RESUMO

PURPOSE: To explore the effect of milk fat globule-epidermal growth factor 8 (MFG-E8) on sepsis-induced acute kidney injury (SAKI). METHODS: Male C57BL/6 mice were randomized to control, sham, CLP, CLP+PBS, and CLP+rmMFG-E8 groups. SAKI was induced by cecal ligation and puncture (CLP). Recombinant mouse MFG-E8 (rmMFG-E8) (20 µg/kg) or PBS (vehicle) was administered intraperitoneally. Blood, urine and renal tissue were collected at 24 h after CLP. Blood samples were tested for serum kidney injury biomarker and cytokines. Urine samples were collected to detect KIM-1, and NGAL. Real-time PCR was tested for Bax and Bcl-2. TUNEL staining was used to determine renal apoptosis. Western blot was used to detect the expression of Bax, Bcl-2, and proteins in the NF-κB pathway. RESULTS: MFG-E8 alleviated SAKI by decreasing serum Cre, BUN, urine KIM-1 and NGAL and by mitigating renal pathological changes significant (p < 0.05). IL-1ß, IL-6, TNF-α were significantly inhibited by MFG-E8 (p < 0.05). Apoptosis induced by SAKI was markedly suppressed by MFG-E8. Finally, MFG-E8 attenuated the activation of the NF-𝜅B signaling pathway in SAKI. CONCLUSION: MFG-E8 has beneficial effects on SAKI, which may be achieved by inhibiting the NF-κB pathway.

UNLABELLED: Functional foods containing bioactive compounds of whey may play an important role in prevention and treatment of obesity. The aim of this study was to investigate the prospects of the biotechnological process of coacervation of whey proteins (CWP) in chitosan and test its antiobesogenic potential. METHODS: CWP (100 mg · kg · day) was administered in mice with diet-induced obesity for 8 weeks. The animals were divided into four groups: control normocaloric diet gavage with water (C) or coacervate (C-CWP), and high fat diet gavage with water (HF) or coacervate (HF-CWP). RESULTS: HF-CWP reduced weight gain and serum lipid fractions and displayed reduced adiposity and insulin. Adiponectin was significantly higher in HF-CWP group when compared to the HF. The level of LPS in HF-W group was significantly higher when compared to HF-CWP. The IL-10 showed an inverse correlation between the levels of insulin and glucose in the mesenteric adipose tissue in the HF-CWP group. CWP promoted an increase in both phosphorylation AMPK and the amount of ATGL in the mesenteric adipose tissue in HF-CWP group. CONCLUSION: CWP was able to modulate effects, possibly due to its high biological value of proteins. We observed a protective effect against obesity and improved the inflammatory milieu of white adipose tissue.

UNLABELLED: Crohn's disease (CD) is a chronic inflammatory disorder that primarily affects the intestines, resulting in breakage of the intestinal barrier, pathological inflammation and nutritional disorders that encompass from trace elements deficiency to severe malnutrition. Nutritional interventions either alone or associated to drug therapy may be effective to achieve and maintain inflammation remission. OBJECTIVE: To evaluate usual food intake as quantitative and qualitatively, in CD patients; and describe the effect of a supplement containing whey proteins and TGF- on their body composition. PATIENTS AND METHODS: Dietary intake was assessed considering 42 consecutive patients, followed in a tertiary center, and by using the 3-day food recall and food intake frequency questionnaire. Body composition was assessed previously and 8 weeks after supplementation with a diet containing whey proteins and TGF-ß (N = 22). RESULTS AND DISCUSSION: Considering carbohydrates and lipids, most patients had adequate dietary intake according recommendations. Protein, saturated fat, B12 vitamin and zinc intakes were higher than the recommended values. The dietary fiber, A, D, C and E vitamins, calcium, iron, folate, potassium and sodium intakes did not reach the recommended requirements in most patients. Patients supplemented with the whey protein and TGF-ß dietary presented a positive increment in their lean body mass, when compared to non-supplemented group. CONCLUSION: CD patients require nutritional orientation. Whey protein intake resulted in significant differences, such as improvement in Lean Body Mass and reduction in Fat percentage.

PURPOSE OF REVIEW: This review discusses the mechanisms of the dysfunctional gut during the critical illness and the possibility that an immunonutrient such as whey protein can play a role in better tolerance of enteral nutrition, also decreasing inflammation and increasing anti-inflammatory defenses. RECENT FINDINGS: Impaired gastric motor function and associated feed intolerance are common issues in critically ill patients. Some studies have been published with enteral nutrition enriched with whey protein as a dietary protein supplement that provides antimicrobial activity, immune modulation, improving muscle strength and body composition, and preventing cardiovascular disease and osteoporosis. SUMMARY: Early enteral feeding will enhance patient recovery and the use of enteral diets enriched with whey protein may play a role in these patients.

