RESUMO

The present study evaluated the use of the probiotic Lacticaseibacillus paracasei DTA-83 as a nitrite-reducing agent to produce potentially probiotic or postbiotic pre-converted nitrite from celery. The results obtained were compared to those achieved by direct addition of sodium nitrite for the typical reddish color formation in cooked pork sausages and the inhibitory potential against the growth of target microorganisms, including the clostridia group. Regarding the sausages color, similar findings were observed when comparing the use of pre-converted nitrite from celery produced by L. paracasei DTA-83 and the direct addition of sodium nitrite. Additionally, it presented an inhibitory effect against Salmonella spp., which was not observed with the direct addition of nitrite, revealing a potential strategy to control salmonellosis in the matrix. However, a non-equivalent preservative effect against Clostridium perfringens (INCQS 215) was determined. The results highlight a promising alternative to produce probiotic or postbiotic meat ingredients; however, further studies should be conducted to investigate doses that achieve microbial control.

Objetivo. Caracterizar flavonoides presentes en el extracto y subextracto metanólico de las hojas de Apium graveolens var. Rapaceum, DC. "Apio-nabo". Materiales y métodos. Se elaboró un extracto y subextracto metanólico de las hojas Apium graveolens var. Rapaceum, DC. "Apio-nabo". Se determinó la solubilidad del extracto metanólico en solventes de polaridad creciente. Se detectaron los componentes químicos del extracto y subextracto metanólico mediante un tamizaje fitoquímico empleando gelatina, tricloruro férrico, reactivo de Shinoda, reactivo de Dragendorff y reactivo de Ninhidrina, entre otros. Se ejecutó cromatografía en capa fina y mediante espectroscopia UV/Vis se propuso estructuras químicas para los metabolitos tipo flavonoides presentes en el extracto y subextracto metanólico de hojas de Apium graveolens var. Rapaceum, DC. "Apio-nabo". Resultados. El extracto metanólico de hojas de Apium graveolens var. Rapaceum, DC fue soluble en solventes polares. Los metabolitos secundarios encontrados son compuestos fenólicos tipo flavonoides, taninos y alcaloides en el extracto y subextracto metanólico. Se propuso tres estructuras químicas de flavonoides a través del análisis de los espectros UV/Vis, y mediante comparación con lo publicado por TJ Mabry y Olga Lock. Conclusión. Se caracterizó la posible estructura química de tres metabolitos secundarios tipo flavona encontrados en el subextracto metanólico de hojas de Apium graveolens var. Rapaceum, DC.


Objective. To characterize flavonoids present in the methanol extract and subextract of the leaves of Apium graveolens var. Rapaceum, DC. "Celery-turnip". Materials and methods. An methanolic extract and subextract of Apium graveolens var. Rapaceum, DC. "Celery-turnip" was prepared. The solubility of the methanolic extract in solvents of increasing polarity was determined. The chemical components of the methanolic extract and subextract were detected by means of a phytochemical screening using gelatin, ferric trichloride, Shinoda reagent, Dragendorff reagent and Ninhydrin reagent, among others. Thin layer chromatography was carried out and by means of UV / Vis spectroscopy chemical structures were proposed for the flavonoid metabolites present in the methanolic extract and subextract of Apium graveolens var. Rapaceum, DC. "Celery-turnip". Results. It was determined that the methanolic extract of the leaves of Apium graveolens var. Rapaceum, DC. is soluble in polar solvents. The metabolites, the phenolic compounds, the flavonoids, the tannins and the alkaloids in the extract and methanol subextract. Flavonoid chemical structures are proposed through the analysis of the UV / Vis spectra, and it has been published with TJ Mabry and Olga Lock. Conclusion. The possible chemical structure of three secondary flavone metabolites found in methanolic subextract of leaves of Apium graveolens var. Rapaceum, DC.

