RESUMO

Mantle cell lymphoma (MCL) is a B-cell non-Hodgkin lymphoma; data indicate that blastoid and pleomorphic variants have a poor prognosis. We report characteristics and outcomes of patients with blastoid/pleomorphic variants of MCL. We retrospectively studied adults with newly diagnosed MCL treated from 2000 to 2015. Primary objectives were to describe progression-free survival (PFS) and overall survival (OS). Secondary objectives included characterization of patient characteristics and treatments. Of the 1029 patients with MCL studied, a total of 207 neoplasms were blastoid or pleomorphic variants. Median follow-up period was 82 months (range, 0.1-174 months); median PFS was 38 months (95% confidence interval [CI], 28-66) and OS was 68 months (95% CI, 45-96). Factors associated with PFS were receipt of consolidative autologous hematopoietic transplantation (auto-HCT; hazard ratio [HR], 0.52; 95% CI, 0.31-0.80; P < .05), MCL International Prognostic Index (MIPI) intermediate (HR, 2.3; 95% CI, 1.2-4.3; P < .02) and high (HR, 3.8; 95% CI, 2.0-7.4; P < .01) scores, and complete response to induction (HR, 0.29 (95% CI, 0.17-0.51). Receipt of auto-HCT was not associated with OS (HR, 0.69; 95% CI, 0.41-1.16; P = .16) but was associated with MIPI intermediate (HR, 5.7; 95% CI, 2.5-13.2; P < .01) and high (HR, 10.8; 95% CI, 4.7-24.9; P < .01) scores. We report outcomes in a large cohort of patients with blastoid/pleomorphic variant MCL. For eligible patients, receipt of auto-HCT after induction was associated with improved PFS but not OS. Higher MIPI score and auto-HCT ineligibility were associated with worse survival.

Bacteria may be the initial cause of certain pathologies as well as a secondary agent responsible for the development of complications such as pressure ulcer infections. Pressure ulcers are a persistent health problem, especially in immunocompromised patients, and associated with infection by opportunistic microorganisms with antimicrobial resistance, such as Klebsiella pneumoniae, highlight the need for the development of new antimicrobial approaches. Thus, the aim of this study was to evaluate the antibacterial and anti-adherent activity of Origanum vulgare L. (oregano) essential oil against Klebsiella pneumoniae strains, as well as the effect of its association with synthetic antimicrobials. To this end, the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) analyses were performed on microdilution plates. The assay of the Minimum Inhibitory Adherence Concentration (MIAC), with test tubes. As well as, the association study through the infusion disc method containing ampicillin (AMP), gentamicin (GEN), ciprofloxacin (CIP) and ceftriaxone (CEF). Therefore, it was possible to obtain that the essential oil of oregano presents antimicrobial and bactericidal activity, with MIC ranging between 128µg/mL and 256 µg/mL and MBC between 256 µg/mL and 512 µg/mL, on the tested K. pneumoniae strains. When used in association with ampicillin and gentamicin, oregano essential oil showed synergistic effect for some strains. Therefore, it is observed that the tested essential oil can act as a promising antibacterial in the treatment of diseases caused by K. pneumoniae.

As bactérias podem ser a causa inicial de determinadas patologias como também um agente secundário responsável pelo desenvolvimento de complicações como as infecções das lesões por pressão. As lesões por pressão configuram um problema de saúde persistente, principalmente em pacientes imunocomprometidos, e associadas a infecção por microrganismos oportunistas com resistência antimicrobiana, como por exemplo a Klebisiella pneumoniae, ressaltam a necessidade do desenvolvimento de novas abordagens antimicrobianas. Assim, o objetivo deste estudo foi avaliar a atividade antibacteriana e antiaderente do óleo essencial de Origanum vulgare L. (orégano) contra cepas da Klebisiella pneumoniae, bem como o efeito da sua associação com antimicrobianos sintéticos. Para isso foram realizadas as análises da Concentração Mínima Inibitória (CIM) e Concentração Bactericida Mínima (CBM) em placas de micro diluição. O ensaio da Concentração Mínima de Aderência (CIMA), com tubos de ensaio. Como também, o estudo de associação através do método de disco de infusão contendo ampicilina, gentamicina, ciprofloxacino e ceftriaxona. Assim sendo, foi possível obter que o óleo essencial de orégano apresenta atividade antimicrobiana e bactericida, com CIM variando entre 128µg/mL e 256 µg/mL e CBM entre 256 µg/mL e 512 µg/mL, sobre as cepas de K. pneumoniae testadas. Quando utilizado em associação com a ampicilina e gentamicina, o óleo essencial de orégano demonstrou efeito sinérgico para algumas cepas. Portanto, observa-se que o óleo essencial testado pode atuar como um promissor antibacteriano no tratamento de doenças causadas por K. pneumoniae.

