RESUMO

Salmonella enterica is a pathogenic bacterium responsible for intestinal illness and systemic diseases such as typhoid and paratyphoid fevers. Among clinical manifestation classification, non-typhoidal Salmonella is mainly known as foodborne pathogen associated with the consumption of fecal contaminated food and water. Even though Salmonella hosts include humans and warm-blooded animals, it has been found in non-host environments as river water where the bacteria use different strategies to fitness the environment persisting and establishment. Now with the availability of WGS and bioinformatics tools, we can explore bacterial genomes with higher resolution to increase our understanding of specific genetic signatures among environmental and clinical isolates, being the goal of this work. Pangenome construction allowed the detection of specific environmental and clinical gene clusters related to metabolism and secretion systems as the main signature respectively. Specifically, D-galactonate degradation pathway was observed mainly in environmental genomes while T3SS and flagellum genes were detected for all clinical but not for all environmental isolates. Gene duplication and pseudogenes accumulation were detected as the main adaptation strategy for environmental isolates; thus, isolation source may play an important role in genome plasticity, conferring a selective advantage to survive and persist for environmental Salmonella isolates. Intact prophage sequences with cargo genes were observable for both isolation sources playing an important role in virulence contribution.

RESUMO

Here, we report the genome sequences of three Salmonella enterica strains belonging to serovars Weltevreden (CFSAN047349), Saintpaul (CFSAN047351), and Thompson (CFSAN047352), isolated from river water in Sinaloa, Mexico. The genomes were closed by a combination of long-read and short-read sequencing. The strain sequence types (STs) are ST365, ST50, and ST26, respectively.

This work presents a comparison of the length-weight and length-length allometric relationships between six measurements: length pattern (LP), cephalic length (CL), maximum depth (MA), caudal peduncle depth (CPD) ocular diameter (OD) and weight (W) of three species of the genus Selene (S. brownii, S. vomer and S. setapinnis) caught in the port of Veracruz, Mexico, using a beach seine of 800 m. The results show certain relationships between all measurements of S. brownii (an isometric relationship and four negative allometric relationships); S. vomer showed only four relationships (two isometric and two negative allometric); and S. setapinnis showed three relationships (all negative allometric); the results of S. vomer and S. setapinnis are consistent with those reported by Muto et al. (2010). The comparison of the coefficients of allometry between the three species allowed us to determine that S. brownii and S. vomer differed significantly in two of the four allometric coefficients of length-weight relationships common to them, and in three of the four allometric coefficients of length-length relationships common to them. S. brownii and S. setapinnis differed significantly in two of the three allometric coefficients of length-weight relationships common to them and in the only allometric coefficient of length-length relationships common to them. Finally, S. vomer and S. setapinnis differed significantly in the three common allometric coefficients of length-weight relationships, and in the only common allometric coefficient of length-length relationship.


En este trabajo se presentan las comparaciones de las relaciones alométricas de longitud- peso y longitud-longitud de seis medidas (longitud patrón (LP), longitud cefálica (LC), amplitud máxima (AM), amplitud del pedúnculo (AP), diámetro ocular (DO) y Peso (W) de tres especies del género Selene (S. brownii, S. vomer y S. setapinnis) capturados en el puerto de Veracruz México, mediante el uso de un chinchorro playero de 800 m. Los resultados obtenidos muestran que S. brownii posee relaciones entre todas sus medidas (una isométrica y cuatro alométricas negativas), S. vomer posee solo cuatro relaciones (dos isométricas y dos alométricas negativas) y S. setapinnis posee tres relaciones (todas alométricas negativas), los resultados de S. vomer y S. setapinnis concuerdan con los reportados por Muto et al. (2010). Al comparar los coeficientes de alometría entre las tres especies se pudo determinar que S. brownii y S. vomer difieren significativamente en dos de los cuatro coeficientes alométricos comunes en las relaciones longitud-peso, y tres de los cuatro coeficientes alométricos comunes en las relaciones longitud-longitud; S. brownii y S. setapinnis difieren significativamente en dos de los tres coeficientes alométricos comunes en las relaciones longitud-peso y en el único coeficiente alométricos común en las relaciones longitud-longitud; por último S. vomer y S. setapinnis difieren significativamente en los tres coeficientes alométricos comunes en la relaciones longitud-peso, y en el único coeficiente alométrico común en las relaciones longitud-longitud.

