RESUMO

The study characterized the lactoferrin (Lf) mRNA gene in different goat breeds in the Philippines and determined its association with subclinical mastitis (SCM). The study involved collection of milk at second week of lactation (n=75) and blood samples (n=5) to obtain extracted RNA and using cDNA to amplify Lf gene through polymerase chain reaction. The nucleotide and amino acid sequences were determined and used as reference in the evaluation of phylogenetic relationship. Amplified products were utilized for RFLP analysis before determining the association of the gene with SCM. Results of the study demonstrated that Lf gene in goats registered a molecular weight of 2135. Nucleotide and amino acid sequence of Lf gene revealed high similarity (99%) in Saanen, Anglo-Nubian and Philippine native goats with that of Capra hircus (U53857) Lf gene submitted to GenBank. Phylogenetic studies showed that Lf gene of Anglo-Nubian, Saanen and Native goats clade together with Lf gene of C. hircus (U53857). Three genotypes in goats were documented using the restriction enzymes AluI and HaeIII. Based on the Statistical analysis, association (comp 5.65, p = 0.0308) has been established between the Lf genes of goats with genotype BB to SCM using HaeIII restriction enzyme.(AU)

Adding ionophores to ruminant diets is a strategy to manipulate ruminal fermentation and improve milk yield. This study evaluates the effects of narasin supply to lactating ewes on dry matter intake (DMI), milk yield and composition, and performance of the lambs. Thirty lactating Santa Inês and Santa Inês × Dorper ewes fed a basal diet containing 50 % coastcross hay and 50 % concentrate were randomly assigned to two treatments: control (CON; without ionophores) or NAR (addition of 13 mg narasin kg-1 DM). From the 2nd to 10th week of lactation, DMI of ewes was determined, and once a week, their milk production and composition was measured over a 3-h interval. At the 10th week of lactation, lambs were weaned and their average daily gain (ADG) and starter DMI continued to be evaluated for two more weeks. Narasin supply did not affect weight and DMI of ewes. Ewes fed NAR had greater feed efficiency for milk production and displayed tendency for higher milk yield. Narasin supply reduced milk protein levels, but it did not affect other milk component levels. Ewes fed NAR had greater production of milk urea nitrogen and showed tendency for higher production of fat and total solids. Starter DMI of lambs was not affected by treatments; however, there was a tendency for greater weaning weight for NAR lambs. At the end of experiment, no differences were observed in the performance of lambs. The supply of 13 mg narasin kg-1 to lactating ewes improved milk yield efficiency and tended to increase the weaning weight of their lambs.(AU)

Dopamine from tuberoinfundibular dopaminergic (TIDA) neurones tonically inhibits prolactin (PRL) secretion. Lactational hyperprolactinaemia is associated with a reduced activity of TIDA neurones. However, it remains controversial whether the suckling-induced PRL surge is driven by an additional decrease in dopamine release or by stimulation from a PRL-releasing factor. In the present study, we further investigated the role of dopamine in the PRL response to suckling. Non-lactating (N-Lac), lactating 4 hour apart from pups (Lac), Lac with pups return and suckling (Lac+S), and post-lactating (P-Lac) rats were evaluated. PRL levels were elevated in Lac rats and increased linearly within 30 minutes of suckling in Lac+S rats. During the rise in PRL levels, dopamine and 3,4-dihydroxyphenylacetic acid (DOPAC) levels in the median eminence (ME) and neurointermediate lobe of the pituitary did not differ between Lac+S and Lac rats. However, dopamine and DOPAC were equally decreased in Lac and Lac+S compared to N-Lac and P-Lac rats. Suckling, in turn, reduced phosphorylation of tyrosine hydroxylase in the ME of Lac+S. Domperidone and bromocriptine were used to block and activate pituitary dopamine D2 receptors, respectively. Domperidone increased PRL secretion in both N-Lac and Lac rats, and suckling elicited a robust surge of PRL over the high basal levels in domperidone-treated Lac+S rats. Conversely, bromocriptine blocked the PRL response to suckling. The findings obtained in the present study provide evidence that dopamine synthesis and release are tonically reduced during lactation, whereas dopamine is still functional with respect to inhibiting PRL secretion. However, there appears to be no further reduction in dopamine release associated with the suckling-induced rise in PRL. Instead, the lower dopaminergic tone during lactation appears to be required to sensitise the pituitary to a suckling-induced PRL-releasing factor.