This study aimed to identify the Staphylococcus species responsible for bovine mastitis in dairy herds in northern Brazil, to investigate the capacity of biofilm production, and to analyze the association of biofilm production with multiresistance and intensity of California Mastitis Tests (CMT) reactions that can make treatment more difficult and cause misdiagnoses, respectively. Milk samples were collected from 23 dairy farms located in five municipalities in the state of Acre. A total of 339 crossbred cows were tested by CMT, with 109 animals (229 udder ceilings) reacting to the test. After bacterial isolation in blood agar, the catalase-positive and gram-positive cocci were submitted for identification by MALDI-TOF MS. Of 103 strains identified as staphylococci, Staphylococcus chromogenes (58.3%) and Staphylococcus aureus (19.4%) were the most prevalent species. Biofilm production was quantitatively evaluated using a microplate adherence test. Among the Staphylococcus strains, 71.8% were biofilm producers. Most strains of S. chromogenes (68.3%) had the capacity to produce biofilms, ranging from weak (43.3%), moderate (13.3%), and strong (11.7%) producers. Among S. aureus strains, 50% were non-biofilm producers, and none were strong producers. Our data showed an association between biofilm production capacity and multidrug resistance. In addition, there was a reduction in the response to the CMT test, which can mask the diagnosis.


O presente estudo teve como objetivos identificar as espécies de estafilococos envolvidas com mastite em rebanhos leiteiros no norte do Brasil, investigar a capacidade de produção de biofilme e analisar a associação da produção de biofilme com multirresistência e intensidade de reações de CMT que podem dificultar o tratamento e causar erros de diagnósticos, respectivamente. Amostras de leite foram coletadas em 23 propriedades leiteiras, situadas em cinco municípios do estado do Acre. Um total de 339 vacas mestiças foram testadas pelo CMT, sendo detectados 109 animais (229 tetos) reagentes ao teste. Após isolamento em ágar sangue, os cocos Gram positivos produtores de catalase foram submetidos à identificação por MALDI TOF MS. Um total de 103 estirpes foram identificadas como estafilococos, sendo mais prevalentes as espécies Staphylococcus chromogenes (58,3%) e S. aureus (19,4%). A produção de biofilme foi avaliada quantitativamente pelo teste de aderência em microplaca. Dentre todas as amostras de estafilococos, 71,8% foram produtoras de biofilme. A maioria das amostras de S. chromogenes (68,3%) apresentou capacidade de produzir biofilme, variando de fraco (43,3%), moderado (13,3%) a forte (11,7%) produtor. Entre as amostras de S. aureus, 50% não foram produtoras e nenhuma foi produtora forte. Essa capacidade de produção de biofilme mostrou-se associada à multirresistência a antimicrobianos, além de diminuir a intensidade no teste CMT, podendo mascarar o diagnóstico.

Doxorubicin (DOX) is known to cause cognitive impairments in patients submitted to long-term chemotherapy (deficits also known as chemobrain). Therefore, there is an urgent need for therapeutic strategies capable of returning cancer survivors back to their previous quality of life. The present study investigated whether resveratrol (RSV) or curcumin (CUR) administration could affect mnemonic function and brain morphological changes following DOX administration in rats. Male Wistar rats were divided into 4 groups: DOX group (2.5 mg/kg/week for 4 weeks, i.p., plus distilled water for 28 days, oral gavage - OG), DOX + RSV group (DOX, 2.5 mg/kg/week for 4 weeks, i.p., plus RSV, 10 mg/kg/day for 28 days, OG), DOX + CUR group (DOX, 2.5 mg/kg/week for 4 weeks, i.p., plus CUR, 100 mg/kg/day for 28 days, OG) and control (CTR) group (0.9% saline solution weekly for 4 weeks, i.p., plus distilled water for 28 days, OG). Behavioral analyses (open field - OF - and the novel object recognition test - NORT) were performed. Brains were collected and analyzed by hematoxylin-eosin and luxol fast blue staining techniques and by immunohistochemistry for GFAP (glial fibrillary acidic protein) expression in astrocytes and Iba1 (ionized calcium-binding adaptor molecule 1) expression in microglia. DOX-injected rats presented short-term and long-term memory impairments as seen in the NORT at 3 and 24 h after habituation and increased GFAP and Iba1 expression, respectively, in astrocytes and microglia of the frontal cortex, hypothalamus and hippocampus. Such cognitive deficits were prevented by CUR at both periods and by RSV at 24 h. DOX-induced astrogliosis and microgliosis were avoided by RSV and CUR. No signs of demyelination or neuronal loss were found in any group. Thus, CUR and RSV prevented memory loss, astrogliosis and microgliosis induced by DOX monotherapy.

