RESUMO

Synaptic plasticity constitutes a fundamental process in the reorganization of neural networks that underlie memory, cognition, emotional responses, and behavioral planning. At the core of this phenomenon lie Hebbian mechanisms, wherein frequent synaptic stimulation induces long-term potentiation (LTP), while less activation leads to long-term depression (LTD). The synaptic reorganization of neuronal networks is regulated by serotonin (5-HT), a neuromodulator capable of modify synaptic plasticity to appropriately respond to mental and behavioral states, such as alertness, attention, concentration, motivation, and mood. Lately, understanding the serotonergic Neuromodulation of synaptic plasticity has become imperative for unraveling its impact on cognitive, emotional, and behavioral functions. Through a comparative analysis across three main forebrain structures-the hippocampus, amygdala, and prefrontal cortex, this review discusses the actions of 5-HT on synaptic plasticity, offering insights into its role as a neuromodulator involved in emotional and cognitive functions. By distinguishing between plastic and metaplastic effects, we provide a comprehensive overview about the mechanisms of 5-HT neuromodulation of synaptic plasticity and associated functions across different brain regions.

Serotonergic neurons constitute one of the main systems of neuromodulators, whose diffuse projections regulate the functions of the cerebral cortex. Serotonin (5-HT) is known to play a crucial role in the differential modulation of cortical activity related to behavioral contexts. Some features of the 5-HT signaling organization suggest its possible participation as a modulator of activity-dependent synaptic changes during the critical period of the primary visual cortex (V1). Cells of the serotonergic system are among the first neurons to differentiate and operate. During postnatal development, ramifications from raphe nuclei become massively distributed in the visual cortical area, remarkably increasing the availability of 5-HT for the regulation of excitatory and inhibitory synaptic activity. A substantial amount of evidence has demonstrated that synaptic plasticity at pyramidal neurons of the superficial layers of V1 critically depends on a fine regulation of the balance between excitation and inhibition (E/I). 5-HT could therefore play an important role in controlling this balance, providing the appropriate excitability conditions that favor synaptic modifications. In order to explore this possibility, the present work used in vitro intracellular electrophysiological recording techniques to study the effects of 5-HT on the E/I balance of V1 layer 2/3 neurons, during the critical period. Serotonergic action on the E/I balance has been analyzed on spontaneous activity, evoked synaptic responses, and long-term depression (LTD). Our results pointed out that the predominant action of 5-HT implies a reduction in the E/I balance. 5-HT promoted LTD at excitatory synapses while blocking it at inhibitory synaptic sites, thus shifting the Hebbian alterations of synaptic strength towards lower levels of E/I balance.

Background: The species Dasyprocta prymnolopha is a wild rodent with a geographic distribution that extends from Mexico to South America, including Brazil. Agouti has been the subject of morphophysiological research, but data on the cardiovascular system remains limited. Therefore, the objective was to describe the macroscopic and microscopic morphology, including the study of the cardiac and coronary system syntopy of the species D. prymnolopha. Materials, Methods & Results: Twelve Dasyprocta primnolopha adults were used, 6 males and 6 females. Topographic analysis of the heart was evaluated in situ, with subsequent measurement, anatomovascular description and macroscopic study of cardiac and coronary vascularization. A microscopic investigation and identification of structural cardiac aspects were also carried out in adult agoutis, the biological samples of the heart were submitted to histological techniques and stained with Hematoxylin and Eosin and Masson's Trichrome. The heart is located between the end of the 2nd and the beginning of the 5th intercostal space, with the apex reaching the 6th space. It presents the presence of 2 ligaments: phrenopericardium and sternopericardium. The right atrioventricular valve is composed of 2 cusps, the parietal and the septal, with variations within the same species for 3 cusps. Projections that interconnect the papillary muscles with each other were observed. In the left ventricle there are variations in the number of papillary muscles (range 2 to 4). In the region of the aortic valve, 5 ostia were observed in the left aortic sinus in all animals. The coronary circulation has a predominantly left distribution. Histologically, the heart consists of 3 main layers: endocardium, myocardium, and epicardium. The cusp valves of the atrioventricular openings are composed of endocardial folds that contain a central plate of dense connective tissue, and inserted in this cardiac musculature was observed the cardiac skeleton, with its fibrous ring consisting of collagen and elastic fibers that surrounds the atrioventricular opening. Discussion: Cardiac assessment in wild animals is challenging, as in-depth knowledge of the morphology of the cardiovascular system is required for the use of diagnostic tools. In this first anatomical study of the heart, this organ presents syntopy with those of other rodents, but the topography may vary in individuals of the same species, which may be related to the accentuated vertebral curve. The phrenopericardial and sternopericardial ligaments were observed in this research, although there are no reports in other species of Dasyprocta sp. The arrangement of the arteries has, as a particularity, the larger left atrium in relation to the heart/atrium size ratio when compared to other domestic species, covering the left coronary sinus until it reaches the left atrial surface. In the agouti, it was observed that the vascularization is left, with the left coronary artery giving rise to both the paraconal and subsinuous interventricular branch, a fact found in animals such as ruminants, dogs. In the histological observations of the present study, the heart was similar to that of other mammals. Our data reveal morphological characteristics similar to those of other mammals, but with very expressive characteristics that differ even within the species. It is important to generate new information to elucidate cardiac and coronary diagnostic analyses, which can be extended to different species.

