RESUMO

The study evaluated the reliability and repeatability of the force and surface electromyography activity (EMG) outcomes obtained through voluntary and electrically evoked contractions of knee extensors in females (n = 18) and males (n = 20) and compared these data between sexes. Maximal isometric voluntary contractions (iMVCs) of knee extensors associated with electrical stimulation of the femoral nerve were performed over 4 days (48-h interval), with the first day involving familiarization procedures, the second involving three trials (1-h interval), and the third and fourth involving just one trial. The intraclass correlation coefficient (ICC), coefficient of variation (CV), and repeatability of outcomes from within- and between-day trials were determined for each sex. Females presented lower maximal voluntary force during iMVC (iMVCForce) and associated vastus lateralis EMG activity (root mean square, RMSVL), force evoked by potentiated doublet high-frequency (Db100Force) and single stimuli (Qtw), and M-wave amplitude than males (P ≤ 0.01, partial eta squared ≥0.94). Voluntary activation (VA) and RMSVL/M-wave amplitude did not differ between sexes. iMVCForce, VA, Db100Force, Qtw, and M-wave amplitude were the most reliable outcomes in within-day trials, with similar results between sexes (ICC > 0.62; CV < 6.4%; repeatability: 12.2%-22.6%). When investigating between-day trials, the iMVCForce, VA, Db100Force, and Qtw were the most reliable (ICC > 0.66; CV < 7.5%; repeatability: 13.2%-33.45%) with similar results between sexes. In conclusion, females presented lower iMVCForce and evoked response than males. Although reliability and repeatability statistics vary between trials, data (e.g., from EMG or force signal), and sexes, most of the outcomes obtained through this technique are reliable in females and males.NEW & NOTEWORTHY Although reliability and repeatability of knee extensors vary according to the type of neuromuscular function outcome (e.g., from force or EMG responses), the trial intervals (i.e., hours or days), and the sex of the participant, most force and EMG outcomes obtained through these neuromuscular assessment protocols present ICC > 0.75, very good CV (<10%), and repeatability <25% in within- and between-day trials in both sexes.

BACKGROUND: Betaglycan, also known as the TGFß type III receptor (Tgfbr3), is a co-receptor that modulates TGFß family signaling. Tgfbr3 is upregulated during C2C12 myoblast differentiation and expressed in mouse embryos myocytes. RESULTS: To investigate tgfbr3 transcriptional regulation during zebrafish embryonic myogenesis, we cloned a 3.2 kb promoter fragment that drives reporter transcription during C2C12 myoblasts differentiation and in the Tg(tgfbr3:mCherry) transgenic zebrafish. We detect tgfbr3 protein and mCherry expression in the adaxial cells concomitantly with the onset of their radial migration to become slow-twitch muscle fibers in the Tg(tgfbr3:mCherry). Remarkably, this expression displays a measurable antero-posterior somitic gradient expression. CONCLUSIONS: tgfbr3 is transcriptionally regulated during somitic muscle development in zebrafish with an antero-posterior gradient expression that preferentially marks the adaxial cells and their descendants.

OBJECTIVE: The goal of this study was to evaluate and compare lower-leg muscle fatigue, edema, and discomfort induced by the prolonged standing of security guards wearing regular socks and those wearing 15-20 or 20-30 mmHg compression stockings as intervention. BACKGROUND: Compression stockings are sometimes used by individuals standing all day at work. However, quantitative evidence showing their potential benefits for lower-leg health issues in healthy individuals during real working conditions is lacking. METHOD: Forty male security employees participated in the study. All were randomly assigned to the control or one of the two intervention groups (I15-20 or I20-30). Lower-leg muscle twitch force, volume, and discomfort ratings were measured before and after their regular 12-hr standing work shift. RESULTS: Significant evidence of lower-leg long-lasting muscle fatigue, edema, and discomfort was observed after standing work for guards wearing regular socks. However, no significant changes were found for guards wearing either compression stockings. CONCLUSION: In healthy individuals, compression stockings seem to attenuate efficiently the tested outcomes in the lower leg resulting from prolonged standing. APPLICATION: Occupational activities requiring prolonged standing may benefit from 15-20 or 20-30 mmHg compression stockings. As similar benefits were observed for both levels of compression, the lower level may be sufficient.

