This study aimed to evaluate the effects of calcium propionate (PrCa), PrCa + monensin sodium (PrCa + Mon), and PrCa + Saccharomyces cerevisiae (PrCa + Sc) on the productive performance of Holstein steers. Twenty-four Holstein steers (270.0 ± 25.85 kg) were distributed individually into four treatments of six replicates. The treatments were control (no additives), PrCa (10 g/kg), PrCa + Mon (10 g/kg + 30 mg/kg), and PrCa + Sc (10 g/kg + 12.8 × 109 cfu). The steers were fed for 43 days, and afterwards, nutrient intake and digestibility as well as volatile fatty acids were determined, while the weight gained, feed efficiency, and CH4 production were calculated. Diet of PrCa + Sc had the highest (P < 0.0001) acid detergent fiber intake and propionate acid as well as the nutrient digestibility, with lowest (P < 0.0001) rumen acetic acid, methane, and protozoa concentration versus other diets. In conclusion, dietary inclusion of PrCa + Sc (10 g/kg + 12.8 × 109 cfu) improved nutrient digestibility, rumen fermentation, and reduced methane emission, thereby enhancing the possibility of ecofriendly ruminant farming.

Monensin sodium is one of the most common ionophores used in livestock feeding; however, ionophores are condemned by organized consumer groups. Bioactive compounds from plants found in the seasonally dry tropical forest have similar mechanisms of action as ionophores. The aim was to investigate the effects of replacing monensin sodium with phytogenic additives on the nutritional efficiency of beef cattle. Five 14-month-old Nellore bulls (average body weight 452.68 ± 42.60 kg) were used in the study. The experiment was designed as a 5 × 5 Latin Square (five treatments and five 22-day experimental periods). Within each period, 15 days were used for adaptation of animals to experimental conditions and 7 days for data collection. Bulls were fed a control diet (without additives), monensin (a diet containing monensin sodium 40% as a synthetic additive), and three diets containing phytogenic additives prepared from Anadenanthera macrocarpa (Benth) Brenan, Mimosa tenuiflora (Willd) Poiret, or Prosopis juliflora (Sw.) DC. Nutritional efficiency was assessed through feed intake, nutrient digestibility, feeding behavior, and hematological parameters. Monensin and phytogenic additives did not influence (P > 0.05) feeding behavior or hematological parameters, but the nutrient intake was highest for bulls supplemented phytogenic additives (P < 0.05). Monensin supplementation did not influence (P > 0.05) feed intake. The phytogenic additives and monensin sodium increased (P < 0.05) the nutrient digestibility. Therefore, the phytogenic additives from P. juliflora, A. macrocarpa, and M. tenuiflora can be recommended to enhance the nutritional efficiency of confined Nellore cattle.

This study aimed to evaluate the influence of sodium monensin on the hepatic accumulation of copper in sheep. Twenty-four Santa Inês crossbred sheep were used and allocated in a 2 × 2 factorial experiment with six repetitions and considering the factors dietary copper (basal and high) and supplementation (with and without sodium monensin). Thus, four homogeneous groups were formed: control (basal diet); monensin (Mon), 30 ppm of monensin; copper (Cu), 10 10 mg/kg BW per day of copper; monensin + copper (MonCu). The experimental period lasted 14 weeks. Liver and bile samples were collected at the beginning and end of the experiment to determine mineral element concentrations, and weekly blood samples for biochemical, hematological, and mineral evaluation. Liver copper concentrations at the beginning of the experiment did not vary between groups, while mean liver copper concentrations at the end of the experiment were higher in the MonCu, Cu, and Mon groups when compared to the control. At the end of the study, hepatic copper concentration was influenced by copper (p = 0.0001) and monensin (p = 0.0003) supplementation. Copper-supplemented groups had reduced liver iron contents (p = 0.0287) and increased copper concentrations in bile. The biochemical evaluation showed increased serum GGT and AST activity (p < 0.05) in the Cu and MonCu groups from the eleventh week on compared to the control and Mon groups. The increase in activity of these enzymes was influenced by copper supplementation (p = 0.0340). Monensin interferes positively with the hepatic accumulation of copper and the supplementation of this additive may predispose sheep to copper poisoning.