The aim of this work was to determine the presence of galectin-10 in nasal lavage fluid (NLF) of patients with aspirin-sensitive respiratory disease (ASRD) before and after challenge with L-ASA (aspirin) by ELISA. Fifteen ASRD patients, ten aspirin-tolerant asthmatics (ATA), and fifteen healthy controls (HC) were studied. The baseline presence of Galectin-10 in PBMC was determined using real time PCR. Galectin-10 was evaluated in tissue of nasal polyps by western blot. Our results showed a lower expression in PBMC of ASRD patients than in ATA and healthy controls. However, a higher concentration of galectin-10 in NLF was found in ASRD patients before and after L-ASA challenge; western blot confirmed a high expression of galectin-10 in tissue from nasal polyps obtained from ASRD patients. Our results suggest a probable role of galectin-10 in the inflammatory response observed in ASRD patients; however, confirmatory studies are needed.

PURPOSE: The prevalence of diabetes mellitus in Puerto Ricans has been identified and reported as being disproportionately higher as compared to other metabolic pathologies. Recently, diabetes has been identified as the third cause of mortality in Puerto Rico (Puerto Rico Health Department, Vital Statistics Annual Report, 1999-2001). The Research Center, Education and Medical Services for Diabetes in Puerto Rico (also known as the [quot ]Centro de Diabetes para Puerto Rico[quot ] [CDPR]) is a public corporation in the island created by the government to reduce diabetes prevalence, mortality and morbidity. METHOD: The CDPR offers Diabetes Self Management Educational Training Program Schools for patients (DSMETPS) island wide. The research design was an ex-post facto. As part of the process, patients are administered an extensive sociodemographic and health information questionnaire, which also includes the CES-D (a symptomatology depressive scale). This study pretends to describe the diabetic patient profiles (n=27) using information from the DSMETPS of the CDPR and explore the association with the CES-D. Variables such as patients' needs, knowledge and understanding of the condition (i.e., pathology management, type and medications utilized and exercise and nutritional patterns), patient attitudes to diabetes and their relations with the CES-D were explored. RESULTS: Results show a negative association, controlling for age and gender, between patients diabetic education/knowledge and CES-D score. CONCLUSIONS: Diabetes educators in Puerto Rico need to identify depressive symptomatology in order to prevent mental health complications in their patients since this may affect their future treatment and prognosis. An interdisciplinary team is recommended to improve the effectivity of the intervention.

Fourier transform infrared and Raman spectra of dimethylterephthalate (DMT), as microcrystalline powder, have been investigated. The vibrational spectra were calculated using the AM1 and PM3 semi empirical procedures, and the Møller-Plesset (MP2/DZV), and the Becke-Lee, Yang and Parr gradient-corrected correlation functional: B3LYP/6-31G(d) ab initio calculations. On this basis, and assisted with the FT-IR and Raman spectra of the terephthalic acid, an assignment of the vibrational spectra of dimethylterephthalate was proposed. In the calculations, remarkable differences concerning the assignments of the vibrational spectra were noted between the AM1 and PM3 methods. Also, the ab initio procedure shows differences in interpreting the spectra compared with the semi empiric procedures, and among themselves. Calculated geometrical parameters were compared with the experimental values of dimethylterephthalate, diethylterephthalate and terephthalic acid.

