderately comedogenic and acanthosis compared to the control group. Inflammatory reactions, erythema, and desquamation were not observed in treated and controls animals. The phase-resolved photoacoustic spectroscopy method showed the penetration of the developed formulations throughout the rabbit's skin, identifying their absorption bands at the dermal side of the skin. In conclusion, the results of this study provide a step towards the application of the developed natural antioxidant encapsulated in a multiple microemulsion for skincare, concerned with the physical, chemical, and biological properties of the formulation.

Fish oil (FO) is a natural source of omega-3 fatty acids, with well-established beneficial effects in inflammatory diseases when FO is orally administered. This study investigated the effects of a topically applied FO preparation (FOP) on phenol-induced ear edema and evaluated the percutaneous penetration of FOP in ear tissue. After applying phenol, groups of mice received FOP on the ear. After 1 h, ear tissue was collected to determine the percent inhibition of edema, myeloperoxidase activity, and to perform photoacoustic spectroscopy (PAS). Treatment with FOP did not reduce edema, but reduced myeloperoxidase activity. The FOP decreased the area of bands that characterize inflamed tissue and penetrated into the tissue. These results indicated an inhibitory effect of FOP on leukocyte recruitment in phenol-induced ear edema. These data support the applicability of PAS as a non-destructive method for evaluating the inflammatory response, percutaneous penetration and antiinflammatory activity of compounds.

BACKGROUND: Agrobacterium sp. IFO 13140 cells were immobilized on a loofa sponge and used to produce curdlan over five successive cycles. The interaction between microbial cells and the loofa sponge as well as the produced curdlan were characterized by Fourier transform infrared-attenuated total reflectance (FTIR-ATR) spectrometry. The purity of the curdlan was also evaluated. The storage stability of the immobilized cells was assessed and the produced curdlan was used in a functional yogurt formulation. RESULTS: The average curdlan production by immobilized cells was 17.84 g L(-1) . The presence of the microorganism in the sponge was confirmed and did not cause alterations in the matrix, and the chemical structure of the curdlan was the same as that of commercial curdlan. The purity of both was similar. The immobilized cells remained active after 300 days of storage at -18 °C. The use of the produced curdlan in a functional yogurt resulted in a product with lower syneresis. CONCLUSION: A large number of cells physically adhered to the surface of loofa sponge fibers, and its use as an immobilization matrix to produce curdlan was effective. The use of the produced curdlan in yogurt allowed the development of a more stable product. © 2015 Society of Chemical Industry.
