RESUMO

The association among environmental cues, ethylene response, ABA signaling, and reactive oxygen species (ROS) homeostasis in the process of seed dormancy release is nowadays well-established in many species. Alternating temperatures are recognized as one of the main environmental signals determining dormancy release, but their underlying mechanisms are scarcely known. Dry after-ripened wild cardoon achenes germinated poorly at a constant temperature of 20, 15, or 10 °C, whereas germination was stimulated by 80% at alternating temperatures of 20/10 °C. Using an RNA-Seq approach, we identified 23,640 and annotated 14,078 gene transcripts expressed in dry achenes and achenes exposed to constant or alternating temperatures. Transcriptional patterns identified in dry condition included seed reserve and response to dehydration stress genes (i.e., HSPs, peroxidases, and LEAs). At a constant temperature, we observed an upregulation of ABA biosynthesis genes (i.e., NCED9), ABA-responsive genes (i.e., ABI5 and TAP), as well as other genes previously related to physiological dormancy and inhibition of germination. However, the alternating temperatures were associated with the upregulation of ethylene metabolism (i.e., ACO1, 4, and ACS10) and signaling (i.e., EXPs) genes and ROS homeostasis regulators genes (i.e., RBOH and CAT). Accordingly, the ethylene production was twice as high at alternating than at constant temperatures. The presence in the germination medium of ethylene or ROS synthesis and signaling inhibitors reduced significantly, but not completely, germination at 20/10 °C. Conversely, the presence of methyl viologen and salicylhydroxamic acid (SHAM), a peroxidase inhibitor, partially increased germination at constant temperature. Taken together, the present study provides the first insights into the gene expression patterns and physiological response associated with dormancy release at alternating temperatures in wild cardoon (Cynara cardunculus var. sylvestris).

OBJECTIVE: With high-resolution real-time ultrasonography we investigated the muscle architectural parameters of vastus lateralis in healthy volunteers. PURPOSES: We determined the reproducibility and validity of ultrasonography and the role of the ultrasonographer in assessing muscle architecture. We proposed the most appropriate clinical parameters for objective measurements and an ultrasound protocol of muscle architecture. METHODS: We conducted an intraobserver and interobserver study. We investigated 21 healthy male volunteers. The subjects were independently evaluated by four different operators using high-resolution real-time ultrasonography. To assess the reproducibility of ultrasound examinations, four operators repeated measurements using the same ultrasound device. Muscle thickness, muscle volume, muscle fiber pennation angle, and subcutaneous adiposity of the vastus lateralis muscle were measured. RESULTS: Intra-observer (ICC 0.92-0.97), interobserver (ICC 0.78-0.92) reproducibility was good to excellent for all measurements. CONCLUSION: Simple, reproducible, non-invasive ultrasound measurements of muscle structure easily demonstrated differences in muscle morphology. With a protocol and with objective and repeatable measurements, sonographers from different backgrounds could obtain an objective measurement of ultrasound images with little differences and low variability in results, thanks to the upgrading of diagnostic ultrasound imaging and their clinical skills.

RESUMO OBJETIVO: Nós utilizamos ultrassonografia de alta resolução em tempo real para investigar os parâmetros da arquitetura muscular do vasto lateral em voluntários saudáveis. PROPÓSITO: Nós determinamos da reproduzibilidade e validade da ultrassonografia e o papel do ultrassonografista na avaliação da arquitetura muscular. Nós propomos os parâmetros clínicos mais apropriados para uma medição objetiva e um protocolo de ultrassonografia para arquitetura muscular. MÉTODOS: Nós conduzimos um estudo intra-observador e inter-observador. Investigamos 21 voluntários saudáveis do sexo masculino. Os participantes foram avaliados de forma independente por quatro operadores diferentes usando ultrassonografia de alta-resolução em tempo real. Para avaliar a reprodutibilidade dos exames de ultrassonografia, quatro operadores repetiram as medições usando o mesmo equipamento de ultrassonografia. A espessura e o volume do músculo, o ângulo de penação da fibra muscular, adiposidade subcutânea do músculo vasto lateral foram medidos. RESULTADOS: A reprodutibilidade intra-observador (ICC 0,92-0,97) e inter-observador (ICC 0,78-0,92) foi boa a excelente para todas as medições. CONCLUSÃO: Medições de ultrassonografia simples, reprodutíveis e não-invasivas da estrutura muscular demostraram facilmente as diferenças na morfologia muscular. Seguindo um protocolo e com medições objetivas e reproduzíveis, ultrassonografistas com diferentes formações e experiências podem conseguir uma medição objetiva de imagens de ultrassonografia com poucas diferenças e pouca variação nos resultados graças à melhoria da qualidade nos exames de diagnóstico por imagem de ultrassonografia e das habilidades clínicas.

