RESUMO

In brief: Social insect queens display both extraordinary longevity and fertility. In this point of view, we describe their distinctive traits that make them useful models for reproductive longevity, holding implications for human health discoveries. Abstract: Social insects present an extraordinary opportunity as models for reproductive longevity because they challenge the conventional patterns of aging and reproduction seen in other model organisms. Their queens are simultaneously long-lived and highly fecund, and understanding how these traits co-occur may lead to discoveries with important implications for human health.

The coffee berry borer (CBB) Hypothenemus hampei Ferrari is the most serious pest of coffee worldwide. Management of the CBB is extremely difficult because its entire life cycle occurs inside the fruit, where it is well protected. Knowing which life stages contribute most to population growth, would shed light on the population dynamics of this pest and help to improve CBB management programs. Two staged-classified matrices were constructed for CBB populations reared in the lab on artificial diets and CBB populations from artificial infestations in the field. Matrices were used to determine demographic parameters, to conduct elasticity analyses, and to perform prospective perturbation analysis. Higher values of the intrinsic rate of natural increase (rm) and population growth rate (λ): were observed for CBB populations growing in the lab than in the field (rm: 0.058, λ: 1.74 lab; rm: 0.053, λ: 1.32 field). Sensitivity values for both CBB populations were highest for the transitions from larva to pupa (G2: 0.316 lab, 0.352 field), transition from pupa to juvenile (G3: 0.345 lab, 0.515 field) and survival of adult females (P5: 0.324 lab, 0.389 field); these three vital rates can be important targets for CBB management. Prospective perturbation analyses indicated that an effective management for the CBB should consider multiple developmental stages; perturbations of >90% for each transition are necessary to reduce λ to <1. However, when the three vital rates with highest sensitivity are impacted at the same time, the percentage of perturbation is reduced to 25% for each transition; with these reductions in survival of larvae, pupae and adult females the value of λ was reduced from 1.32 to 0.96. Management programs for CBB should be focused on the use of biological and cultural measures that are known to affect these three important targets.

Health and lifespan are influenced by dietary nutrients, whose balance is dependent on the supply or demand of each organism. Many studies have shown that an increased carbohydrate-lipid intake plays a critical role in metabolic dysregulation, which impacts longevity. Caenorhabditis elegans has been successfully used as an in vivo model to study the effects of several factors, such as genetic, environmental, diet, and lifestyle factors, on the molecular mechanisms that have been linked to healthspan, lifespan, and the aging process. There is evidence showing the causative effects of high glucose on lifespan in different diabetic models; however, the precise biological mechanisms affected by dietary nutrients, specifically carbohydrates and lipids, as well as their links with lifespan and longevity, remain unknown. Here, we provide an overview of the deleterious effects caused by high-carbohydrate and high-lipid diets, as well as the molecular signals that affect the lifespan of C. elegans; thus, understanding the detailed molecular mechanisms of high-glucose- and lipid-induced changes in whole organisms would allow the targeting of key regulatory factors to ameliorate metabolic disorders and age-related diseases.

Parkinson's disease (PD) is a complex progressive neurodegenerative disorder involving impairment of bodily movement caused by the specific destruction of dopaminergic (DAergic) neurons. Mounting evidence suggests that PD might be triggered by an interplay between environmental neurotoxicants (e.g., paraquat, PQ), heavy metals (e.g., iron), and gene alterations (e.g., PARKIN gene). Unfortunately, there are no therapies currently available that protect, slow, delay, or prevent the progression of PD. Melatonin (Mel, N-acetyl-5-methoxy tryptamine) is a natural hormone with pleiotropic functions including receptor-independent pathways which might be useful in the treatment of PD. Therefore, as a chemical molecule, it has been shown that Mel prolonged the lifespan and locomotor activity, and reduced lipid peroxidation (LPO) in wild-type Canton-S flies exposed to PQ, suggesting antioxidant and neuroprotective properties. However, it is not yet known whether Mel can protect or prevent the genetic model parkin deficient in flies against oxidative stress (OS) stimuli. Here, we show that Mel (0.5, 1, 3 mM) significantly extends the life span and locomotor activity of TH > parkin-RNAi/ + Drosophila melanogaster flies (> 15 days) compared to untreated flies. Knock-down (K-D) parkin flies treated with PQ (1 mM) or PQ (1 mM)/iron (1 mM) significantly diminished the survival index and climbing abilities (e.g., 50% of flies were dead and locomotor impairment by days 4 and 3, respectively). Remarkably, Mel reverted the noxious effect of PQ or PQ/iron combination in K-D parkin. Indeed, Mel protects TH > parkin-RNAi/ + Drosophila melanogaster flies against PQ- or PQ/iron-induced diminish survival, locomotor impairment, and LPO (e.g., 50% of flies were death and locomotor impairment by days 6 and 9, respectively). Similarly, Mel prevented K-D parkin flies against both PQ and PQ/iron. Taken together, these findings suggest that Mel can be safely used as an antioxidant and neuroprotectant agent against OS-stimuli in selective individuals at risk to suffer early-onset Parkinsonism and PD.

