The work aims to analyze the associations of polymorphic variants of the PRL and BLG genes with resistance and susceptibility to mastitis in Holstein cows. The experimental study consisted of the selection of biomaterial samples from 250 heads of Holstein cows aged 3 years divided into two groups (healthy and with a confirmed diagnosis of mastitis). The determination of animal genotypes was carried out using polymerase chain reaction and restriction fragment length polymorphism. The study of the nature of the association of polymorphic variants of the PRL and BLG gene with resistance/increased risk of mastitis established a significant deviation from the theoretically expected distribution of bBLG-HaeIII genotypes in the group of animals suffering from mastitis (the value of χ2 was 0.24). The bBLG-HaeIIIBB genotype can act as a marker of an increased risk of developing mastitis in Holstein cows; its frequency in the group of sick animals exceeds the frequency in the control group by more than 2 times (44.0 compared to 17.0%, respectively). The bBLG-HaeIIIAB genotype is significantly associated with mastitis resistance in Holstein cows; its frequency is 2 times lower than in the control group (28.0 compared to 54.0%).

κ-Cn and ß-lactoglobulin are important candidate genes associated with milk yield and milk protein content. The present investigation is carried out to determine the polymorphisms status of κ-Cn and ß-lactoglobulin genes in Anatolian Black cattle and Holstein breeds. PCR-RFLP technique was used to determine Kappa-Casein and ß-lactoglobulin polymorphisms in both cattle breeds. The allele frequency of Anatolian Black cattle in terms of κ-Cn and ß-lactoglobulin genes were 0.50 (A) 0.50 (B) and 0.20 (A) 0.80 (B) respectively, whereas in Holstein were 0.29 (A) 0.71 (B) and 0.44 (A) 0.56 (B) respectively. The chi-square test showed that each cattle breed was in Hardy-Weinberg equilibrium (P>0.05).


κ-Cn e ß-lactoglobulina são importantes genes candidatos associados à produção de leite e ao teor de proteína do leite. A presente investigação é realizada para determinar o status de polimorfismos dos genes κ-Cn e ß-lactoglobulina nas raças Anatolian Black cattle e Holstein. A técnica PCR-RFLP foi utilizada para determinar os polimorfismos Kappa-Casein e ß-lactoglobulina em ambas as raças de gado. A freqüência dos alelos do gado negro anatólio em termos de κ-Cn e ß-lactoglobulina foi de 0,50 (A) 0,50 (B) e 0,20 (A) 0,80 (B) respectivamente, enquanto que em Holstein foi de 0,29 (A) 0,71 (B) e 0,44 (A) 0,56 (B) respectivamente. O teste do qui-quadrado mostrou que cada raça bovina estava em equilíbrio de Hardy-Weinberg (P>0,05).

O objetivo deste trabalho foi identificar polimorfismos genéticos de leptina, ß-lactoglobulina e fator de transcrição pituitária (PIT1) e avaliar seus efeitos na composição química e na contagem de células somáticas de leite de vacas leiteiras mestiças que vivem em um clima quente. Um total de 291 vacas leiteiras mestiças foram investigadas. Foram coletadas amostras de sangue para extração de DNA e amostras de leite. As amostras foram classificadas em três grupos genéticos: 12/ (42), 34/ (83) e 78/ (166) Holandês x Guzerá. As frequências de alelos e genótipos foram determinadas e o equilíbrio Hardy-Weinberg foi avaliado. Foram realizadas análises da composição do leite (gordura, proteína, lactose e extracto seco desengordurado), contagem de células somáticas e rendimento leiteiro. Os grupos genéticos e os polimorfismos genéticos para cada gene foram utilizados como efeitos fixos na análise. O único polimorfismo encontrado em equilíbrio de Hardy-Weinberg foi para o genótipo da ß-lactoglobulina. No presente estudo, era esperado que a maioria das variáveis de composição variasse entre os genótipos. Já se sabe que os cruzamentos dão origem a animais com características fenotípicas e genotípicas. No entanto, os polimorfismos não influenciaram a composição e a qualidade do leite nas vacas 12/ , 34/ e 78/ Holstein x Guzerá mantidas em um clima quente.(AU)

