RESUMO

This study aims to evaluate the association between reproductive disorders and the main serogroups of Leptospira spp. in dairy herds. Blood samples from 216 cows from nine herds were collected for a Microscopic Agglutination Test (MAT), and a reproductive follow-up with ultrasonography was carried out monthly for at least 12 months. A total of 75 (16.5%) cows were reactive, while seroreactivity to a herd level ranged from 16.0% to 52.8%. Out of the nine herds, in five (55.0%) Sejroe was found to be the predominant serogroup and in four (45.0%) it was Icterohaemorrhagiae. The major reproductive problems identified were embryonic death (ED) and abortions. All herds presented ED and abortion was observed in three (33.3%). Herds with high seroreactivity (>20%) had more reproductive problems. Those with a predominance of Sejroe were associated with ED (P < 0.05), while those with Icterohaemorrhagiae were associated with abortion. The results show that different leptospiral strains lead to different reproductive problems, as well as showing that the main problem - ED - is caused by leptospirosis, particularly for those determined by strains of the serogroup Sejroe.

For different ovine breeds to maximize their reproductive capacity in countries with tropical climate, it is important to evaluate their potential for thermal resilience and consequences on their reproductive traits. Therefore, the objective of this study was to evaluate the effect of thermal environment temperatures of climate seasons in a tropical climate region on the surface temperatures of the scrotum, testicular biometric characteristics, seminal quality and serum testosterone concentration of rams of different genotypes. Breeders of four different genotypes (Dorper, n = 8, Texel, n = 8, Santa Inês, n = 9 and Morada Nova, n = 8) were used throughout the four climate seasons. Higher thermal challenge was recorded in the spring and summer. In the summer increase in scrotal surface temperature was detected by infrared thermography (P < 0.05), mainly in the regions of the distal testicular pole and tail of the epididymis. The animals of the Texel genotype had higher rectal temperature in the summer. In spring, this genotype also had the highest testicular pole (32.2 ±â€¯0.5 °C; P < 0.05) and distal (29.9 ±â€¯0.4 °C; P < 0.05) temperatures and a higher mean testicular temperature (31.7 ±â€¯0.4 °C; P < 0.05). The Morada Nova genotype showed a higher surface temperature gradient between testicular poles (2.96 ±â€¯0.1 °C; P < 0.05), especially in spring. Genotype-dependent thermal sensitivity was detected for the thermal gradient between the testicular poles, reflecting the seminal quality. There was a positive correlation of the thermal gradient between testicular poles with sperm membrane integrity and negative correlation with total sperm defects. The Texel genotype showed less progressive motility and higher percentage of sperm defects. There was no difference in testosterone concentration between genotypes and in the different seasons (P > 0.05). Thus, the indigenous genotypes showed a greater capability to maintain the scrotum-testicular thermoregulation. Dorper animals resembled the indigenous sheep genotypes, in terms of seminal characteristics, unlike Texel animals, which showed lower adaptability and lower seminal quality.

ABSTRACT: Bulk tank somatic cell counts (BTSCC) is widely used to monitore the mammary gland health at the herd and regional level. The BTSCC time series from specific regions or countries can be used to compare the mammary gland health and estimate the trend of subclinical mastitis at the regional level. Three time series of BTSCC from dairy herds located in the USA and the Southeastern Brazil were evaluated from 1995 to 2014. Descriptive statistics and a linear regression model were used to evaluate the data of the BTSCC time series. The mean of annual geometric mean of BTSCC (AGM) and the percentage of dairy herds with a BTSCC greater than 400,000 cells mL-1 (%>400) were significantly different (P 0.05) according to the countries and the times series. Linear regression model used for the USA time series was statistically significant for AGM and the %>400 (P 0.05). The first and second USA time series presented an increasing and decreasing trend for AGM and the %>400, respectively. The linear regression model for the Brazil time series was not significant (P>0.05) for both dependent variables (AGM and %>400). The Brazil time series showed no increasing or decreasing trend for the AGM and %>400. Consequently, approximately 40 to 50% of the dairy herds from southeastern Brazil will not achieve the regulatory limits for BTSCC over the next years.