Chondroitin sulfate (CS) is a type of glycosaminoglycan described as an antioxidant molecule that has been found in animal species such as fish. Tilapia (Oreochromis niloticus) represents an eco-friendly source of this compound, since its economical processing generates usable waste, reducing the negative environmental impact. This waste was used for CS extraction, purification, characterization by enzymatic degradation, and evaluation of its antioxidant effect. CS obtained from tilapia presented sulfation mainly at carbon 4 of galactosamine, and it was not cytotoxic at concentrations up to 200 µg/mL. Furthermore, 100 µg/mL of CS from tilapia reduced the levels of reactive oxygen species to 47% of the total intracellular reactive oxygen species level. The ability of CS to chelate metal ions in vitro also suggested an ability to react with other pathways that generate oxidative radicals, such as the Haber-Weiss reaction, acting intracellularly in more than one way. Although the role of CS from tilapia remains unclear, the pharmacological effects described herein indicate that CS is a potential molecule for further study of the relationship between the structures and functions of chondroitin sulfates as antioxidants.

The aim of this work was to measure HMGB1, TNF-alpha, and IL-8 in bronchoalveolar lavage (BAL), serum and TLR2 and TLR4mRNA expression in lung tissue of rabbits with two grades of acute lung injury (ALI). The animals were randomly assigned to groups with severe (S) and mild/moderate (MM) ALI, induced with warm saline, and a control group. HMGB1, TNF-alpha, IL-8, TLR2mRNA and TLR4mRNA were measured after ALI induction. The results showed increased levels of IL-8, TNF-alpha, HMGB1 and TLR4mRNA in the ALI groups. HMGB1, IL-8 and TNF-alpha concentrations in BAL were higher in S compared MM. Increased TLR4mRNA was observed in S and MM versus control. The results suggest an early participation of HMGB1 in ALI together with IL-8 and TNF-alpha and association with severity. TLR4 has early expression and role in ALI pathophysiology but is not associated with severity.(AU)


O objetivo deste trabalho é determinar os níveis de HMGB1, TNF-alfa e IL-8 no lavado broncoalveolar (BAL), bem como quantificar a expressão sérica de TLR2 e TLR4 mRNA em tecido pulmonar de coelhos com dois graus de lesão pulmonar aguda (LPA). Os animais foram distribuídos aleatoriamente em grupos com LPA grave (S) e leve / moderada (MM), induzidas com solução salina morna, e um grupo controle. HMGB1, TNF-alfa, IL-8, TLR2mRNA e TLR4mRNA foram medidos após a indução de LPA e quatro horas de ventilação mecânica. Os resultados mostraram níveis aumentados de IL-8, TNF-alfa, HMGB1 e TLR4mRNA nos grupos com LPA. As concentrações de HMGB1, IL-8 e TNF-alfa no LBA foram maiores no S comparado ao MM. Aumento de TLR4mRNA foi observado em S e MM versus controle. Os resultados sugerem uma participação precoce da HMGB1 na LPA em conjunto com IL-8 e TNF-alfa e associação com a gravidade da LPA. O TLR4 foi expresso na ALI e possivelmente possui papel precoce na fisiopatologia da LPA, mas sem associação com a gravidade.(AU)

A total of 640 one-day-old Cobb broiler chicks, half male and half female, were reared up to 49 days of age. A completely randomized 2 × 4 factorial design was used. Experimental treatments consisted of four levels of organic zinc supplementation and one control (inorganic zinc supplementation) and two levels of sex (female and male), with four replicates of 16 birds per experimental plot. Animal performance, carcass and cut yields, and meat quality were evaluated. Feed intake increased with zinc supplementation. Broilers that received the lowest level of organic zinc showed better feed conversion than those fed an equivalent level of inorganic zinc. Increased levels of organic zinc reduced the abdominal fat content in females but impaired the oxidative stability of meat. The results showed that male broilers have better performance, carcass yield, and meat quality than females and that organic zinc improves broiler feed conversion and reduces abdominal fat but increases lipid oxidation in meat.(AU)


Foram alojados 640 pintainhos de corte, sendo metade de machos e metade de fêmeas, da linhagem Cobb, de um a 49 dias de idade. Foi adotado um delineamento inteiramente ao acaso, em esquema fatorial 2 × 4. Os tratamentos experimentais consistiram em quatro níveis de suplementação de zinco orgânico e um controle inorgânico, e diferentes sexos, com quatro repetições de 16 aves por parcela experimental. Foram avaliados o desempenho, a característica da carcaça e dos cortes, bem como a qualidade de carne dos frangos. O aumento dos níveis de zinco elevou o consumo de ração dos frangos. Os frangos que receberam zinco orgânico na exigência mínima apresentaram melhor conversão alimentar comparados àqueles que receberam ração de fonte inorgânica. O aumento nos níveis de zinco orgânico proporcionou redução no teor de gordura abdominal para fêmeas, mas níveis de zinco orgânico prejudicaram a estabilidade oxidativa da carne. Conclui-se que frangos machos apresentam melhor desempenho, carcaça e qualidade de carne em relação às fêmeas, e que o zinco orgânico melhora a conversão alimentar dos frangos, reduz a gordura abdominal, porém eleva a oxidação lipídica da carne.(AU)
