RESUMO

Stray dogs are considered as sentinels in the epidemiology of Toxoplasma gondii because they are carnivores and eat a variety of foods, including garbage. In the present study, tissues and sera of 249 stray dogs (Canis familiaris) from Grenada, West Indies were examined for T. gondii infection. Sera were examined for antibodies to T. gondii by the modified agglutination test (MAT); 89 (35.7%) of 249 were seropositive with titers of 25 in seven dogs, 50 in 22 dogs, 100 in 22 dogs, 200 or higher in 38 dogs. Hearts of 76 seropositive dogs were bioassayed in mice. Viable T. gondii was isolated from 12 dogs; these isolates were designated TgDogGr1 to TgDogGr12. These isolates were further propagated in cell culture and DNA isolated from cell culture derived tachyzoites of 12 isolates was genotyped using 10 PCR-restriction fragment length polymorphism markers (SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico). The results revealed six genotypes, including ToxoDB PCR-RFLP #1, #2, #3, #7, #13 and #224, with 1, 6, 1, 2, 1 and 1 isolates, respectively. The result supports previous findings that T. gondii population genetics is highly diverse in Grenada.

Nothing is known of the genetic diversity of Toxoplasma gondii circulating in wildlife in Mexico. In the present study, a mouse virulent T. gondii strain was isolated from the heart of a wild puma (Felis concolor). The puma was found roaming in outskirt of Durango City, Mexico and tranquilized for moving to a zoo. The puma died during translocation and a necropsy examination was performed. The puma had an antibody titer for T. gondii of 200 by the modified agglutination test. Its heart and brain tissue were bioassayed into 2 outbred Swiss Webster (SW) and 1 gamma interferon gene knockout (KO) mouse. The KO mouse and the 2 SW mice that became infected after inoculation with homogenate of puma heart died of acute toxoplasmosis 12, 19 and 20 days p.i. respectively and tachyzoites were found in lungs of all 3 mice. None of the 4 SW and 1 KO mouse inoculated with digest of the puma brain became infected with T. gondii. Tachyzoites from the lungs of mice were propagated in cell cultures. Tachyzoites from cell culture were inoculated into 5 SW; the mice died or had to be killed 14 days p.i. and a cat fed tissues of these mice shed T. gondii oocysts. Results of mortality and infectivity of tachyzoites and oocysts in SW mice indicated that the puma T. gondii strain (designated TgPumaMe1) was virulent for outbred mice. DNA isolated from culture-derived tachyzoites was characterized using 11 PCR-RFLP markers (SAG1, 5'- and 3'-SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 and Apico) revealed a new genotype (ToxoDB PCR-RFLP #222). Isolation of atypical genotype T. gondii from wild puma indicates that mouse virulent strains are circulating in wildlife in Mexico.

BACKGROUND AND OBJECTIVE: Simple Measure of the Impact of Lupus Erythematosus in Youngsters (SMILEY) is a health-related quality of life (HRQOL) assessment tool for pediatric systemic lupus erythematosus (SLE), which has been translated into Portuguese for Brazil. We are reporting preliminary data on cross-cultural validation and reliability of SMILEY in Portuguese (Brazil). METHODS: In this multi-center cross-sectional study, Brazilian children and adolescents 5-18 years of age with SLE and parents participated. Children and parents completed child and parent reports of Portuguese SMILEY and Portuguese Pediatric Quality of Life Inventory (PedsQL™) Generic and Rheumatology modules. Parents also completed the Childhood Health Assessment Questionnaire (CHAQ). Physicians completed the SLE disease activity index (SLEDAI), Physician's Global Assessment of disease activity (PGA) and Systemic Lupus Erythematosus International Collaborating Clinics ACR Damage Index (SDI). RESULTS: 99 subjects (84 girls) were enrolled; 93 children and 97 parents filled out the SMILEY scale. Subjects found SMILEY relevant and easy to understand and completed SMILEY in 5-15 minutes. Brazilian SMILEY was found to have good psychometric properties (validity and reliability), and the child-parent agreement was moderate. CONCLUSION: SMILEY may eventually be used routinely as a research/clinical tool in Brazil. It may be also adapted for other Portuguese-speaking nations offering critical information regarding the effect of SLE on HRQOL for children with SLE.