Parasitoids can be used as biological agents of pest control. Anagyrus saccharicola Timberlake (Hymenoptera: Encyrtidae) is a parasitoid of the pink sugarcane mealybug Saccharicoccus sacchari (Cockerell) (Hemiptera: Pseudococcidae). Although this mealybug is present in all sugarcane-producing countries, there is limited information regarding this pest and its parasitoid. Aiming to elucidate information on bioecological parameters of A. saccharicola, were evaluated the survival of parasitoid females and males at three temperatures, the host preference of the parasitoid, and the fecundity and longevity of the host. In addition, the parasitism rate of A. saccharicola was estimated based on three factors, feeding, mating, and time. Survival was evaluated at 20, 25, and 30°C. Host preference was conducted on 15-, 20-, and 30-day-old mealybugs. And the parasitism rate was evaluated in fed and unfed, mated and unmated parasitoids and with 24 h and newly emerged. The temperature of 20°C was the most favorable for parasitoid survival. Parasitism occurred at all evaluated ages of the mealybug; however, the preference was for those that were 30-days-old. The parasitized mealybugs longevity was approximately 8 additional days after parasitization, and non-parasitized mealybugs lived for an additional 20 days for mealybugs aged 30 and 20 days at the outset of the tests, and a further 13 days for the 15 days. Feeding and mating after 24 h of emergence resulted in a higher parasitism rate. These findings can contribute to more efficient rearing of A. saccharicola and in the planning of the biological control of S. sacchari in the integrated pest management programs.

Our ability to effectively prevent the transmission of the dengue virus through targeted control of its vector, Aedes aegypti, depends critically on our understanding of the link between mosquito abundance and human disease risk. Mosquito and clinical surveillance data are widely collected, but linking them requires a modeling framework that accounts for the complex non-linear mechanisms involved in transmission. Most critical are the bottleneck in transmission imposed by mosquito lifespan relative to the virus' extrinsic incubation period, and the dynamics of human immunity. We developed a differential equation model of dengue transmission and embedded it in a Bayesian hierarchical framework that allowed us to estimate latent time series of mosquito demographic rates from mosquito trap counts and dengue case reports from the city of Vitória, Brazil. We used the fitted model to explore how the timing of a pulse of adult mosquito control influences its effect on the human disease burden in the following year. We found that control was generally more effective when implemented in periods of relatively low mosquito mortality (when mosquito abundance was also generally low). In particular, control implemented in early September (week 34 of the year) produced the largest reduction in predicted human case reports over the following year. This highlights the potential long-term utility of broad, off-peak-season mosquito control in addition to existing, locally targeted within-season efforts. Further, uncertainty in the effectiveness of control interventions was driven largely by posterior variation in the average mosquito mortality rate (closely tied to total mosquito abundance) with lower mosquito mortality generating systems more vulnerable to control. Broadly, these correlations suggest that mosquito control is most effective in situations in which transmission is already limited by mosquito abundance.