RESUMO: Contagens de células somáticas de tanque (BTSCC) são amplamente utilizadas para monitorar a saúde da glândula mamária em níveis regional e de rebanho. Séries temporais de BTSCC, de regiões ou países específicos, podem ser usadas para comparar a saúde da glândula mamária e estimar a tendência de mastite subclínica em nível regional. Três séries temporais de BTSCC de rebanhos leiteiros, localizados nos EUA e na região sudeste do Brasil, foram avaliadas de 1995 a 2014. A estatística descritiva e modelos de regressão linear foram usados para avaliar as informações das séries temporais de BTSCC. A média anual geométrica de BTSCC (AGM) e a porcentagem de rebanhos com BTSCC, maior que 400.000 células mL-1 (%>400), foram significantemente diferentes (P 0,05) de acordo com os países e as séries temporais analisadas. O modelo de regressão linear usado para as séries temporais dos EUA foram estatisticamente significantes para AGM e %>400 (P 0,05). A primeira e a segunda séries temporais dos EUA apresentaram uma tendência de aumento e diminuição, tanto para AGM quanto para %>400, respectivamente. O modelo de regressão linear para as séries temporais do Brasil não foi significante (P>0,05) para ambas as variáveis dependentes (AGM e %>400). As séries temporais do Brasil não apresentaram tendência de aumento ou diminuição para AGM e %>400. Consequentemente, aproximadamente, 40 a 50% dos rebanhos leiteiros do sudeste do Brasil não alcançarão os limites regulatórios para BTSCC ao longo dos próximos anos.

BACKGROUND: Staphylococcus aureus is associated with chronic mastitis in cattle, and disease manifestation is usually refractory to antibiotic therapy. Biofilm production is a key element of S. aureus pathogenesis and may contribute to the treatment failure that is consistently reported by veterinarians. Minas Gerais State is the largest milk-producing state in Brazil, and the characterization of bacterial isolates is an important aspect of disease control for dairy farmers. Here, we investigated the potential of S. aureus isolated from bovine mastitis to produce slime and biofilm in a skim-milk medium and classified the isolates according to their agr type. RESULTS: Slime was detected using the Congo Red agar (CRA) test in 35.18% (19/54) of the strains; however, 87.04% (47/54) of the strains were considered biofilm-positive based on crystal violet staining. Compared to TSB supplemented with 0.25% glucose, skim milk significantly increased the production of biofilm, but this effect was only observed in slime-producing strains. The bacteria belonged to agr groups I (12/54), II (34/54), III (6/54), and IV (2/54), and bacteria in agr group III were found to be stronger biofilm producers than those in groups I and II. Again, milk had a significant influence only on slime-positive agr I and II isolates, revealing an association between milk and slime. CONCLUSIONS: The present study demonstrated that skim-milk medium and slime production are two factors that together influence biofilm formation by bovine strains of S. aureus. A predominance of bacteria belonging to agr group II was observed, and bacteria from agr group III showed the highest proportion of biofilm producers. The majority of bacteria characterized in this study formed biofilm in milk, which suggests that biofilm formation has an important role in the virulence of S. aureus isolated from bovine intramammary infections.

This study was conducted to establish the relationship between somatic cell count (SCC) and bacterial shedding from mammary quarters according to mastitis pathogens. Milk samples from 638 mammary quarters were examined for mastitis pathogens, SCC and total bacterial count (TBC). The raw data of SCC and TBC were used to perform descriptive statistics. The significance of the arithmetic mean differences between SCC and TBC according to bacteriological examination results was determined by a two-tailed unpaired t-test. Pearson and Spearman´s correlations were done with logarithmic data and linear regression analyses. The geometric means of the bacteriological examination results were (cells mL-1; CFU mL-1): no growth (52,000; 12,000), coagulase-negative staphylococci (85,000; 17,000), Staphylococcus aureus (587,000; 77,000); other streptococci (432,000; 108,000) and Streptococcus agalactiae (1,572,000; 333,000). The Pearson and Spearman's correlations between SCC and TBC were higher than 0.60 for all mastitis pathogens. The regression analyses slopes showed different increase in TBC with the same increase in SCC according to mastitis pathogens. The slope for S. agalactiae (0.542) was higher than that for other mastitis pathogens. The results suggest that the intensity of inflammatory process was associated with number of mastitis pathogens shedding from the mammary gland.