The purpose of this study was to investigate the effects of three simulated goalball games on neuromuscular, physiological, perceptual, and technical parameters. Ten male players underwent assessments before and immediately after each game. Heart rate was recorded at rest and during all games that were entirely filmed for further technical performance analysis. Exercise significantly decreased knee extensor muscles peak force and percentage of voluntary activation after the second and third games, indicating the presence of central fatigue. Heart rate responses remained predominantly in a range equivalent to moderate activity intensity in all games. In addition, perceptual parameters were associated with reduced frequency of throws and density of actions. These findings suggest significant implications for the management of physical training, game strategy during a competition, and fixture change from three to two games per day.

Purpose: The objective of this study was to evaluate a plyometric conditioning activity (3 sets of 5 countermovement jumps, [CA]) for twitch properties and voluntary knee extension. Methods: After a familiarization session, fourteen highly trained sprint athletes, 12 men (23.25 ± 7.17 years) and 2 women (23.0 ± 2.8 years) performed 2 experiments, each in a randomized order (crossover design). In one experiment, the time-course of twitch contractile properties was evaluated with and without the previous CA at 2, min intervals to 10 min of recovery. In the second session, maximal voluntary knee extension was evaluated at the same recovery intervals, for control and experimental condition in random order. Results: Mixed-model ANOVA with Bonferroni post-hoc revealed significant differences between pre-test and 2 min (p < 0.01, ES = 0.42) and 4 min (p < 0.01, ES = 0.20) for peak twitch torque of quadriceps femoris muscles confirming postactivation potentiation [PAP] at these times. Twitch rate of torque development (RTD) was significantly greater than pre-test value only at 2 min (p < 0.01, ES = 0.58) after the CA. Twitch contraction time and ½ relaxation time were not significantly difference from pre-test values after the CA (p > 0.05). No significant difference was observed for voluntary RTD following CA. Conclusion: The plyometric CA increased twitch peak torque and RTD consistent with PAP; however, there was no effect of CA on voluntary RTD of knee extension at any time after the plyometric CA. Even with PAP confirmed, we observed that the CA fails to improve isometric RTD of quadriceps femoris muscles. HighlightsA plyometric CA significantly increased twitch peak torque (at 2 and 4 min) and twitch rate of torque development (at 2 min) of quadriceps femoris muscles, indicating postactivation potentiation (PAP).No effect was observed for twitch contraction time and ½ relaxation time after the CA.No improvement was observed on voluntary rate of torque development evaluated at the same time intervals.

BACKGROUND: The relevance of local twitch response (LTR) during dry needling technique (DNT) is controversial, and it is questioned whether LTR is necessary for successful outcomes. Furthermore, because the LTR during the deep DNT may be evoked with different intensities, it is unknown whether the magnitude of LTR intensity is associated with optimal clinical results, especially concerning to the effects of joint maximal range of motion (ROM). This study aimed to (i) determine whether visual inspections can quantify the LTR intensity during the DNT through a qualitative ultrasonography scale of LTR intensity (US-LTR scale), and (ii) assess the differences of US-LTR scale associated with changes in the maximal joint ROM. METHODS: Using a cross-sectional design, seven asymptomatic individuals were treated with DNT in the latent myofascial trigger point in both medial gastrocnemius muscles. During DNT, three consecutive LTRs were collected. The US-LTR scale was used to classify the LTRs into strong, medium, and weak intensities. The categories of US-LTR were differentiated by the velocity of LTRs using the optical flow algorithm. ROM changes in ankle dorsiflexion and knee extension were assessed before and immediately after DNT. RESULTS: The US-LTR scale showed the third LTR was significantly smaller than the first one (p < 0.05). A significant difference in velocity was observed between US-LTR categories (p < 0.001). A significant difference in the ROM was observed between the strong and weak-medium intensity (p < 0.05). CONCLUSIONS: The present findings suggest that the LTR intensity can be assessed using a qualitative US-LTR scale, and the effects of DNT on joint maximal ROM is maximized with higher LTR intensity. This study reports a novel qualitative method for LTR analysis with potential applications in research and clinical settings. However, further research is needed to achieve a broader application.