Biomaterials used in tissue regeneration processes represent a promising option for the versatility of its physical and chemical characteristics, allowing for assisting or speeding up the repair process stages. This research has characterized a polyurethane produced from castor oil monoacylglyceride (Ricinus communis L) and tested its effect on reconstructing bone defects in rat calvaria, comparing it with commercial castor oil polyurethane. The characterizations of the synthesized polyurethane have been performed by spectroscopy in the infrared region with Fourier transform (FTIR); thermogravimetric analysis (TG/DTG); X-ray diffraction (XRD) and Scanning Electron Microscopy (SEM). For the in vivo test, 24 animals have been used, divided into 3 groups: untreated group (UG); control group treated with Poliquil® castor polyurethane (PCP) and another group treated with castor polyurethane from the Federal University of Piauí - UFPI (CPU). Sixteen weeks after surgery, samples of the defects were collected for histological and histomorphometric analysis. FTIR analysis has shown the formation of monoacylglyceride and polyurethane. TG and DTG have indicated thermal stability of around 125 °C. XRD has determined the semi-crystallinity of the material. The polyurethane SEM has shown a smooth morphology with areas of recesses. Histological and histomorphometric analyzes have indicated that neither CPU nor PCP induced a significant inflammatory process, and CPU has shown, statistically, better performance in bone formation. The data obtained shows that CPU can be used in the future for bone reconstruction in the medical field.

In this study, we demonstrated the potential associative effect of combining conventional amphotericin B (Amph B) with gallic acid (GA) and with ellagic acid (EA) in topical formulations for the treatment of cutaneous leishmaniasis in BALB/c mice. Preliminary stability tests of the formulations and in vitro drug release studies with Amph B, GA, Amph B plus GA, EA, and Amph B plus EA were carried out, as well as assessment of the in vivo treatment of BALB/c mice infected with Leishmania major After 40 days of infection, the animals were divided into 6 groups and treated twice a day for 21 days with a gel containing Amph B, GA, Amph B plus GA, EA, or Amph B plus EA, and the negative-control group was treated with the vehicle. In the animals that received treatment, there was reduction of the lesion size and reduction of the parasitic load. Histopathological analysis of the treatments with GA, EA, and combinations with Amph B showed circumscribed lesions with the presence of fibroblasts, granulation tissue, and collagen deposition, as well as the presence of activated macrophages. The formulations containing GA and EA activated macrophages in all evaluated parameters, resulting in the activation of cells of the innate immune response, which can generate healing and protection. GA and EA produced an associative effect with Amph B, which makes them promising for use with conventional Amph B in the treatment of cutaneous leishmaniasis.
