RESUMO

Paspalum notatum Flüggé is an economically important subtropical fodder grass that is widely used in the Americas. Here, we report a new chromosome-scale genome assembly and annotation of a diploid biotype collected in the center of origin of the species. Using Oxford Nanopore long reads, we generated a 557.81 Mb genome assembly (N50 = 56.1 Mb) with high gene completeness (BUSCO = 98.73%). Genome annotation identified 320 Mb (57.86%) of repetitive elements and 45,074 gene models, of which 36,079 have a high level of confidence. Further characterisation included the identification of 59 miRNA precursors together with their putative targets. The present work provides a comprehensive genomic resource for P. notatum improvement and a reference frame for functional and evolutionary research within the genus.

The Paspalum genus has potential for further genetic improvement because of its adaptability to different ecosystems and production of high yields for grazing livestock. We estimate the genetic parameters of 195 intraspecific P. notatum hybrids using Restricted Maximum Likelihood (REML), followed by selection based on Best Linear Unbiased Prediction (BLUP) through multivariate analysis. The intraspecific hybrids studied showed considerable genetic variability in the evaluated forage traits, displaying their potential for progression in subsequent stages of the genetic improvement program. Notably, plant height emerged as an important trait for indirect selection to enhance forage production. The use of the REML/BLUP procedure proves to be a robust tool for data analysis, particularly for perennial species. Furthermore, multivariate analysis based on BLUPs should be used in the selection process within breeding programs. Based on the BLUP values, hybrids D3, D16, C17, C2 and B17 were identified as superior for forage production, and they hold promise for future breeding programs for future breeding initiatives aimed at direct selection to improve yield.

KEY MESSAGE: Interspecific comparison of two Paspalum species has demonstrated that mating systems (selfing and outcrossing) contribute to variation (genetically and morphologically) within species through similar but mutually exclusive processes. Mating systems play a key role in the genetic dynamics of populations. Studies show that populations of selfing plants have less genetic diversity than outcrossing plants. Yet, many such studies have ignored morphological diversity. Here, we compared the morphological and molecular diversity patterns in populations of two phylogenetically-related sexual diploids that differ in their mating system: self-sterile Paspalum indecorum and self-fertile P. pumilum. We assessed the morphological variation using 16 morpho-phenological characters and the molecular diversity using three combinations of AFLPs. We compared the morphological and molecular diversity within and among populations in each mating system. Contrary to expectations, selfers showed higher morphological variation within populations, mainly in vegetative and phenological traits, compared to outcrossers. The high morphological variation within populations of selfers led to a low differentiation among populations. At molecular level, selfing populations showed lower levels of genotypic and genetic diversity than outcrossing populations. As expected, selfers showed higher population structure than outcrossers (PhiST = 0.301 and PhiST = 0.108, respectively). Increased homozygous combinations for the same trait/locus enhance morphological variation and reduce molecular variation within populations in selfing P. pumilum. Thus, selfing outcomes are opposite when comparing morphological and molecular variation in P. pumilum. Meanwhile, pollen flow in obligate outcrossing populations of P. indecorum increases within-population molecular variation, but tends to homogenize phenotypes within-population. Pollen flow in obligate outcrossers tends to merge geographically closer populations; but isolation by distance can lead to a weak differentiation among distant populations of P. indecorum.

The genetic systems of Paspalum species have not been extensively studied. We analyzed the ploidy, reproductive mode, mating system, and fertility of four Paspalum species-Paspalum durifolium, Paspalum ionanthum, Paspalum regnellii, and Paspalum urvillei. An analysis of 378 individuals from 20 populations of northeastern Argentina was conducted. All populations of the four Paspalum species were pure tetraploid and had a sexual and stable reproductive mode. However, some populations of P. durifolium and P. ionanthum showed low levels of apospory. Populations of P. durifolium and P. ionanthum had low seed sets under self-pollination but were fertile under open pollination, showing that self-incompatibility likely caused self-sterility. In contrast, populations of P. regnellii or P. urvillei showed no evidence of apospory, and seed sets in both self- and open pollination conditions were high, suggesting that they are self-compatible due to the absence of pollen-pistil molecular incompatibility mechanisms. The evolutionary origin of the four Paspalum species could explain these differences. This study supplies valuable insights into the genetic systems of Paspalum species, which could have implications for their conservation and management.

