This study evaluated the effects of chromium picolinate (CrPic) with or without vitamin C on the reproductive potentials of cocks raised under high ambient temperature. Four diets contained CrPic at 0.00 to 1.20 mg kg-1 without vitamin C and another four diets contained CrPic with 200 mg vitamin C each. A total of 192 White Leghorn and Noiler cocks (96 cocks each) were randomly assigned to the 8 treatments and replicated 4 times with 6 cocks per replicate in a 2 x2 x 4 factorial experiment. At the end of the sixteen weeks study, twelve birds per treatment were randomly sacrificed. The testes were carefully sampled, weighed and processed for estimation of daily sperm production using the homogenate method. The results revealed that the interactions of 0.40 mg CrPic kg-1 with or without vitamin C significantly (p < 0.05) influenced the gonadal and extra-gonadal weights, semen characteristics among the cocks while the daily sperm production and reproductive hormones were not negatively affected. This study, therefore, concluded that 0.40 mg CrPic kg-1 diet with vitamin C could significantly reduce the effects of heat stress on cocks raised under high ambient temperature without negatively impacting on their normal reproductive functions.(AU)

In this work, a principle of flow injection analysis (FIA) was applied for sample introduction to an electrospray ionization - ion trap mass spectrometer (ESI-ITMS) with the aim to quantify chromium(III) picolinate (CrPic3) in commercial supplements by multiple reaction monitoring, and using cobalt(II) picolinate as internal standard (IS). FIA system was operated with ammonium formate 10 mmol L-1 in methanol-water (1:1, v/v) as a carrier solution at a flow rate 200 µL min-1; 100 µL injections were performed in 2-min intervals. Setting ion transitions m/z 419 â†’ 270 and 304 â†’ 260 for the analyte and IS, respectively, and 100 ms integration time, the method detection and quantification limits 12 ng g-1 and 40 ng g-1 of Cr (as CrPic3) in the air-dried powder. Acetonitrile extracts of the real-world samples presented varying from sample-to-sample chemical composition and IS efficiently compensated for ionization interferences. Mean results from triplicate analysis of four different supplements were obtained with relative standard deviation 0.1-4.0%, indicating acceptable precision. Trueness of the proposed FIA-ESI-ITMS/MS procedure was demonstrated by 95.8-108% percentage recoveries attained in the analysis of the CrPic3-spiked samples. For comparative purposes, total Cr was determined by ICP-MS. The quantitative results obtained indicate the necessity of analytical control of Cr(III) supplements commercially available and demonstrate that the proposed FIA-ESI-ITMS/MS procedure is well-suited for the determination of CrPic3 in such products.

We synthesized ten enamine naphthoquinones with yields ranging from 43 to 76%. These compounds were screened for their in vitro antiproliferative activities by MTT assay against four types of human cancer cell lines: HCT116, PC3, HL60 and SNB19. The naphthoquinones bearing the picolylamine (7) and quinoline (12) moieties were the most actives (IC50 < 24 µM for all the cell lines), which were comparable or better to the values obtained for the control drugs. In silico evaluations allowed us to develop a qualitative Structure-Activity Relationship which suggest that electrostatic features, particularly the C2-C3 internuclear repulsion and the molecular dipole moment, relate to the biological response. Furthermore, Molecular Docking simulations indicate that the synthetic compounds have the potential to act as anticancer molecules by inhibiting topoisomerase-II and thymidylate synthase.

