RESUMO

This study examined the effects of exogenous hCG administration on ovarian function and pregnancy rates in estrous-induced dairy goats during the transition into the breeding season. Eighty-six Toggenburg does received 60 mg of medroxyprogesterone acetate intravaginal sponge for 6 d plus 200 IU of equine chorionic gonadotropin and 30 µg of d-cloprostenol i.m. 24 h before sponge removal, and were then bred for 96 h. Seven days (D7) after first mating the does received either 1 mL of saline (the control group, n = 43) or 300 IU of hCG (the hCG-treated group, n = 43) i.m. Transrectal ovarian ultrasonography (B-mode and color Doppler) was performed on D7, D13, D17, and D21 and ultrasonographic pregnancy detection on D30. Pregnancy rate was higher (P < 0.05) in hCG-treated goats (90.7%; 39/43) than that in control animals (74.4%; 32/43). Accessory luteal structures (ALSs) were detected in 46.5% (20/43) of hCG-treated does. All hCG-treated does that had ALSs and 82.6% of goats without ALS post-treatment remained pregnant. The total luteal area increased (P < 0.05) from D7 to D13 in pregnant animals of both groups, whereas mean vascular area declined (P < 0.05) by D21 in all nonpregnant does. Serum progesterone concentrations increased (P < 0.05) on D21 in pregnant goats of both groups, but they were related to changes in luteal tissue content only in control does throughout the present study. Mean daily numbers of small- and medium-sized antral follicles decreased (P < 0.05) only in pregnant animals of both groups with a decline in medium follicle numbers occurring earlier in hCG-treated (D13) compared with control does (D17). To summarize, a single dose of hCG given on D7 after estrus was followed by a decrease in the number of medium-sized antral follicles in gestating hCG-treated does, induced the formation of ALSs in ~47% of all hCG-treated does, and significantly increased the pregnancy rate in estrous-induced Toggenburg goats in the transition to the breeding season.

Pododermatitis, also known as "bumblefoot", is an inflammatory lesion of the footpad. The objective of this study was to evaluate the influence of genetic strain and sex on the incidence of footpad lesions in broilers. The experiment was carried out at São Paulo State University, using 480 broilers distributed in a completely randomized experimental design in a 2X2 factorial arrangement (2 strains X2 sexes), with four replicates of 30 broilers each, totaling 16 experimental units. Feed intake, weight gain, feed conversion ratio, and livability were evaluated at 21, 35 and 42 days of age; production efficiency factor was determined at the end of the rearing period. On day 42, the footpads of 100 broilers were grossly examined and assigned a lesion score in a 1-3 scale (Almeida Paz & Martins, 2014). Three samples per score within treatment were collected for microscopic evaluation. Analysis of variance was applied and performance parameter means were compared by Tukey's test. Footpad lesion incidence was analyzed by the c2 test using SAS (version8.2). Ross(r) broilers presented higher feed intake during all evaluated periods, and higher weight gain only in the period 1 to 21 days compared with Cobb(r) broilers. On the other hand, Cobb(r) presented better feed conversion ratio in the periods of 1 to 35 and 1 to 42 days. The incidence of foot pad lesions was statistically different (p < 0.05) between strains and sexes, but there were no interactions between these factors. The lesion scores assigned were compatible with the histopathological results, showing that the higher the score, the more severe were the dermal and epidermal lesions.(AU)

The objective of this study was evaluated the villus integrity of commercial turkeys submitted to coccidiosis prevention methods and challenged with Eimeria field oocysts, using scanning electron microscopy. Sixty BUT 9 female commercial turkeys were distributed in a completely randomized block design split with two treatments: T1- control diet without vaccinations against coccidiosis and anticoccidial drug, and T2- vaccinated against coccidiosis (commercial vaccine). On d 21 of life, all birds of all treatments were challenged with a mixed-species containing E. meleagrimitis and E. galopavonis, via crop intubation with 1 mL of ~20,000 sporulated oocysts/bird. The size of the inoculum was determined in previous experiments. At 27 and 70 days of age, five birds per treatment were randomly removed to evaluate the intestinal integrity. Duodenum, jejunum and ileum segments were collected and processed according to routine scanning electron microscope. The enteric mucosa integrity of the commercial turkeys subjected to coccidiosis vaccination was reduced when compared to the birds of the control treatment. There was no difference in the performance results, therefore these results cannot be attributed to the vaccination as well as to the ineffectiveness against the challenge that was administered.

This study aimed at evaluating the intestinal integrity, using light and scanning electron microscopy (SEM), and the performance of broiler chickens fed additives alternative to antimicrobials. A total of 1080 male chicks were distributed according to a completely randomized experimental design, with six treatments with six replicates of 30 birds each. The following treatments were evaluated: basal diet (control), basal diet supplemented with an antimicrobial, basal diet supplemented with a probiotic, basal diet supplemented with a prebiotic, basal diet with a symbiotic, and basal diet supplemented with organic acids. Weight gain, feed intake, feed conversion ratio and livability were recorded when broiler chickens were 10, 21, 35, and 42 days old. On day 42, 72 birds were individually weighed and sacrificed. In order to evaluate the morphometrics of the different intestinal wall layers, segments of the small intestine and the cecum were collected from two birds per replicate, and intestinal integrity (SEM) was evaluated in the same segments of two birds per treatment. During the starter period (1-21 days old), birds fed the alternative additives presented similar weight gain as those fed the antimicrobial product, but were not different from control birds. Feed conversion ratio of birds fed alternative additives was better than that of the control birds from one to 10 days of age, but not during the remaining rearing period, and was similar to the birds receiving the antimicrobial. The morphometric parameters of the different intestinal wall layers was not influenced by the treatments. During the total rearing period, the evaluated alternative additives did not improve intestinal integrity or broiler performance.