The tetraploid germplasm of Paspalum contains a large diversity that can be used to generate better forages. The objective was to evaluate a group of Paspalum notatum and Paspalum simplex apomictic hybrids for a set of agronomic traits and apomixis expressivity. Forage yield, cold tolerance, winter regrowth, and seed yield were evaluated. The expressivity of apomixis was evaluated in P. simplex hybrids by flow cytometry. Progeny testing with molecular markers was used to determine the genotypic variability in the progeny. Differences within P. notatum and P. simplex hybrids were observed for all traits, and some of them were superior in comparison with the controls. The accumulated forage yield during three years was 988 g m-2 in the P. notatum hybrids, whereas, in P. simplex, the average forage yield per harvest (40 days of regrowth) was 180 g m-2. In P. simplex, the apomixis expressivity varied between 0 and 100%, and 65% of the hybrids showed high apomixis expressivity (superior to 70%). The genotypic mean homogeneity in the progeny was 76% and 85% in P. notatum and P. simplex, respectively. The generation of hybrids with high apomixis expressivity that combine good agronomic performance and homogeneity in the offspring is possible in tetraploid P. notatum and P. simplex.

The objectives were (a) to evaluate whether grasses native to the Pampa biome, Axonopus affinis Chase, Paspalum notatum Flüggé and Paspalum plicatulum Michx, and the invasive grass Cynodon dactylon (L.). Pers have the potential to phytoremediate soil contaminated with Cu (0, 35 and 70 mg Cu kg-1); (b) assess whether the growth of these species is compromised by the excess of Cu available in the soil; and (c) determine the impact of excess Cu on the physiological responses of the studied species. C. dactylon presented the best performance in soil contaminated with 35 mg of Cu kg-1. In C. dactylon, the concentrations of chlorophyll b and carotenoids increased, as did the photosynthetic rate and plant growth. Phytotoxic effects of Cu in soil contaminated with 70 mg of Cu kg-1 were more severe on A. affinis and led to plant death. The other species presented reduced photosynthetic and growth rates, as well as increased activity of antioxidant enzymes such as superoxide dismutase and guaiacol peroxidase. This very same Cu level has decreased photosynthetic pigment concentrations in P. notatum and P. plicatulum. On the other hand, it did not change chlorophyll a and b concentrations in C. dactylon and increased carotenoid concentrations in it. High values recorded for Cu bioaccumulation-in-grass-root factor, mainly in P. plicatulum, have indicated that the investigated plants are potential phytostabilizers. High C. dactylon biomass production-in comparison to other species-compensates for the relatively low metal concentration in its tissues by increasing metal extraction from the soil. This makes C. dactylon more efficient in the phytoremediation process than other species.

RESUMEN El objetivo del estudio fue la caracterización productiva y de emisiones modeladas de gases de efecto invernadero (GEI) en 61 sistemas lecheros localizados en cinco regiones de Honduras. Durante las fases inicial (FI) y final (FF), con encuestas aplicadas individualmente a los productores, se identificaron aspectos técnicos y de productividad. Variables numéricas expresadas en Microsoft Excel ® permitieron, con el modelo FAO de evaluación ambiental de la ganadería global-interactivo (GLEAM-i, por sus siglas en inglés) de ciclo de vida, estimar emisiones anuales de metano (CH4), óxido nitroso (N2O) y dióxido de carbono (CO2) en cada finca. Cálculos intermedios (GEI/animal) fueron derivados de la modelización GLEAM-i en Excel®. Durante la FI las fincas conjuntamente emitieron 25.038 t CO2 equivalente (CO2-eq), mientras que dichas emisiones fueron 10,5% menores en la FF. Emisiones de GEI/animal (2,85 ± 0,08 t CO2-eq) y de GEI/kg de proteína láctea (96,91 ± 4,50 kg CO2-eq) durante la FI fue-ron 13 y 21% menores en la FF, respectivamente. Valores de 52,82 ± 1,64 (CH4) y 2,66 ± 0,10 (N2O) kg/animal en la FI fueron 13% y 17% menores en la FF, respectivamente. La región Centro-Sur-Oriente emitió la menor cantidad de CH4 (42,95 ± 2,37 kg/animal) y N2O (1,82 ± 0,15 kg/animal, mientras las regiones Occidente y Norte experimentaron una reducción del 27% en GEI/kg proteína láctea entre la FI y FF. Se concluyó que la metodologia usada identificó los impactos productivos y medioambientales, derivados de alternativas técnicas implementadas en sistemas de producción lechera de Honduras.