The objective of this trial was to evaluate the behavioral patterns and performance of lactating sows and their litters under the effect of artificial vocalization. Twenty-eight sows and their litters were distributed in a completely randomized design in a 2x2 factorial scheme (artificial vocalization x lactation week). The behavior of the animals was monitored during 24 hours on the 7th and 15th days of lactation, analyzing the number, interval, and frequency of nursings. The body condition and performance of the sows were also evaluated. Artificial vocalization promoted higher frequencies of eating for sow and nursing for piglets (P <0.05), increased inactive sow behavior (P <0.05), and reduced sow alert in activity (P <0.05). The number and duration of suckling sessions at the 15thday of lactation were reduced (P <0.05). The use of artificial vocalization did not affect the body condition or milk production of the lactating sows, or the performance of the litter during lactation (P> 0.05). The use of maternal artificial vocalization during lactation of sows promoted greater lactation efficiency and longer rest time, favoring the sows' welfare.

We aimed to investigate the effect of maternal exposure to NaF on mandibular bone microarchitecture and phosphocalcic plasma parameters of the offspring. For this purpose, 10-, 15-, and 21-day-old pups (n = 6-8 per group) from two groups of mothers, control and NaF 50mg/L treated dams, were used. Plasma calcium (Ca) and phosphorus (P) levels and alkaline phosphatase activity (ALP) were measured. Fluoride concentration (F-) in bone and in stomach content was measured using potentiometry after isothermal distillation. Morphometric, histological, and histomorphometric analyses of the jaw bones were performed. Plasma Ca and P levels and ALP activity increased in 10-day and decreased in 21-day-old pups from NaF-treated mothers. Fluoride concentration in stomach content samples of 15- and 21-day-old nursing pups from mothers exposed to NaF in their drinking water was higher compared to that observed in control dam offspring. Mandibular F- content was higher in 21-day-old pups born to F--exposed dams compared to those observed in age-matched control pups. Mandibular area increased in 21-day-old pups born to treated mothers as compared to controls. Mandibular bone volume BV/TV (%) was higher in offspring from NaF-exposed dams than in controls at all the studied times. The increase in bone volume after exposure to F- was concomitant with the increase in trabecular thickness and the decrease in trabecular separation. Altogether, our results showed that exposure to NaF during gestation and lactation increased mandibular area and bone volume of pups, with concomitant changes in phosphocalcic parameters associated with the bone modeling process.

The objective of the present study was to evaluate the effect of different schemes of calves' supplementation in a creep feeding system, on the behavior of Bos indicus calves and dams, and also the influence of the calves' supplementation on dams' performance. Forty-eight Nellore male calves (147 ± 7 kg body weight and 3 months of age) in the suckling phase and their dams (476 ± 9 kg and 6 years of age) were studied in a completely randomized design. The experiment was divided into two periods of 71 days. The treatments were 5- and 10-g supplement dry matter (DM)/kg BW day offered in periods 1 and 2, respectively (5S/10S); 10- and 5-g supplement DM/kg BW day offered in periods 1 and 2, respectively (10S/5S); 7.5-g supplement DM/kg BW day in both periods 1 and 2 (7.5S); and mineral mix ad libitum in both periods 1 and 2 (MM). No differences (P < 0.05) in body condition score (BCS), final body weight (FBW), and average daily gain (ADG) were found in dams' performance. Calves from MM treatment spent more time (P < 0.05) grazing than the supplemented calves from 5S/10S and 10S/5S treatments, in the first period. No difference in suckling time was found between the treatments (P > 0.05) in the first evaluated period. Calves from 10S/5S treatment spent more time suckling and less time eating supplements (P < 0.05) than 5S/10S treatment animals, in the second evaluated period. Dams of MM treatment's calves had more idle time and lower grazing time when compared with the mothers of calves from 5S/10S and 10S/5S treatments. It was concluded that different schedules of Nellore calves' supplementation on pasture do not affect their mothers' performance, and supplementation decreases the grazing time of calves in the suckling phase.

Despite the increasing use of oregano (Origanum vulgare L.) essential oil for therapeutic purposes, pre- and postnatal development of animals offspring exposed to this oil has not yet been evaluated. In line with previous concerns of genotoxicity, in this study adult rats were exposed to different doses of oregano essential oil (3, 9 and 27% vol/vol) during pre-mating, mating, gestation and lactation. Prenatal screening included fetal development and uterine inspection, where the reproductive rate of females such as breeding, pregnancy, delivery, viability and post-implantation loss rate were measured. Postnatal evaluation of rat offspring included motor development, neuroendocrine and behavioral assessment. Body weight of rat dams and signs of dystocia were evaluated daily. Development of physic characteristics and reflex tests of puppies were also assessed. Additionally, these rats, when adults, were submitted to sexual and open field behavioral tests. The main differences among the groups were observed in the indices of mating, pregnancy and post-implantation loss (P < 0.01). Results demonstrated that the treatment of parental generation with oregano essential oil has the potential to affect the developing fetuses at the highest dose used, but without causing maternal toxicity and changes in general behavior and development of the progeny.