The objective of the present study was to investigate the effects of 3-n-butylphthalide (NBP) on a 1-methyl-4-phenylpyridinium (MPP+)-induced cellular model of Parkinson’s disease (PD) and to illustrate the potential mechanism of autophagy in this process. For this purpose, rat PC12 pheochromocytoma cells were treated with MPP+ (1 mM) for 24 h following pretreatment with NBP (0.1 mM). Cell metabolic viability was determined by the MTT assay and cell ultrastructure was examined by transmission electron microscopy. The intracellular distribution and expression of α-synuclein and microtubule-associated protein light chain 3 (LC3) were detected by immunocytochemistry and Western blotting. Our results demonstrated that: 1) NBP prevented MPP+-induced cytotoxicity in PC12 cells by promoting metabolic viability. 2) NBP induced the accumulation of autophagosomes in MPP+-treated PC12 cells. 3) Further study of the molecular mechanism demonstrated that NBP enhanced the colocalization of α-synuclein and LC3 and up-regulated the protein level of LC3-II. These results demonstrate that NBP protects PC12 cells against MPP+-induced neurotoxicity by activating autophagy-mediated α-synuclein degradation, implying that it may be a potential effective therapeutic agent for the treatment of PD.

The aim of this study was to characterize the quality of celery petioles (Apium graveolens L. cv Golden Clause) from the external, middle, and internal zones of each plant. Harvest dates were 93 (HD1) and 124 (HD2) days after transplantation. Total weight (TW), total length (TL), total leaves number (LN), and petiole length of leaves (PL) for the 3 zones were measured. Physicochemical (color, b* and h; texture; total soluble solids, TSS; titratable acidity, TA; water content, WC), nutraceutical (ascorbic acid, AA; total quinones, TQ; browning potential, BP), and sensorial analysis (flavor, fibrosity, crunchiness) were done at harvest on petioles of each zone. No differences between harvesting dates were found in TW, TL, LN, and PL. Middle leaves had the highest PL. Harvest dates were not different in b*, h, TA, AA, and WC. Texture, TSS, BP, and TQ resulted higher in petioles of HD2 than in HD1. Similar TSS and TA were found in leaves of different plant zones. The yellow color of both external and internal zones was significantly higher than in the middle zone. The texture and BP was similar between the external and middle zones but the WC was lower in the external zone. Similarly, the AA content as well as the TQ was also lower than in the middle zone. Harvest delay improved the nutraceutical value in terms of BP and TQ, even though it also resulted in pithiness and fibrosity of the leaves. This study therefore suggests that the petioles of the middle zone when harvested at HD1 are the most suitable for consumption. Practical Application: Celery is a vegetable reduced in calories, has a high nutritional value and its fresh petioles are mainly consumed in salads. The texture and flavor are the most important attributes that define consumers' acceptability. As nutritional value, texture, and flavor may change with plant age and different zones of the plant, harvest date plays an important role on quality. Results indicate that harvest delay improved the nutraceutical value even though it also resulted in pithiness and fibrosity of the stalks. Petioles of the middle zone, when harvested at 93 d after transplanting, are the most suitable for consumers' consumption.

A simple and low cost method able to detect the presence of pesticides, organophosphates and carbamates based on the inhibition of serum and erythrocytic cholinesterases, was used in lettuce (Lactuca sativa), cilantro (Coriandum santivum) and celery (Apium graveolens) obtained from the Ferias del Agricultor from Valle Central of Costa Rica. The percentage inhibition of cholinesterases is related to the presence of plaguicide in the vegetable. Thirteen percent of the analyzed samples were positive for plaguicides using serum cholinesterase and 33% for erythrocytic cholinesterase. Washing and cooking the vegetables does not eliminate the presence of plaguicides but they lower slightly the concentration. Statistical evidence (p = 0.0001) indicates that erythrocytic cholinesterase has higher analytical sensitivity than serum cholinesterase. It is very important to establish the degree of contamination with pesticides in these agricultural products because they are exposed to direct contamination by fumigation, soil contamination and irrigation water, and are products that are often consumed without adequate cooking and washing.
