RESUMO

Soluble extracts of Bufo ovaries agglutinate sialidase-treated rabbit erythrocytes. Unlike other amphibian lectins this agglutination activity does not require the presence of calcium ions. It is specifically inhibited by D-galactose and its derivatives. Thiodi-D-galactoside is the most potent saccharide inhibitor followed by lactose and methyl-beta-D-galactoside, respectively. D-Fucose, D-glucose and D-mannose do not inhibit the activity at concentrations at or above 100 mM. The lectin has been purified 500-fold to apparent homogeneity from the ovaries by salt extraction and affinity chromatography on lactose-aminophenyl-agarose, with a yield of about 0.2%. The molecular mass determined by gel filtration under native conditions was 30 kDa; polyacrylamide gel electrophoresis in SDS gave a molecular mass of 15 kDa, suggesting that the lectin is a dimer. The lectin has an isoelectric point of 40 and contains a high proportion of acidic amino acids.

RESUMO

1. Gas-liquid chromatography and mass spectrometry were used to verify the presence of arachidonic acid (20:4 omega 6) in adult male and female organs of the blood-suckling bug Triatoma infestans. Fat body, gonad and head lipids were analyzed. 2. Male gonads contained the highest percentage of phospholipids and the highest percentage of arachidonic acid. 3. Phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol, phosphatidylserine and sphingomyelin were isolated from gonads and heads of sexed insects and the fatty acid composition analyzed. 4. All the phospholipids analyzed contained relevant percentages of linoleic acid and fair percentages of arachidonic acid, except for sphingomyelin where no 20:4 omega 6 acid was found. 5. However, arachidonic acid was specifically incorporated in phosphatidylinositol since this phospholipid contained very large percentages of the acid. 6. In male gonads, phosphatidylcholine also contained a similar high percentage of the acid. Arachidonic acid is apparently incorporated, selectively, into these lipids from the blood of the host.

PIP: The study investigates hormonal control of the menstrual cycle, adrenal function role, and endouterine menstrual disorders by an analysis of peripheral hormones and of their concentration at endometrial level. The authors also present a new radioimmunoassay method for determining hormone presence in plasma and in the endometrium. 59 ovulatory cycles were considered, and ovarian hormones were found to be 10-20 times higher in tissue than in plasma, but not in women under sequential or combined oral contraceptive treatment. The relation between endometrial estrogens and progesterone with plasma follicle stimulating hormone and luteinizing hormone levels is discussed, together with the significance of tissue hormone concentration.^ieng