This investigation presents the development and validation process of a scale whose principal purpose is to identify those areas in which one is required to achieve improved functioning and adequate emotional health resulting in improved sports performance with adolescent athletes. The "Hechavarria Wellness Scale for Adolescent Athletes" (HWSAA) was developed. The concept of wellness was measured utilizing the following 4 sub-scales measuring; depressive symptomatology, trait anxiety, self-esteem and social relations. A pilot study was undertaken with a universe of 149 adolescent athletes from both sexes with an average of 15.6 years old and residing within the different municipalities of Puerto Rico. The alpha coefficient reliability of the HWSAA and its' subscales fluctuated between 0.71 and 0.82. The results suggest that the HWSAA and its' subscales possess a psychometrically adequate internal consistency. This means that the instrument is valid and reliable for the sample utilized. We suggest further research with the HWSAA

The humoral and cytotoxic T-lymphocyte (CTL) responses have been shown to be determinant in the clearance of many viral infections and because of those characteristics, vaccine candidates against AIDS are designed to enhance both arms of the immune system. While a protocol of immunization able to confer protection in humans against HIV will have to await the results of current clinical trials, it remains important to identify protocols of immunization in animals that achieve significant levels of humoral and cellular immune responses to HIV. In this study we have carried out a comparative analysis of the immune responses elicited in mice immunized with recombinants based on the modified vaccinia virus Ankara strain (rMVA) versus the Western Reserve strain (WR) of vaccinia virus (rVV), both expressing a V3 loop multi-epitopic protein from eight different HIV isolates (TAB13). We found that during priming, rMVA elicited a two- to three-fold higher specific CD8+ T cell response than rVV. Similar enhancement was observed during priming with purified protein TAB13 followed by a booster with rMVA. The epitopes LR150, MN and IIIB, located at the ends and in the middle of the chimeric protein, were able to induce a specific CD8+ T cell response, both after priming or prime/booster with the recombinant viruses but not after prime/booster with TAB13. By examining the cytokine pattern, the immune response triggered by these vectors was of Th-1 type. Humoral immune responses were higher in animals immunized with TAB13/TAB13 or TAB13/rVV than in animals immunized with TAB13/rMVA. These findings demonstrate that during priming or in a prime/booster immunizations, rMVA is superior to rVV in the ability to enhance specific cellular responses to an HIV-1 protein, and that both humoral and cellular immune responses to theV3 loop epitope of HIV-1 Env can be obtained by priming with TAB13 followed by a booster with viral vectors.

Adriamycin (ADM) is an oncostatic of the anthracycline family with confirmed experimental and clinical efficiency. This antitumoral drug has been reported to stimulate macrophage activity and is able to induce apoptosis (AP) in some tumour cells. The objective of the present work was to investigate if in vivo administration of ADM to mice induces AP in their peritoneal macrophages (PM). AP was expressed by the apoptotic index (AI) of peritoneal macrophages observed under fluorescence microscope after ethidium bromide and acridine orange staining and confirmed by detection of the ladder pattern on DNA electrophoresis, indicates DNA fragmentation in 80-120 bp characteristic of apoptotic state. 24 hours after i.p. ADM administration, AP was observed in PM. The effect was best visible after the injection of 5 mg/kg ADM. (Al: 76.3+/-8.9 vs untreated control group AI: 2.8+/-1.1). In the ADM treated group a DNA ladder electrophoretic pattern was observed while DNA from normal PM was genomic. Since ADM toxicity has been attributed to reactive oxygen species generation, we investigated its possible participation in AP induction by pretreating mice with antioxidants: (+)-alpha-tocopherol acid succinate (30 IU/mouse per os) for 3 days before ADM administration with E. coli lipopolysacharide (0.15 microg/mouse i.p.) 24 hours before ADM administration or with superoxide dismutase (10,000 IU/mouse i.p.) 1 hour before ADM administration. AI was significantly decreased, with values close to those of the untreated control group (AI: 15+/-5.7, 9.6+/-8.0 and 32.9+/-6.9, respectively). Antioxidants given before ADM treatment significantly increased the live cell index (p < or = 0.001) in PM the groups while inactivated antioxidants no longer protect PM against the ADM AP induction. DNA analysis confirmed the effect: in the untreated control and in the antioxidant protected groups DNA was genomic while in either ADM or inactivated-antioxidants + ADM treated groups, DNA presented the ladder pattern. AP can thus be induced in PM by ADM and inhibited by antioxidants. These observations may have clinical applications.