In order to study the morphological changes that occur in cells of the testes of isogenic black mouse C57BL/6/Uni into three periods during spermatogenetic used 15 mice divided into 3 groups of 5 animals with 40, 50 and 60 days of age. The mice were sacrificed and weighed. Testicles were weighed and measured, and histologically processed and stained with HE, PAS and Masson Massom-H and evaluated under light microscopy. It was observed that group I with 40 days of age in the seminiferous tubules had a lumen with sparse small amount of interstitial tubular cells. In the seminiferous epithelium type A spermatogonia, intermediate and B were identified, which occupied the compartment adbasal and intermingled with these cells in spermatocytes I in Pachytene and leptotene was observed, whereas in the adluminal compartment Golgi phase spermatids we observed the presence of acrosomal granule. In group II, the cells of the seminiferous epithelium were developed and it was observed in round spermatids cephalic hood phase plus many elongated spermatids in acrosome phase and Sertoli cells. In group III, 60 days old, it was found that seminiferous epithelium which was of the tubules had elongated spermatids in acrosome phase and maturation, with elongated nuclei and acrosomal system typical of spermiation in the presence of sperm and residual bodies near the tubular lumen. Therefore morphological evolution of germ cell testicular spermatids can be checked and recognized in its four phases: golgi, cap, acrosome and maturation over the age of the animal.


Con el fin de estudiar los cambios morfológicos que ocurren en las células de los testículos del ratón negro isogénico C57BL/6/Uni en tres períodos durante el proceso espermatogenético, se utilizaron 15 ratones divididos en 3 grupos de 5 animales con 40, 50 y 60 días de edad. Los ratones fueron sacrificados y pesados. Luego se pesaron y midieron los testículos, para ser finalmete procesados histológicamente y teñidos con HE, PAS y tricrómico Massom-H y evaluados bajo microscopía de luz. Se observó en el grupo I con 40 días de edad que los túbulos seminíferos tenían un lumen pequeño con escaza cantidad de células tubulares intersticiales. En el epitelio seminífero se identificaron espermatogonias tipo A, intermedio y B, que ocuparon el compartimiento adbasal y entremezcladas con estas células, se observó los espermatocitos I en paquiteno y leptoteno, mientras que en el compartimiento adluminal se observaron espermátidas de fase Golgi en presencia de gránulos acrosomales. En el grupo II, las células del epitelio seminífero se desarrollaron y fue posible observar espermátidas redondas en fase de capuz cefálico además de numerosas espermátidas elongadas en fase acrosómica y células sustentaculares. En el grupo III, de 60 días de edad, se encontró que el epitelio seminífero que revestía los túbulos presentaban espermátidas alargadas en fase acrosómica y de maduración, con sus núcleos alargados y el sistema acrosomal típico de la espermiación, con la presencia de espermatozoides y cuerpos residuales cerca del lumen tubular. Así, se puede comprobar con el paso de la edad del animal, la evolución morfológica de las células germinativas testiculares y reconocer las espermátidas en sus cuatro fases: golgi, capuchón, acrosomal y de maduración.

Con el fin de estudiar los cambios morfológicos que ocurren en las células del testículo del ratón negro isogénico C57BL/6/Uni en tres períodos diferentes del proceso espermatogenético; fueron utilizados 15 ratones divididos en 3 grupos (n=5) con 40, 50 y 60 días de edad respectivamente. Todos los animales fueron sacrificados y pesados. Posteriormente sus testículos se pesaron, midieron y procesaron histológicamente para HE, PAS y tricrómico Massom-H. Las muestras obtenidas fueron evaluados con microscopía de luz. En el grupo I con 40 días se observaron túbulos seminíferos con un lúmen pequeño y escaza cantidad de células intersticiales. En el epitelio seminífero se identificaron espermatogonias tipo A, intermedio y B, quienes ocuparon el compartimiento basal entremezclándose con espermatocitos I en paquiteno y leptoteno. En el compartimiento adluminal se observaron espermatidas de fase Golgi y presencia de gránulos acrosomales. En el grupo II de 50 días, se observaron células del epitelio seminífero desarrolladas, espermatidas en fase de capuz cefálico, muchas espermatidas elongadas en fase acrosomica y células sustentaculares. En el Grupo III de 60 días se observó el epitelio del túbulo seminífero con espermátidas alargadas en fase acrosómica y de maduración, con núcleos alargados y un sistema acrosomal típico de la espermiación, con presencia de espermatozoides y cuerpos residuales cerca del lúmen tubular. En conclusión se observa la evolución morfológica de las células germinativas testiculares y se reconocen las espermatides en sus cuatro fases: Golgi, capuchón, acrosomal y de maduración en las diferentes edades del animal.