Apesar do aumento do uso do óleo essencial de orégano (Origanum vulgare L.) para fins terapêuticos, o desenvolvimento pré e pós-natal da progênie de animais expostos a este óleo ainda não foi avaliado. Partindo das suspeitas prévias de genotoxicidade desse óleo, neste estudo, ratos Wistar adultos foram expostos a diferentes doses do óleo essencial de orégano (3, 9 e 27% vol/vol) durante o acasalamento, a gestação e a lactação. Para a avaliação pré-natal, o desenvolvimento gestacional foi observado e os úteros inspecionados, assim como os índices reprodutivos das fêmeas, como a taxa de acasalamento, de gestação, parto e perdas pós-implantação. Na avaliação pós-natal, observou-se o desenvolvimento motor, neuroendócrino e comportamental da prole. Observou-se, diariamente, o peso das ratas e sinais de distocia. Após o parto, as características de desenvolvimento e testes de reflexos dos filhotes foram avaliadas, enquanto que, na puberdade, foram realizadas análises histopatológicas e dosagem hormonal. Adicionalmente, na idade adulta, esses ratos foram submetidos ao teste de comportamento em campo aberto e ao comportamento sexual. As principais diferenças entre os grupos foram nos índices de acasalamento, de gestação e de perdas pós-implantação (P < 0,01). Os resultados demonstram que o tratamento da geração parental com óleo essencial de orégano tem potencial para afetar os índices reprodutivos das ratas e o desenvolvimento dos fetos na maior dose utilizada, mas sem causar toxicidade materna e alterações no desenvolvimento geral e comportamental da sua progênie.

The aim of this study was to evaluate the effects of mammary biopsy procedures on gland function of goats submitted to hormonal lactation. Ten female Caninde goats, at the age of around 3 years, were used and lactation was induced by using estrogen, progesterone, and prednisolone. Ultrasonographic examinations of mammary gland, milking, and analysis of physicochemical properties of milk were performed immediately before (0 h), and 24, 48, and 72 h after each biopsy procedure. Surgical mammary gland biopsies were obtained at days 5 and 26 of the lactation. Even after drying initiation, all animals produced milk for five months. No abnormalities were observed on the ultrasonographic appearance of biopsied glands. All 20 biopsies were technically successful and any inflammation or infection was associated with biopsy procedures. Regarding the physicochemical properties and volumes of milk produced, no significant differences (P > 0.05) were observed between right and left glands nor in the comparison of the results from time 0 with 24, 48, and 72 h for biopsied teats. The mammary biopsy did not induce neither severe changes in production and physicochemical properties of the milk nor in ultrasonographic appearance of mammary gland of goats in hormonal lactation.


O objetivo deste estudo foi avaliar os efeitos dos procedimentos de biópsia mamária sobre a função da glândula de caprinos submetidos à lactação induzida. Dez cabras Canindé com idade de cerca de 3 anos foram utilizadas e a lactação foi induzida pelo uso de estrógeno, progesterona e prednisolona. Exames ultrassonográficos da glândula mamária, ordenha e análise das propriedades físico-químicas do leite foram realizados imediatamente antes (0 h), e 24, 48 e 72 h após cada procedimento de biópsia. As biópsias cirúrgicas de glândula mamária foram obtidas aos dias 5 e 26 da lactação. Mesmo após o início do processo de secagem, todos os animais continuaram produzindo leite por cinco meses. Nenhuma anormalidade foi observada na aparência ultrassonográfica das glândulas biopsiadas. Todas as 20 biópsias foram realizadas com sucesso e nenhuma inflamação ou infecção foi observada. Com relação às propriedades físico-químicas e volumes de leite produzido, nenhuma diferença significativa (P > 0,05) foi observada entre a glândula direita e esquerda, nem comparando resultados do momento 0 com 24, 48 e 72 h para as glândulas biopsiadas. A biópsia mamária não induz severas mudanças na produção e nas propriedades físico-químicas do leite nem na aparência ultrassonográfica da glândula mamária de caprinos em lactação.