This study assessed the association between encapsulated nitrate product (ENP) and monensin (MON) to mitigate enteric methane (CH4) in vitro and possible effects on ruminal degradability, enteric fermentation characteristics, and microbial populations. Six treatments were used in randomized complete design in a 2×3 factorial arrangement with two levels of MON (0 and 2.08 mg/mL of buffered rumen fluid) and three levels of ENP (0, 1.5 and 3.0%). The substrate consisted of 50% Tifton-85 hay and 50% concentrate mixture (ground corn and soybean meal). ENP replaced soybean meal to achieve isonitrogenous diets (15% CP). No ENP×MON interaction was observed for any measured variable (P > 0.05) except for the relative abundance of F. succinogenes (P = 0.02) that linearly increased in diets with MON when ENP was added. The ENP addition decreased CH4 production (P < 0.01) without affecting (P > 0.05) truly degraded organic matter nor the relative abundance of methanogens. Hydrogen production was reduced with MON (P = 0.04) and linearly decreased with ENP inclusion (P = 0.02). We concluded that use of nitrate is a viable strategy for CH4 reduction, however, no additive effect of ENP and MON was observed for mitigating CH4 production.

BACKGROUND: Ionophore antibiotics improve the efficiency of energy metabolism, which has driven their use as a feed additive in ruminants for decades. Currently, they have not been approved in many countries, generating a challenge for the immediate search for plant extracts with a similar mode of action on rumen metabolism. This study evaluated the effects of enriched Prosopis juliflora (mesquite) piperidine alkaloid extract (MPA) levels as an alternative phytoadditive to sodium monensin (MON) in sheep. RESULTS: The MPA diet did not differ from MON with regard to nutrient intake. A quadratic effect (P < 0.05) was observed for organic matter and neutral detergent fibre digestibility, with respective maximum point at 25.40 and minimum point at 0.95 mg kg-1 MPA. The MPA levels linearly decreased (P < 0.05) faecal nitrogen loss. MPA did not differ from MON with regard to nutrient digestibility, and MPA levels increased (P < 0.05) the proportion of digestible energy and metabolizability from dietary gross energy. The MPA levels linearly decreased (P < 0.05) enteric CH4 production, the yield showing lower (P < 0.05) energy loss as CH4 than MON. CONCLUSION: The results show that MPA levels of 17.3 and 27.8 mg kg-1 are enteric CH4 inhibitors and enhance energy and protein utilization, indicating a promising alternative to MON for ruminants. © 2022 Society of Chemical Industry.

The objective of this study was to evaluate diets containing monensin (MON) associated or not with virginiamycin (VM) or functional oil based on cashew nut shell and castor beans (FOcc) for beef cattle in feedlots on nutritional (intake and digestibility) and productive parameters. A total of 1410 non-castrated Nellore cattle were selected, with an average age of 18 months and with an initial mean body weight (BW) of 305 ± 41.52 kg. The diet showed a roughage to concentrate ratio of 23:77, with the supply of corn silage as a source of roughage. The following additive inclusions in the diet were evaluated: (1) MON: 27 mg MON/kg dry matter (DM); (2) MON + VM: 22 mg MON/kg DM + 19 mg VM/kg DM; and (3) MON + FOcc: 22 mg MON/kg DM + 500 mg FOcc/kg DM. Statistical analyses were obtained through a linear model using initial BW and days of feedlot as covariables and comparisons between treatments using mutually orthogonal linear contrasts with a 5% significance level. The association or not of MON with VM or FOcc does not affect any of the nutritional and productive parameters evaluated. Animals that receive diets with MON + VM have higher average daily gain and feed efficiency (FE) than those that receive MON + FOcc without showing differences in nutritional parameters. The supply of MON associated with VM or FOcc does not increase intake and productive performance and, consequently, efficiency of feedlot beef cattle. However, in the case of use associated with MON, the VM provides greater performance than FOcc without changing food intake.

This study evaluated the growth, physiology, and coccidiosis infestation of suckling beef calves provided monensin and grazing limpograss (Exp. 1) or bahiagrass (Exp. 2) pastures. Treatments were randomly assigned to pastures (4 pastures/treatment; 3 cow-calf pairs/pasture in Exp. 1; 4 pastures/treatment; 10 cow-calf pairs/pair of pastures in Exp. 2) and comprised of supplementation of 0.40 kg/d of soybean meal added or not with monensin (20 mg/kg of total DM intake) for 112 and 78 days before weaning in Exp. 1 and 2, respectively. In Exp. 1, supplement DM disappearance tended (P = 0.10) to be less for calves supplemented with vs. without monensin, but treatment × day and treatment effects were not observed (P ≥ 0.18) for herbage mass (HM), herbage allowance (HA), cow body condition score (BCS), calf average daily gain (ADG), calf plasma data, and fecal coccidia egg count. In Exp. 2, forage nutritive value, HM and HA, and cow BCS did not differ (P ≥ 0.43) between treatments. Supplemental monensin did not impact (P ≥ 0.78) plasma concentrations of insulin-like growth factor 1 but increased (P ≤ 0.05) calf overall ADG and plasma concentrations on day 78 and reduced plasma concentrations of urea nitrogen (PUN) on day 78 and fecal coccidia egg count on day 78 compared to calves provided no monensin supplementation. Hence, monensin supplementation successfully improved growth performance of creep-fed suckling beef calves, when herbage mass was not a limiting factor and coccidiosis infestation occurred.