In dairy farm animals, one the most important goal of the selection is the improvement of milk yield and composition. Several studies have demonstrated that the candidate genes of the kappa-casein (CSN3) and β - lactoglobulin (β-LG) are associated with milk yield, milk quality and health traits in dairy animals. Therefore the aim of this study was to detect polymorphisms in CSN3 and β-LG genes and its association with milk yield in up to 305 days (305MY) and predicted transmission capacity (PTA) for 305MY in Girolando cattle. Totally, 138 bulls and 729 cows (n=867) were sampled. The genotypes of both genes were obtained by the PCR-RFLP method using HinfI and HaeIII enzymes for CSN3 and β-LG genes, respectively. Statistical results revealed two alleles A and B for both genes. The genotypes and alleles more frequents for CSN3 and β-LG genes were respectively: AA (0.7324) and A (0.8558), and AB (0.4827) and A (0.5017). The x2 test revealed that the two loci were at Hardy-Weinberg equilibrium (p0.05). The allele variants of β-LG and CSN3 might be more investigated before include them into future breeding schemes designed for Girolando dairy cattle with objective of improving milk traits as milk yield in up to 305 days (305MY) and predicted transmission capacity (PTA) for 305MY.


Em rebanhos leiteiros, um dos objetivos mais importantes da seleção é a melhoria da produção e composição do leite. Vários estudos demonstraram que os genes candidatos da kappa-caseína (CSN3) e da β-lactoglobulina (β-LG) estão associados a produção de leite, qualidade do leite e características de saúde em animais leiteiros. Portanto, o objetivo deste estudo foi detectar polimorfismos nos genes CSN3 e β-LG e avaliar possíveis associaçoes desses polimorfismos com a produção de leite em até 305 dias (305MY) e a capacidade de transmissão prevista (PTA) de leite em bovinos da raça Girolando. No total, 138 touros e 729 vacas (n = 867) foram amostrados. A genotipagem foi realizada pelo método PCR-RFLP utilizando as enzimas HinfI e HaeIII para os genes CSN3 e β-LG, respectivamente. Os resultados estatísticos revelaram dois alelos A e B para ambos os genes. Os genotipos e alelos mais frequentes para os genes CSN3 e β-LG foram respectivamente: AA (0,7324) e A (0,8558) e AB (0,4827) e A (0,5017). O teste x2 revelou que os dois loci estavam em equilibrio de Hardy-Weinberg (p 0,05). Portanto, as variantes alélicas identificadas nos genes β-LG e CSN3 devem ser mais investigadas antes de serem incluidas nos programas de melhoramento desenhados para bovinos leiteiros Girolando objetivando melhorar as características do leite analisadas no presente estudo.

The objectives of this study were to determine the effects of a single gene and composite genotype of the casein gene family, including the beta-lactoglobulin gene (beta-LGB), acyl-CoA: diacylglycerol acyltransferase 1 gene (DGAT1), growth hormone gene (GH), and luteinizing hormone receptor gene (LHR) on milk yield, milk composition, the percentage of fat, protein, solids-not-fat, and total solid in crossbred Holsteins. A total of 231 crossbred Holstein cows were examined for the study. The genotype of the beta-casein gene was analyzed by allele-specific polymerase chain reaction, while the alpha-S1, alpha-S2, kappa-casein, DGAT1, beta-LGB, and GH genes were analyzed using a polymerase chain reaction-restriction fragment length polymorphism assay. The association between genes and milk yield and milk composition was analyzed. Three pairs of genes, for which significant associations were detected, were beta + kappa-casein, DGAT1 + beta-casein, and GH + beta-LGB. In the single-gene model, most loci are significantly associated with traits. A significant association between the composite genotype and the traits was detected in all composite genotypes. GH + beta-LGB appears to be the most suitable variants for improving milk production and percentage of milk protein. Overall, the effects of the composite genotype and single gene were different. A physical or functional relationship between genes is necessary for investigating gene markers.

The objective was to estimate the allelic and genotypic frequencies, genetic diversity and polymorphic information content for the β-casein, κ-casein and β-lactoglobulin genes. Blood and frozen semen samples were collected from 453 Jersey individuals registered by the Mexican Jersey Cattle Association. Twenty eight breed specific SNP primers for whole genes were used. The B allele of κ-casein had higher frequency (0.69) than the A (0.26) and E (0.05). For β-lactoglobulin, the highest frequency was for B (0.72), followed by A and C alleles (0.26 and 0.02, respectively). The β-casein allele with the highest frequency was A² (0.71), followed by A¹ (0.19), A³ (0.05), B (0.04) and C (0.01). The average genetic diversity (He) was 0.53. The average locus effective allele number was 1.79. These results indicate a high allelic diversity for κ-caseín, β-casein and β-lactoglobulin that could be included in breeding programs in the population studied, aimed to improve the milk quality traits of economic importance.