INTRODUÇÃO: Idosos com 80 anos ou mais, ou longevos, são o subgrupo etário que mais cresce no mundo. Nesse segmento, a incapacidade funcional (IF) é mais prevalente que em outras faixas etárias. OBJETIVO: Caracterizar a capacidade funcional e analisar potenciais associações em idosos longevos de uma capital brasileira, com base em um modelo de decisão. MÉTODO: Estudo transversal com 100 idosos longevos, não institucionalizados, assistidos pela Estratégia Saúde da Família do Recife (PE), selecionados por amostra probabilística. Foram avaliados dados sociodemográficos, econômicos e clínicos coletados por meio de entrevistas domiciliares, aferição de medidas antropométricas e buscas nos prontuários. Para análise bivariada, foi empregado o teste χ2 de Pearson, considerando o nível de significância p < 0,05, e para a multivariada foi construída uma árvore de decisão com base em um algoritmo Exhaustive CHAID. RESULTADOS: A prevalência de IF na amostra correspondeu a 67,0%. Na análise bivariada, contribuíram para esse desfecho: renda (p = 0,032), situação previdenciária (p < 0,01), situação nutricional (p = 0,010), doenças neurológicas (p < 0,01), neoplasias (p < 0,01), saúde autopercebida (p = 0,025) e rede de apoio social (p = 0,032), permanecendo na análise multivariada: renda (p = 0,003), rede de apoio social (p = 0,032) e situação nutricional (p = 0,040). A árvore de decisão possibilitou a identificação das variáveis mais associadas ao desfecho, sendo capaz de prever adequadamente a dependência moderada, com assertividade de 72,1%. CONCLUSÃO: O modelo de decisão mostrou-se uma ferramenta oportuna na dedução dos determinantes mais relevantes da IF. Seu uso potencialmente contribui para ampliar a precisão diagnóstica e identificação de populações de risco.


INTRODUCTION: The oldest old adults, aged 80 years and above, is the fastest growing age group in the world. In this section of the population, functional disability (FD) is more prevalent compared to other age groups. OBJECTIVE: To characterize functional capacity and analyze potential associations in the oldest old from a Brazilian capital city, based on a decision model. METHODS: Cross-sectional study of 100 non-institutionalized oldest old participants assisted at the Family Health Strategy of Recife, in the Brazilian northeastern state of Pernambuco, selected by probabilistic sample. Sociodemographic, economic, and clinical data were collected by means of home interviews, anthropometric measurements, and medical records. For bivariate analysis, Pearson's chi-square test was used, establishing significance at p < 0.05. For the multivariate analysis, a decision tree was built from the Exhaustive CHAID algorithm. RESULTS: The prevalence of FD in the sample corresponded to 67.0%. In the bivariate analysis, the following data contributed to this outcome: income (p = 0.032), social security status (p < 0.01), nutritional status (p = 0.010), neurological diseases (p < 0.01), neoplasms (p < 0.01), self-perceived health (p = 0.025) and social support network (p = 0.032), remaining in the multivariate analysis: income (p = 0.003), social support network (p = 0.032), and nutritional status (p = 0.040). The decision tree allowed the identification of the variables most strongly associated with the outcome, being able to adequately predict moderate dependence, with 72.1% assertiveness. CONCLUSION: The decision model proved to be a timely tool in deducing the most relevant determinants of FD. Its use potentially contributes to increase the accuracy of the diagnosis and to identify populations at risk.