An improved methodology of achieving an accurate separation of pure fractions of betacyanins and betaxanthins from Ullucus tuberosus Caldas has been carried out. For that purpose, an in-depth chemical identification of each betalain fraction using HPLC-DAD-ESI-MS was developed. This procedure allowed to evaluate the fractionation efficiency and also identify a large number of betalains, most of which have not been described so far in this raw material: betanidin- and isobetanidin-5-O-(4'-O-malonyl-ß-glucoside), 2-decarboxy-phyllocactin, betanidin- and isobetanidin-6-O-(6'-O-feruloyl)-ß-glucoside (gomphrenin and isogomphrenin III), dehydro-phyllocactin and isophyllocactin, and arginine and glycine-betaxanthins (portulacaxanthin III). Moreover, the availability of pure betalain fractions by the proposed methodology permitted to establish the total betalain content and the antioxidant activity of both separated betacyanin and betaxanthin fractions of ulluco for the first time. The results suggest Ullucus tuberosus as an underutilized food bioactive source with a high concentration of total betaxanthins (21.8 µg indicaxanthin/g fresh ulluco) and betacyanins (44.5 µg betanin/g fresh ulluco), strongly correlated to the Folin-Ciocalteau reduction capacity.

Chromium (Cr) is a micromineral that is involved in the metabolism of carbohydrates, lipids, ammonia, and nucleic acids; thus, its supplementation can influence the nutritional status of ruminants, and consequently, colostrum profile, since this secretion depends on products secreted by the mammary gland and elements of the maternal bloodstream. The present study investigated the influence of supplementation with Cr bound to organic molecule on the nutritional, immune, and antioxidant quality of ewe colostrum. Thirty-two multiparous Santa Ines ewes (55.3 ± 8.00 kg body weight) were randomly assigned into four groups: T1 (0.0 mg of chromium picolinate (CrPic) supplementation per ewe, n = 8), T2 (0.15 mg of CrPic per ewe, n = 9), T3 (0.30 mg of CrPic per ewe, n = 7), and T4 (0.45 mg of CrPic per ewe, n = 8). Supplementation was supplied during the breeding season, pregnancy, and lactation. Shortly after calving, the first milking colostrum was collected to determine its chemical composition, activity of lysozyme, lactoperoxidase, ceruloplasmin, catalase, glutathione peroxidase, and oxygen radical absorbance capacity. The results show that lactoperoxidase activity decreased with CrPic supplementation (P < 0.01), revealing that this micromineral reduces an important component of defense mechanism in the body. Therefore, the results of this work show that supplementation with chromium picolinate influences colostrum quality.

Polymyxins (colistin and polymyxin B) have recently regained significant importance as last-line drugs to treat infectious diseases due to multidrug-resistant gram-negative bacteria. However, resistance to polymyxins has increased, and the recognition of plasmid-mediated resistance (by the mcr gene) has led to an epidemiological concern. We aimed to evaluate the reduction of the polymyxin B minimum inhibitory concentration (MIC) in the presence of EDTA or dipicolinic acid (DPA) by using the broth microdilution (BMD) method for phenotypic screening of acquired polymyxin resistance mediated by the mcr-1 gene. Overall, 94 Enterobacterales (48 polymyxin-resistant and 46 polymyxin-susceptible) were evaluated: 47 mcr-1 positive (36 Escherichia coli, 2 Klebsiella pneumoniae, and 9 Salmonella spp.) and 47 mcr-1 negative (3 E. coli and 44 K. pneumoniae-27 isolates with MIC from ≤0.125 to 8 µg/mL and 20 isolates with MIC from 16 to 64 µg/mL). Results were categorized as positive when the chelator decreased the original BMD MIC by ≥2 logs. The majority (95.7%) of mcr-1 positive isolates displayed at least a 3 log dilution decrease in the MIC of polymyxin B with EDTA or DPA. The EDTA-based BMD assay detected 45 mcr-1-positive isolates, with only one false-positive among the mcr-1-negative isolates (sensitivity [SN], 95.7%; specificity [SP], 97.9%), whereas the DPA-based BMD assay detected 44 mcr-1-positive isolates (SN, 93.6%; SP, 95.7%), with two false-positive results. The accuracy of EDTA- and DPA-based BMD assays were 97% and 95%, respectively. The EDTA- and DPA-based assays were demonstrated to be reliable methods to detect mcr-1 positive isolates with excellent accuracy.