ABSTRACT The study aimed to characterize production dynamics and greenhouse gas (GHG) emissions from 61 dairy farms in five regions in Honduras. Farm data were collected through individual surveys during the initial and final phases (IP; FP). Using Microsoft Excel, data was incorporated into the global livestock environmental assessment model-interactive (GLEAM-i, FAO) life cycle framework to estimate annual emissions of methane (CH4), nitrous oxide (N2O), and carbon dioxide (CO2) at the farm system level. Animal emissions (GHG/animal) were derived in Excel® from the GLEAM-i predictions. Together, farms during the IP emitted 25.038 t CO2 equivalent (CO2-eq) while these emissions were 10,5% lower in the FP. Emissions of GHG/animal (2,85 ± 0,08 t CO2-eq) and GHG/kg of milk protein (96,91 ± 4,50 kg CO2-eq) during the IP were 13% and 21% lower in the FP, respectively. Methane and N2O emission values (52,82 ± 1,64 vs. 2,66 ± 0,10 kg/animal) were 13% and 17% higher in the IP than in FP. The South-Central region emitted the lowest amount of CH4 and N2O (42,95 ± 2,37 kg/animal vs. 1,82 ± 0,15 kg/animal) while 27% lower GHG/kg milk protein was observed between the IP and FP of the Western and Northern regions. It was concluded that the used methodology identified productive and environmental impacts derived from implemented technical interventions in dairy production systems in Honduras.

Severe water stress is responsible for reducing plant growth and reproduction. This study aimed to evaluate the physiological and biochemical mechanisms associated with the tolerance of two genotipes of bahiagrass (Paspalum notatum Flügge) with different ploidy level to water deficit and flooding at the reproductive stage. Photosynthetic performance of diploid and tetraploid plants was not affected by flooding. In contrast, the water deficit decreased stomatal conductance, increased leaf temperature, and resulted in a decrease in the assimilation rate of the two genotypes. Despite the greater activities of antioxidant enzymes, flooded roots accumulated hydrogen peroxide and malondialdehyde. Roots of plants exposed to water deficit maintained an accumulation of biomass similar to that of control plants; however, with higher levels of total phenol content, total soluble sugars and proline. Diploid plants subjected to flooding had more inflorescences, however, the drought reduced the total number of filled florets per plant. Less starch degradation allows the maintenance and recovery of biomass in the tetraploid genotype, which allows it to maintain its reproductive performance even under drought conditions. Overall, the synthesis of osmoprotectants and activation of antioxidant machinery are important strategies in the tolerance of bahiagrass to water stress at the reproductive stage.

This work aimed to isolate and characterize plant growth promoting rhizobacteria (PGPR) from 10 Paspalum genotypes and evaluate the effect of their inoculation on P. regnellii, P. atratum, and P. malacophyllum genotypes. The bacterial population ranged from undetectable to 107 bacterial cells per gram of fresh matter in the Paspalum genotypes. Initially, we isolated 164 bacteria from rhizospheric soil and roots of the Paspalum genotypes using media N-free LG agar plate, semi-solid NFb, and LGI. The isolates were characterized genetically and physiologically. The sequencing of 16S rRNA showed the presence of many genera, and some are new in association with Paspalum. The most common was Bacillus followed by Rhizobium, Paraburkholderia, Enterobacter, Cupriavidus, Pseudomonas, Dyadobacter and Acinetobacter. Thirty-eight per cent of isolates produced siderophores, 25 % produced solubilized phosphate, and only 9 % produced indolic compounds. Three greenhouse experiments were performed in randomized blocks with six replicates using representative bacterial strains isolated from P. regnellii, P. malacophyllum and P. atratum cv. Pojuca. We also included strain Sp245 (Azospirillum baldaniorum), uninoculated control, and nitrogen control (150 kg N ha–1). There was an increase of up to 53 % in shoot dry matter in P. regnellii inoculated with strain Sp245 and the shoots accumulated more N. In contrast, only small effects were observed for the other Paspalum genotypes inoculated with PGPR from the host genotypes. This study shows a high diversity of diazotrophic rhizosphere bacteria and suggests no strain specificity between the bacterial isolates and the Paspalum genotypes.

The objective of this work was to describe morphology and grouping of Paspalum notatum accessions, based on multicategorical data which discards the redundant variables for quantification of genetic diversity. We also tested the hypothesis that geographical distance was correlated with morphological divergence. In our study, multivariate analyzes successfully demonstrated the geographic and morphological variability of the P. notatum accessions characterized. Many of these evaluated accessions can be included in future genetic improvement programs. Based on two methodologies for discarding variables, it was possible to identify the potentially important morphological characteristics from genetic diversity studies and characterize new accessions aimed at improving forage and seed production. The methodologies used to discard variables are biometric tools that can be used successfully in future plant breeding programs, especially when a large number of traits and accessions are being evaluated. Although significant, geographic distance had a low association with morphological traits. This indicated the need to use other characteristics, such as forage and seed yield, in addition to molecular analysis. Our analyzes showed genetic variability in P. notatum for all the characteristics studied.(AU)