OBJETIVO: Avaliar o conhecimento científico produzido em relação aos fatores associados à prática de atividade física de idosos diabéticos assistidos na atenção primária à saúde no Brasil. MÉTODOS: Trata-se de uma revisão integrativa, e a busca por estudos ocorreu de forma on-line, sendo incluídos artigos originais da língua portuguesa, inglesa ou espanhola, publicados de outubro de 2008 a setembro de 2018, nas bases de dados Medical Literature and Retrieval System Online (MEDLINE), Literatura Latino-Americana e do Caribe em Ciências da Saúde (LILACS) e Scopus. Os descritores utilizados foram: idosos, diabetes mellitus, exercícios e atenção primária à saúde. A qualidade metodológica dos artigos foi avaliada por meio de dois instrumentos: Critical Appraisal Skills Programme e Hierarchy of Evidence for Intervention Studies. Após análise, quatro artigos foram incluídos. RESULTADOS: Não foram encontrados artigos com amostras constituídas apenas de idosos, e somente um deles fazia uma análise diferenciada para essa faixa etária, chamando atenção para a necessidade de estudos abordando idosos diabéticos, por causa das especificidades desses indivíduos. Nenhum dos artigos fez uso de instrumentos para avaliação da atividade física. CONCLUSÃO: A prática da atividade física esteve associada à melhora da qualidade de vida, do autocuidado e da adesão ao tratamento do diabetes mellitus, evidenciando a importância do aconselhamento dessa prática por meio dos profissionais de saúde. Fatores como frequência e tipo da prática de atividade física não foram verificados nos estudos, o que destaca a necessidade do uso de instrumentos para a sua avaliação.


OBJECTIVE: To evaluate the scientific knowledge produced in relation to the factors associated to the practice of physical activity of older diabetic primary care patients in Brazil. METHODS: It involved an on-line research including original articles published between October 2008 and September 2018 in Portuguese, English, and Spanish, from the databases: Medline, Lilacsand Scopus. The keywords used were elderly, diabetes mellitus, exercise, and primary health care. The methodological quality of the articles was evaluated by means of two instruments: Critical Appraisal Skills Programme and Hierarchy of Evidence for Intervention Studies. After analysis, four articles were included. RESULTS: No articles were found with samples consisting only of older people, and only one of them made an exclusive analysis for this age group. There is a need for studies addressing the older diabetic individuals, due to the specificities of this population. None of the articles made use of instruments for physical activity evaluation. CONCLUSION: The practice of physical activity was associated to an improvement in quality of life, self-care, and adherence to treatment for diabetes mellitus, highlighting the importance of professional counseling. Factors such as frequency and type of physical activity were not verified in the studies, which highlights the need for the use of instruments for this evaluation.

The white leg Litopenaeus vannamei shrimp is of importance to the eastern Pacific fisheries and aquaculture industry but suffer from diseases such as the recently emerged early mortality syndrome. Many bacterial pathogens have been identified but the L. vannamei microbiota is still poorly known. Using a next-generation sequencing (NGS) approach, this work evaluated the impact of the inclusion in the diet of mannan oligosaccharide, (MOS, 0.5% w/w), over the L. vannamei microbiota and production behavior of L. vannamei under intensive cultivation in Ecuador. The MOS supplementation lasted for 60 days, after which the shrimp in the ponds were harvested, and the production data were collected. MOS improved productivity outcomes by increasing shrimp survival by 30%. NGS revealed quantitative differences in the shrimp microbiota between MOS and control conditions. In the treatment with inclusion of dietary MOS, the predominant phylum was Actinobacteria (28%); while the control group was dominated by the phylum Proteobacteria (30%). MOS has also been linked to an increased prevalence of Lactococcus- and Verrucomicrobiaceae-like bacteria. Furthermore, under the treatment of MOS, the prevalence of potential opportunistic pathogens, like Vibrio, Aeromonas, Bergeyella and Shewanella, was negligible. This may be attributable to MOS blocking the adhesion of pathogens to the surfaces of the host tissues. Together, these findings point to the fact that the performance (survival) improvements of the dietary MOS may be linked to the impact on the microbiota, since bacterial lines with pathogenic potential towards shrimps were excluded in the gut.

Contrasting with several theories of ageing, bats are mammals with remarkable longevity despite their high metabolic rate, living on average three times more than other mammals of equal size. The question of how bats live a long time has attracted considerable attention, and they have thus been related to immortal fantasy characters like Dracula in the novel by Bram Stoker. Several ecological and physiological features, such as reduction in mortality risks, delayed sexual maturation and hibernation, have been linked to bats' long lifespan. However, there is still very little information about the molecular mechanisms associated with the longevity of bats. In this regard, the present work tries to summarize current knowledge about how bats can live for so long, taking into consideration nutritional factors, oxidative metabolism, protein homeostasis, stress resistance, DNA repair, mitochondrial physiology and cancer resistance.