Este estudo foi realizado com objetivo de estabelecer a relação entre contagem de células somáticas (CCS) e a liberação de bactérias de quartos mamários de acordo com os patógenos da mastite. Amostras de leite de 638 quartos mamários foram examinadas para identificação dos patógenos da mastite, CCS e contagem total de bactérias (CTB). Estatísticas descritivas foram utilizadas para avaliar os dados brutos de CCS e CTB. A diferença entre médias para CCS e CTB de acordo com os resultados dos exames bacteriológicos foi avaliada pelo teste T para amostras independentes. Foram realizadas a correlação de Pearson, de Spearman e regressão linear com os dados transformados. As médias geométricas de acordo com os resultados dos exames bacteriológicos foram (células mL-1; UFC mL-1): sem crescimento (52.000; 12.000), estafilococos coagulase negativo (85.000; 17.000), Staphylococcus aureus (587.000; 77000); outros estreptococus (432.000; 108.000) e Streptococcus agalactiae (1.572.000; 333.000). A correlação de Pearson e Spearman entre CCS e CTB foi maior que 0,60 para todos os patógenos da mastite. O coeficiente angular das regressões lineares mostrou diferentes aumentos da CTB como o mesmo aumento da CCS de acordo com os patógenos da mastite. O coeficiente angular para o S. agalactiae (0.542) foi maior em relação aos outros patógenos da mastite. Os resultados sugerem que a intensidade do processo inflamatório foi associada com a quantidade de bactérias da mastite liberada pela glândula mamária.

With the purpose of identifying management factors that may be influencing the prevalence of bovine tuberculosis under tropicalconditions, namely in Rio de Janeiro, Brazil, 1632 cows were tested through the single cervical tuberculin test. A questionnairewas completed for each herd. A total of 207 positive reactions were observed, corresponding to 12.7% of the studied cattle. Themain factors observed that may be influencing the prevalence of bovine tuberculosis on those farms were the absence orreduced veterinary assistance and the herd size. The presence of adequate cattle houses and the highly intensive managementare also considered to be likely to influence the prevalence of the disease. Under tropical conditions, a tuberculosis controlprogram, in addition to the test-and-slaughter control method, should include an investigation of herd management practices totry to identify factors that are likely to influence the prevalence of the disease.

Bactérias patogênicas e resíduos de produtos químicos presentes no ambiente de produção podem contaminar o leite e causarem problemas tecnológicos e à saúde dos consumidores. Esforços para assegurar a qualidade e a segurança do leite e derivados devem ser iniciados na fazenda e para isso é necessária a adoção de boas práticas de produção, que são requisitos para o sistema APPCC (análise de perigos e pontos críticos de controle). O trabalho teve o objetivo de examinar e descrever o nível de adoção de boas práticas agropecuárias na produção de leite. Com ajuda de um questionário, entrevistas e registro fotográfico, foram avaliadas 48 propriedades leiteiras localizadas nos Estados de Minas Gerais e Rio de Janeiro. Havia predominância de gado mestiço Holandês-Zebu (34/48; 71%), seguido por animais da raça Holandesa (19%), Jersey (8%) e Pardo-Suíça (2%). A média de produção de leite dos rebanhos era de 686 ± 403 kg/dia, e o número médio de vacas em lactação era 65 ± 42. Em duas propriedades as vacas em lactação eram mantidas em confinamento total (sistema free stall) e nas demais se adotava o sistema extensivo, a pasto. Os resultados obtidos indicam que grande parte das propriedades adotava as principais práticas consideradas necessárias para a produção de leite de qualidade. Contudo, foram identificados vários pontos que deveriam receber maior atenção para capacitar os produtores