Cheese yield mainly depends on the amount and proportion of milk constituents; however, genetic variants of the proteins present in milk may also have an important effect. The objective of this research was to study the effect of the variants A and B of ß-lactoglobulin (LG) on cheese yield using a model system consisting of skim milk powder fortified with different levels of a mixture containing α-lactalbumin and ß-LG genetic variants (A, B, or A-B) in a 1:2 ratio. Fortified milk samples were subjected to pasteurization at 65 °C for 30 min. Miniature cheeses were made by acidifying (pH=5.9) fortified milk and incubating with rennet for 1h at 32 °C. The clot formed was cut, centrifuged at 2,600 × g for 30 min at 20 °C and drained for determining cheese yield. Cheese-yielding capacity was expressed as actual yield (grams of cheese curd per 100g of milk) and dry weight yield (grams of dried cheese curd per 100g of milk). Free-zone capillary electrophoresis was used for determining ß-LG A or B recovery in the curd during rennet-induced coagulation. The presence of ß-LG variant B resulted in a significantly higher actual and dried weight cheese yield than when A or A-B were present at levels ≤ 0.675% of whey protein (WP) addition. Results of free-zone capillary electrophoresis allowed us to infer that ß-LG B associates with the casein micelles during renneting, as shown by an increase in the recovery of this variant in the curd when ß-LG B was added up to a maximum at 0.45% (equivalent to 0.675% WP). In general, actual or dried weight cheese yield increased as WP addition was increased from 0.225 to 0.675%. However, when WP addition ranged from 0.675 to 0.90%, a drastic drop in cheese yield was observed. This behavior may be because an increase in the aggregation of casein micelles with a concomitant inclusion of whey protein in the gel occurs at low levels of WP addition, whereas once the association of WP with the casein micelles reach a saturation point at addition levels higher than 0.675%, rearrangements of the gel network result in larger whey expulsion and syneresis. This knowledge is expected to be useful to maximize cheese yield and optimize processing conditions during cheese and cheese analogs manufacturing.

The objective of this study was to characterize the genetic architecture underlying the absolute concentrations of 2 important milk proteins, κ-casein (κ-CN) and ß-lactoglobulin (ß-LG), in a backcross population of (Holstein × Jersey) × Holstein cattle. A genome-wide association analysis was performed using a selective DNA pooling strategy and the Illumina BovineHD BeadChip assay [777,000 (777K) SNP markers; Illumina Inc., San Diego, CA]. After correction for multiple testing, 25 single nucleotide polymorphisms were found to be associated with κ-CN and 36 single nucleotide polymorphisms were associated with ß-LG. A pathway association analysis revealed 15 Gene Ontology (GO) terms associated with the κ-CN trait and 28 GO terms associated with ß-LG. In addition, several GO terms were associated with both milk proteins. Further analysis revealed that κ-CN and ß-LG production is regulated by both kinase and phosphatase activity, including mechanisms regulating the extracellular matrix. These results are in concordance with the complex multihormonal process controlling the expression of milk proteins and interactions between mammary epithelial cells and extracellular matrix components. Although κ-CN and ß-LG milk proteins are expressed by single genes, the results from this study showed that many loci are involved in the regulation of the concentration of these 2 proteins.

The aim of this work was to sequence the promoter region of ß-lactoglobulin (BLG) gene in four sheep breeds, in order to identify polymorphisms, infer and analyze haplotypes, and phylogenetic relationship among the Valle del Belice breed and the other three breeds considered as ancestors. Sequencing analysis and alignment of the obtained sequences showed the presence of 36 single nucleotide polymorphisms (SNPs) and one deletion. A total of 22 haplotypes found in "best" reconstruction were inferred considering the 37 polymorphic sites identified. Haplotypes were used for the reconstruction of a phylogenetic tree using the Neighbor-Joining algorithm. The number of polymorphisms identified showed high variability within breeds. Analysis of genetic diversity indexes showed that the Sarda breed presented the lowest nucleotide diversity, whereas the Comisana breed presented the highest one. Comparing the nucleotide diversity among breeds, the highest value was obtained between Valle del Belice and Pinzirita breeds, whereas the lowest one was between Valle del Belice and Sarda breeds. Considering that polymorphisms in the promoter region of BLG gene could have a functional role associated with milk composition, the lowest value of nucleotide diversity between Valle del Belice and Sarda breeds may be related to a higher similarity of milk composition of these two breeds compared to the others. Further analyses will be conducted in order to evaluate the possible correlation between the genetic diversity indexes and the BLG content in milk of our breeds.