This study aimed to propose a conditioning activity (CA) model to stimulate improvement on neuromuscular responses, mechanical parameters and for the 50-m freestyle swimming. Thirteen male swimmers (19 ± 3 years and performances of 77% in relation to World Championship records) performed four CA protocols followed by a maximum performance in the 50-m freestyle. In the first protocol (P1) swimmers performed a standard warm-up (∼15 min); in the second protocol (P2) lunges (3 × 85% of the one-repetition maximum); in the third (P3) pull-ups (3 maximum repetitions) and box jumps 40 cm high and 60 cm deep (1 × 5 with 10% of the corporal weight); and in the fourth protocol (P4) a combination of exercises from the second and third protocols. CA protocols had no effect on the standard warm-up. However, P2 performance (27.01 ± 1.25 s) was similar to P1 (27.01 ± 1.18 s) and presented higher positive effects in mechanical parameters for the swim start performance in comparison to other protocols, contributing to improvements in the 50-m freestyle. In addition, turnaround time also had a negative effect, mainly in P3 (3.12 ± 0.28 s), signaling the improvement of this variable in all protocols (P1: 3.30 ± 0.38 s; P2: 3.17 ± 0.30 s; P4: 3.17 ± 0.34 s). P2 (after: 80 ± 11%; before: 82.7 ± 9.9%) and P3 (after: 82.7 ± 9.9%; before: 85.1 ± 9.7%) presented a possible positive effect on the percentage of voluntary activation in relation to P1 (after: 79.3 ± 10.7%; before: 76.3 ± 12%). In conclusion, the proposed conditioning activity protocols were not efficient for performance improvement in the 50-m freestyle compared to the standard model and seem to specifically influence each phase of the event.

Neuromuscular fatigue evaluation is widely performed on different muscles through the conventional protocol using maximum voluntary contraction (MVC) with electrical stimuli in the analyzed muscle. In an attempt to use this protocol on elbow extensor musculature, previous studies and pilot studies showed co-contraction effects from antagonist musculature during muscular stimulations. The aim of this study was to propose a new neuromuscular fatigue protocol evaluation on elbow extensor musculature. Twenty participants preformed exercises to induce central (CenFat) and peripheral fatigue (PerFat). Neuromuscular fatigue was evaluated on knee extensor muscles by a conventional protocol that provides Twitch Superimposed (TSK) and Twitch Potentiated (TPK), central and peripheral parameters respectively. For elbow extensor muscles, the protocol used sustained submaximal contraction at 10, 20, 30, 40, and 50% of MVC. The neuromuscular fatigue in upper limbs was identified by Twitch Potentiated (TPE) and multiple Twitch Superimposed (TSE) parameters. Using the relationship between MVC (%) and evoked force, the proposed protocol used several TSE to provide slope, y-intercept and R 2. It is proposed that slope, R 2, and y-intercept change may indicate peripheral fatigue and the identified relationship between y-intercept and R 2 may indicate central fatigue or both peripheral and central fatigue. The results were compared using the non-parametric analyzes of Friedmann and Wilcoxon and their possible correlations were verified by the Spearmann test (significance level set at p < 0.05). After PerFat a decrease in TPE (57.1%, p < 0.001) was found but not in any TSE, indicating only peripheral fatigue in upper limbs. After CenFat a decrease in TPE (21.4%, p: 0.008) and TPK (20.9%, p < 0.001) were found but not in TSK, indicating peripheral fatigue in upper and lower limbs but not central fatigue. A non-significant increase of 15.3% after CenFat and a statistical reduction (80.1%, p: 0.001) after PerFat were found by slope. Despite R 2 showing differences after both exercises (p < 0.05), it showed a recovery behavior after CenFat (p: 0.016). Although PerFat provided only peripheral fatigue, CenFat did not provide central fatigue. Considering the procedural limitations of CenFat, parameters resulting from the proposed protocol are sensitive to neuromuscular alteration, however, further studies are required.

The transient increase in torque of an electrically evoked twitch following a voluntary contraction is called postactivation potentiation (PAP). Phosphorylation of myosin regulatory light chains is the most accepted mechanism explaining the enhanced electrically evoked twitch torque. While many authors attribute voluntary postactivation performance enhancement (PAPE) to the positive effects of PAP, few actually confirmed that contraction was indeed potentiated using electrical stimulation (twitch response) at the time that PAPE was measured. Thus, this review aims to investigate if increases in voluntary performance after a conditioning contraction (CC) are related to the PAP phenomenon. For this, studies that confirmed the presence of PAP through an evoked response after a voluntary CC and concurrently evaluated PAPE were reviewed. Some studies reported increases in PAPE when PAP reaches extremely high values. However, PAPE has also been reported when PAP was not present, and unchanged/diminished performance has been identified when PAP was present. This range of observations demonstrates that mechanisms of PAPE are different from mechanisms of PAP. These mechanisms of PAPE still need to be understood and those studying PAPE should not assume that regulatory light chain phosphorylation is the mechanism for such enhanced voluntary performance. Novelty The occurrence of PAP does not necessarily mean that the voluntary performance will be improved. Improvement in voluntary performance is sometimes observed when the PAP level reaches extremely high values. Other mechanisms may be more relevant than that for PAP in the manifestation of acute increases in performance following a conditioning contraction.