Fungal pathogens produce toxins that are important for their pathogenesis and/or aggressiveness towards their hosts. Picolinic acid (PA), a non-host selective toxin, causes lesions on rice leaves resembling those originated from Pyricularia oryzae infection. Considering that non-host selective toxins can be useful for plant diseases control, this study investigated whether the foliar spray with PA on wheat (Triticum aestivum L.) plants, in a non-phytotoxic concentration, could increase their resistance to blast, stimulate the anti-oxidative metabolism, and minimize alterations in photosynthesis. The PA spray at concentrations greater than 0.1 mg ml-1 caused foliar lesions, compromised the photosynthesis and was linked with greater accumulation of hydrogen peroxide (H2 O2 ) and superoxide anion radical (O2 •- ). Fungal mycelial growth, conidia production and germination decreased by PA at 0.3 mg ml-1 . Blast severity was significantly reduced by 59 and 23%, respectively, at 72 and 96 h after inoculation for plants sprayed with PA (0.1 mg ml-1 ) at 24 h before fungal inoculation compared to non-sprayed plants. Reduction on blast symptoms was linked with increases on ascorbate peroxidase (EC 1.11.1.11), catalase (EC 1.11.1.6), glutathione peroxidase (EC 1.11.1.9), glutathione reductase (EC 1.8.1.7), glutathione-S-transferase (EC 2.5.1.18), peroxidase (EC 1.11.1.7), and superoxide dismutase (EC 1.15.1.1) activities, lower H2 O2 and O2 •- accumulation, reduced malondialdehyde production as well as less impairments to the photosynthetic apparatus. A more efficient antioxidative metabolism that rapidly scavenges the reactive oxygen species generated during P. oryzae infection, without dramatically decreasing the photosynthetic performance, was a remarkable effect obtained with PA spray.

Multiple sclerosis (MS) is one of the most common causes of progressive disability affecting young people with very few therapeutic options available for its progressive forms. Its pathophysiology involves demyelination and neurodegeneration apparently driven by microglial activation, which is physiologically dependent on colony-stimulating factor-1 receptor (CSF-1R) signaling. In the present work, we used microglial modulation through oral administration of brain-penetrant CSF-1R inhibitor BLZ945 in acute and chronic cuprizone (CPZ)-induced demyelination to evaluate preventive and therapeutic effects on de/remyelination and neurodegeneration. Our results show that BLZ945 induced a significant reduction in the number of microglia. Preventive BLZ945 treatment attenuated demyelination in the acute CPZ model, mainly in cortex and external capsule. In contrast, BLZ945 treatment in the acute CPZ model failed to protect myelin or foster remyelination in myelin-rich areas, which may respond to a loss in microglial phagocytic capacity and the consequent impairment in oligodendroglial differentiation. Preventive and therapeutic BLZ945 treatment promoted remyelination and neuroprotection in the chronic model. These results could be potentially transferred to the treatment of progressive forms of MS.

OBJECTIVE: The purpose of this study was to investigate the effects of chromium picolinate (CrPic) supplementation associated with aerobic exercise using measures of oxidative stress in rats exposed to air pollution. METHODS: Sixty-one male Wistar rats were divided into eight groups: residual oil fly ash (ROFA) exposure and sedentary (ROFA-SED); ROFA exposure, sedentary and supplemented (ROFA-SED-CrPic); ROFA exposure and trained (ROFA-AT); ROFA exposure, supplemented and trained (ROFA-AT-CrPic); sedentary (Sal-SED); sedentary and supplemented (Sal-SED-CrPic); trained (Sal-AT); and supplemented and trained (Sal-AT-CrPic). Rats exposed to ROFA (air pollution) received 50 µg of ROFA daily via intranasal instillation. Supplemented rats received CrPic (1 mg/kg/day) daily by oral gavage. Exercise training was performed on a rat treadmill (5×/week). Oxidative parameters were evaluated at the end of protocols. RESULTS: Trained groups demonstrated lower gain of body mass (P < .001) and increased exercise tolerance (P < .0001). In the gastrocnemius, trained groups demonstrated increased SOD activity (P < .0001) and decrease levels of TBARS (P = .0014), although CAT activity did not differ among groups (P = .4487). CONCLUSION: Air pollution exposure did not lead to alterations in oxidative markers in lungs and heart, and exercise training was responsible for decreasing oxidative stress of the gastrocnemius.