BACKGROUND: There is an unmet therapeutic need in endocrine-resistant, hormone receptor (HR)-positive, human epidermal growth factor receptor 2-negative advanced breast cancer (BC). Preclinical studies support the hypothesis that the mammalian target of rapamycin (mTOR) inhibition could potentially overcome resistance to endocrine therapy. MATERIALS AND METHODS: A literature review regarding BC and mTOR inhibitors was undertaken. The reference lists from retrieved manuscripts were reviewed to identify further studies. RESULTS: Phase II studies have reported that the combination of mTOR inhibitors with endocrine therapy shows efficacy in patients with advanced disease that progressed after treatment with aromatase inhibitors. The recent findings of the phase III BOLERO-2 confirmed that everolimus in combination with exemestane significantly improved progression-free survival and response rate, with a manageable safety profile. CONCLUSIONS: The addition of everolimus to exemestane for women with HR-positive metastatic BC is now considered a new therapeutic strategy. However, a word of caution should be added regarding toxic effects, which might limit practical use and compliance. It is essential that clinicians are educated about key recommendations for toxicity management and specific guideline dose modifications. Additional research efforts with the addition of these compounds in the early-stage setting is greatly needed to improve the survival of patients with HR-positive BC.

RESUMO

A suppressed ion chromatography (IC) technique has been evaluated as a chemical monitoring tool for detecting major anions (F-, Cl-, NO3- and SO4(2-)) of condensed steam in geothermal power plants. It is shown that the suppressed IC technique provides a suitable means for preventing possible damage to generating equipment in the geothermal industry. An electrical conductivity detector (0.1 microS sensitivity) with an anion-exchange column (IonPac AS4A-SC), a micro-membrane suppressor (AMMS II), and an isocratic high-pressure pump system were successfully used for detecting low concentrations of inorganic anions. Method detection limits for the anions of interest were <0.184 mg/L. Details of the IC methodology as well as some experimental results obtained during its application for the chemical monitoring of geothermal steam pipes are also described.

RESUMO

Epidemic dropsy, a disease due to Argemone mexicana poisoning, is characterized by pathological accumulation of diluted lymph in body tissues and cavities. Recently, the largest epidemic of the disease in India affected Delhi and its neighboring states during the months of August-September 1998. Over 3000 persons fell ill, and more than 65 died in the state of Delhi alone. Two cases belonging to the same family died, out of the large number of cases admitted in this tertiary care teaching hospital situated in eastern part of Delhi. Autopsy findings of these two cases are presented and discussed here along with the review of toxicity due to this poisoning.

Groundwaters originating from local and regional aquifers surrounding ash deposits produced by a coal-fired power plant were collected. These water samples were chemically analyzed for quantifying their heavy metal composition at trace levels. A highly sensitive analytical technique based on ion chromatography with a UV-Vis detector and under isocratic eluent flow-rate conditions was used. In order to quantify the major heavy metals (Pb, Cu, Cd, Co, Zn and Ni), three ionic separation column systems were evaluated: (1) a cationic column (HPIC-CS2, Dionex) tested with two eluents (10 mM oxalic acid-7.5 mM citric acid; and 40 mM D-tartaric acid-12 mM citric acid); (2) an anionic column (HPIC-AS4, Dionex) evaluated with 25 mM oxalic acid as eluent: and (3) a bifunctional ion-exchange column (Ionpac CS5, Dionex) which was also tested with two eluents (6 mM pyridine, 2,6-dicarboxylic acid; and 50 mM oxalic acid/95 mM lithium hydroxide). The lowest detection limits achieved with the Ionpac CS5 column and the 50 mM oxalic acid-95 mM lithium hydroxide eluent enabled the heavy metal analysis in groundwater samples to be reliably performed. Details of this comparative study, including the ion chromatography procedure selected and its application to heavy metal analysis of groundwater samples, are presented in this work.

An improved capillary electrophoresis (CE) method for quantifying rare-earth elements (REEs) in synthetic geochemical standards was developed. Synthetic standard solutions were obtained from high purity metal oxides. The separation of REE total group (lanthanum to lutetium) was defined as a primary objective. Special attention was also focused on the optimized separation of europium (Eu) and gadolinium (Gd) because in earlier applications they presented overlapping problems. Their separation and quantitative determinations are essential for geological applications. For the rapid separation of REEs in synthetic geochemical standards, the temperature of the separation device was optimized. An analysis temperature of 15 degrees C enabled both the rapid separation of REEs within 2 min and the overlapping problem of Eu-Gd to be resolved. The detection limits (<0.1 ng) and precision estimates (generally better than 5%) were found to be satisfactory for most geological applications.