Highly dynamic ribosomes, glycogen granules, thinly fibrillar material, and multiple membrane-bound vesicles are embedded in the matrix-rich cytoplasm of Entamoeba spp. trophozoites. The absence of a Golgi apparatus in these amoebae has been commonly accepted. Here we challenge this observation by incubating Entamoeba histolytica and Entamoeba dispar with monensin, an ionophore that produces swelling of the Golgi apparatus. We observe changes in the trophozoites through standard transmission electron microscopy, cryofixation and cryosubstitution, and analyze the label and expression of known resident proteins of the cis-GM130 and trans-TGN38 Golgi network through confocal microscopy and Western blot assays. Cryosubstitution and standard methods using the treatment, preserved membranous lamellae resembling Golgi components. GM130 and TGN38 Golgi antigens were found by immunoelectron, immunoblot, and co-localization by confocal microscopy using the reagent NBD C6-ceramide. Our results indicate that previously undetected Golgi apparatus components are present in the cytoplasm of E. histolytica and E. dispar.

This study was performed with the main objective of evaluating the effect of the combination of pelleting and monensin on fatty acids (FA) composition, the concentration of total polyphenols and flavonoids, and the oxidative stability of milk in cows fed a concentrate containing soybean seeds. Eight Holstein multiparous cows were distributed in a replicated Latin square design. The four supplement treatments consisted of the combination of two factors (pelleting and monensin) and one concentrate as follows: (1) unpelleted concentrate with no monensin (CO); (2) pelleted concentrate with no monensin (PE); (3) unpelleted concentrate with 96 mg of monensin/kg of dry matter, DM (MO); and (4) pelleted concentrate with 96 mg of monensin/kg of DM (PM). There was no interaction between pelleting and monensin for milk production and concentration of milk protein, lactose, total polyphenols, flavonoids, conjugated dienes (CD), and reducing power. Fat and total solids concentration in milk were decreased when cows were fed pelleted (PE and PM) concentrates. Feeding cows with PE and PM concentrates increased the CD concentration in milk. Regarding milk FA concentration, there was no difference among treatments for total saturated, monounsaturated, and polyunsaturated FA. The most prominent result was that pelleting increased the milk concentration of omega-3 FA. Altogether, the present study suggests that the pelleting process can improve the milk fat quality by increasing the omega-3 FA, while the combination of pelleting and monensin in the diet of grazing dairy cows fed soybean-based concentrate adds no further improvements to FA profiles and oxidative stability of milk.

The aim of the study was to evaluate the effects of a cashew nut shell oil and commercial castor oil blend (CNSL-Castor oil) on the performance and microbiota of broiler chickens with and without coccidiosis challenge. A total of 864 one-day-old male chicks (Cobb) were randomly distributed to receive 6 treatments (8 pens/treatment; 18 chicks/pen) in a 3 × 2 factorial, with 3 additives (control [non-additives], 100 ppm sodium monensin, or 0.15% CNSL-Castor oil blend), and 2 levels of coccidiosis challenge at 14 D of age (unchallenged or inoculated by gavage with 1 mL of solution containing oocysts sporulated with Eimeria tenella, Eimeria acervulina, and Eimeria maxima). No differences in productive performance were observed among treatments in the pre-challenge period and in unchallenged birds (P > 0.05). Seven-days post-challenge, birds receiving monensin performed better than birds in the positive control group (non-additive and challenge) or in the CNSL-Castor oil group (P > 0.05). However, 14 D post-challenge, birds supplemented with CNSL-Castor oil presented higher weight gain and better feed conversion (P > 0.05), without any change in feed intake (P > 0.05). During the accumulated period (1 to 42 D of age), the live weight, weight gain, and feed intake did not differ between the CNSL-Castor oil and monensin groups, both of which presented higher values than the positive control. Lactobacillus spp. and Clostridium perfringens numbers were increased in the challenged birds (P < 0.05). CNSL-Castor oil supplementation reduced Clostridium cluster XIV, C. perfringens, and S. aureus, compared with the monensin and control groups (P > 0.05). In addition, the CNSL-Castor oil group presented the highest number of Lactobacillus spp. copies, followed by the monensin and positive control groups (P > 0.05). Thus, monensin and CNSL-Castor oil effectively minimized the impact of coccidiosis at different times. While monensin acts as an antimicrobial, CNSL-Castor oil modulates the intestinal microbiota with antimicrobial action against gram-positive bacteria, mainly C. perfringens and S. aureus.