A series of experiments were conducted to evaluate the toxicity of lemon peel extracts incorporated into mediterranean fruit fly Ceratitis capitata diet. Extracts were obtained with different solvents: diethyl ether, ethyl acetate, and methanol. All three extracts were toxic to some extent; the diethyl ether extract was selected for further studies. Ether extracts of lemon peel were prepared weekly over a 2-month period, from fruits collected on the 1st d of the bioassay. Weekly GC-MS and UV analyses of the extracts demonstrated that the concentration of citral and coumarins decreased in the peel after harvest. We conducted a series of bioassays to evaluate the toxicity of the ether extract, and mixtures of this extract with citral, 5,7-dimethoxycoumarin. and linalool incorporated to C. capitata larvae's natural diet (lemon slices endocarp) at a concentration of 250 microg/g of diet. Significant larvicidal activity can be obtained from a fresh lemon peel extract; however, when the extract was obtained from stored lemons, toxicity decreased. Addition of small amounts of citral or 5,7-dimethoxycoumarin, and linalool to the stored lemon peel extract would bring back the toxicity to the rates of fresh lemons extracts. Finally, female adults of C. capitata fed on diets containing additional amounts of ether extract, 5,7-dimethoxycoumarin, and linalool, were exposed to different photoperiods to test for phototoxicity. The treatment was toxic and affected the oviposition capacity of females depending on photoperiod.

A collection of Aspergillus fumigatus mutants highly resistant to itraconazole (RIT) at 100 micro g ml(-1) were selected in vitro (following UV irradiation as a preliminary step) to investigate mechanisms of drug resistance in this clinically important pathogen. Eight of the RIT mutants were found to have a mutation at Gly54 (G54E, -K, or -R) in the azole target gene CYP51A. Primers designed for highly conserved regions of multidrug resistance (MDR) pumps were used in reverse transcriptase PCR amplification reactions to identify novel genes encoding potential MDR efflux pumps in A. fumigatus. Two genes, AfuMDR3 and AfuMDR4, showed prominent changes in expression levels in many RIT mutants and were characterized in more detail. Analysis of the deduced amino acid sequence encoded by AfuMDR3 revealed high similarity to major facilitator superfamily transporters, while AfuMDR4 was a typical member of the ATP-binding cassette superfamily. Real-time quantitative PCR with molecular beacon probes was used to assess expression levels of AfuMDR3 and AfuMDR4. Most RIT mutants showed either constitutive high-level expression of both genes or induction of expression upon exposure to itraconazole. Our results suggest that overexpression of one or both of these newly identified drug efflux pump genes of A. fumigatus and/or selection of drug target site mutations are linked to high-level itraconazole resistance and are mechanistic considerations for the emergence of clinical resistance to itraconazole.

This study presents the results of 26 months of work, from January 1996 through February 1998, of the Distinct Home Hospitalization Service for Acute Patients. This service managed 20 home beds with two teams, each with a physician and a nurse, with a care approach similar to that for a room in a hospital. Among the items evaluated were the attributes of the admitted population, their illnesses, the form of administering drugs, the satisfaction of the caregivers, the indices of performance, and the costs with this approach. A total of 1,789 patients had home hospitalizations over the period, with a median stay of 4 days. Of the patients, 76.5% were admitted from in-hospital care. The most frequent illnesses were cardiorespiratory ones (45.5%), and the proportion of patients with a terminal illness was 14.2%. Drugs were administered orally with 74% of the patients and parenterally in 26%. The patients' satisfaction level was very high, and the cost of the hospitalization was 70% of that for in-hospital care. Family involvement was key in this approach to care.

We revised retrospectively 30 cases of Spontaneous Infectious Spondylodiskitis (SIS) in adults, diagnosed between 1986 and 1997. The mean age of the patients was 68.8 years; 56.7% were males. The identifiable causes were infectious endocarditis 13 (43.3%); tuberculosis 7 (23.3%); urinary tract infection 4 (13.3%); bacteremia with focus 2 (6.7%) and without focus 2 (6.7%). The cause was not identified in other 2 cases (6.7%). Infections were due to pyogenic bacteriae in 19 (63.3%); tuberculosis 6 (20%) and unknown 5 (16.7%). All patients had localized pain, 70% fever, 36.7% irradiated pain and 23.3% paraparesis. Fever was more frequent in patients with pyogenic etiology than in those with tuberculous SIS (p = 0.004). Blood cultures were positive in 70.4%. Percutaneous aspiration of the disc was performed in 13 patients; cultures were positive in 7. Causal germs were Streptococcus spp. 33.3%; Mycobacterium tuberculosis 20%; Staphylococcus spp. 16.6%; Escherichia coli 6.6%; Pseudomonas aeruginosa 6.6%. There was no bacteriological recovery in 5 (16.7%). Localization was lumbar in 18 (60%), dorsal in 8 (26.6%) and cervical in 4 (13.3%). X-ray of the spine was positive in 63.3% of the cases. Technetium scan in 90.5%, CT in 85.7% and MRI in 100% of cases in which it was carried out. All patients received antibiotic treatment with a median duration of 6 weeks for pyogenic SIS and one year for tuberculous SIS. Eighty three percent required immobilizing brace and 10% surgery for stabilization. Thirty six percent of patients presented complications, most of them related to the causal disease. There was a statistically significant association between mortality and diabetes.

Hypersensitivity pneumonitis (HP) is the clinical manifestation of an pulmonary immunological reaction to inhaled antigens. The list of provocative antigens and specific illnesses grows permanently. A woman of 56 years consulted for dyspnea and cough of two months of evolution. She worked in a factory of sausages removing the dust that recovers the salamis during their stationing. She presented hypoxemia and the pulmonary function test revealed a predominantly obstructive pattern. The x-ray of the thorax was normal and the high resolution computed tomography showed a mosaic pattern. A lung biopsy confirmed the diagnosis of HP. She improved with systemic corticosteroids and the avoidance of antigen exposure. In the cultures of the material that recovers the salamis, Penicillium spp. was recovered and was interpreted as the probable etiological agent of this HP, which we have denominated "the salami worker's lung".