SUMMARY: The experiment was conducted to evaluate the effects of dietary supplemental chromium (Cr) on growth performance, meat quality, intestinal morphology, mucosa Hsp70 mRNA expression and antioxidant status of ducks reared under heat stress conditions. All ducks were randomly divided into three treatment groups, respectively, control group (Control, 23 ± 2 °C), heat stress group (HS, 32 ±2 °C), Cr picolinate group (CrPic, 32 ± 2 °C, 0.2 mg Cr/kg). Feed and distilled-deionized water were available ad libitum for an experimental phase of 35 days. Samples were collected on the day 14, 21 and 35 to determine biological and hematological values. Results showed that heat stress or dietary supplemental Cr both didn't have distinct influence on growth performance (P>0.05), compared to controls. Ducks fed 0.2 mg Cr/kg diet had greater ultimate pH (pHu)(P<0.05) than HS group. At day 14, the ratio of villus height to crypt depth (V/C) in CrPic group significantly increased (P<0.05) than that of HS group in jejunum. Heat stress remarkably increased Hsp70 mRNA expression in jejunum compared with controls (P<0.05). While the expression of Hsp70 mRNA in CrPic group was significantly decreased compared with HS (P<0.05). At day 21, the V/C of ileum in CrPic group significantly increased compared with HS group (P<0.05). Serum SOD levels in CrPic group were significantly higher than those in HS group (P<0.05). At day 35, Hsp70 mRNA expression and serum T-SOD levels in CrPic group significantly increased compared with controls (P<0.05). T-AOC in HS group significantly decreased compared with controls (P<0.05). Results indicate that dietary Cr supplementation doesn't influence ducks' growth performance, but has a positive effect on meat quality, small intestine morphology, also regulates Hsp70 mRNA expression under heat stress conditions, and enhances the antioxidant status.


RESUMEN: Se evaluó los efectos del cromo (Cr) dietético suplementario sobre el rendimiento del crecimiento, la calidad de la carne, la morfología intestinal, la expresión del ARNm Hsp70 en la mucosa y el estado antioxidante de los patos criados bajo condiciones de estrés por calor. Todos los patos se dividieron aleatoriamente en tres grupos: grupo control (control, 23 ± 2 °C), grupo de estrés térmico (HS, 32 ± 2 °C) y grupo de picolinato de Cr (CrPic, 32 ± 2 °C, 0,2 mg Cr / kg). El alimento y el agua desionizada destilada estuvieron disponibles ad libitum durante la fase experimental de 35 días. Las muestras se recogieron los días 14, 21 y 35 para determinar los valores biológicos y hematológicos. Los resultados mostraron que el estrés térmico o la suplementación dietética de Cr no tuvieron una influencia distinta en el rendimiento del crecimiento (P> 0,05), en comparación con los controles. Los patos alimentados con 0,2 mg de Cr / kg de dieta tuvieron un mayor pH final (pHu) (P <0,05) que el grupo HS. En el día 14, la relación de la altura de las vellosidades a la profundidad de la cripta (V / C) en el grupo CrPic aumentó significativamente (P <0,05) en relación a la del grupo de HS en el yeyuno. El estrés por calor incrementó notablemente la expresión del ARNm de Hsp70 en el yeyuno en comparación con los controles (P <0,05). Mientras que la expresión del ARNm de Hsp70 en el grupo CrPic se redujo significativamente en comparación con HS (P <0,05). En el día 21, la relación V / C del íleon en el grupo CrPic aumentó significativamente en comparación con el grupo HS (p <0,05).