Obesity and type 2 diabetes mellitus (DM) have grown in prevalence around the world, and recently, related diseases have been considered epidemic. Given the high cost of treatment of obesity/DM-associated diseases, strategies such as dietary manipulation have been widely studied; among them, the whey protein diet has reached popularity because it has been suggested as a strategy for the prevention and treatment of obesity and DM in both humans and animals. Among its main actions, the following activities stand out: reduction of serum glucose in healthy individuals, impaired glucose tolerance in DM and obese patients; reduction in body weight; maintenance of muscle mass; increases in the release of anorectic hormones such as cholecystokinin, leptin, and glucagon like-peptide 1 (GLP-1); and a decrease in the orexigenic hormone ghrelin. Furthermore, studies have shown that whey protein can also lead to reductions in blood pressure, inflammation, and oxidative stress.

OBJECTIVE: The aim of this study was to investigate the effects of an early enteral formula containing whey protein, in comparison to a standard enteral formula containing casein as the protein source, on the levels of glutathione and inflammatory markers in aged patients with acute ischemic stroke. METHODS: Thirty-one elderly patients (12 males and 19 females; median age = 74 [range,65-90] y old) with ischemic stroke were randomized to receive early nasogastric feeding (35 kcal/kg/d and 1.2 g of protein/kg/d) with either a formula containing polymeric [corrected] casein (casein group, n =16) or another isocaloric and isonitrogenous formula containing hydrolyzed whey protein (WP group, n = 15) for 5 d. The primary endpoints of the study were the changes in the serum levels of glutathione peroxidase, C-reactive protein (CRP), and interleukin 6 (IL-6). RESULTS: Twenty-five patients completed the study (10 in the WP group and 15 in the casein group). Mortality was similar between groups (33%; P = 1.00) and was associated with higher serum IL-6 (73.7 ± 24.7 versus 16.6 ± 2.4 pg/dL; P = 0.04) and CRP (82.0 ± 35.6 versus 48.3 ± 14.5 mg/L; P = 0.02) levels. Albumin levels dropped from the first to the fifth feeding day only in the casein group (P < 0.01). Serum IL-6 decreased (62.7 ± 47.2 to 20.6 ± 10.3 pg/dL; P = 0.02) and glutathione increased (32.2 ± 2.1 to 39.9 ± 6.8 U/G Hb; P = 0.03) only in the WP group. Serum IL-6 was lower (P = 0.03) and glutathione was higher (P = 0.03) in whey protein-fed patients than in the casein group. CONCLUSION: Enteral formula containing whey protein may decrease inflammation and increase antioxidant defenses in elderly patients with ischemic stroke, compared to casein-containing formula.

Avaliou-se o efeito protetor das frações proteicas do soro do leite sobre as vilosidades intestinais de camundongos Balb/C, fêmeas, infectadas por Escherichia coli O157:H7. Foram utilizados 48 animais, distribuídos aleatoriamente em oito grupos de seis fêmeas cada um. Os animais dos grupos 1 e 2 (controles) receberam dieta AIN93G padrão; os dos grupos 3 e 4, AIN93G + alfalactalbumina; os dos grupos 5 e 6, AIN93G + betalactoglobulina e os dos grupos 7 e 8, AIN93G + concentrado proteico total e água ad libitum por sete dias. No dia zero, os animais dos grupos 2, 4, 6 e 8 foram inoculados, por meio de cânula de gavagem, com 0,5mL de E. coli O157:H7, na concentração de 7 x 10(10)UFC/mL. Os animais foram acompanhados clinicamente e sacrificados, no oitavo dia experimental. Verificou-se, por meio de exames histológicos e da morfometria, que as frações betalactoglobulina e alfalactalbumina exerceram efeito protetor sobre as vilosidades intestinais do jejuno distal e do íleo (P<0,05), respectivamente. O concentrado proteico total não demonstrou efeito protetor sobre as vilosidades intestinais.(AU)


The protective effect of protein fractions of whey on intestinal villi of Balb/C female mice infected with Escherichia coli O157: H7 was evaluated. A total of 48 animals were randomly distributed into eight groups of six females each. Animals in groups 1 and 2 (controls) received AIN93G standard diet; the groups 3 and 4, AIN93G + alpha-lactalbumin; while groups 5 and 6, AIN93G + beta-lactoglobulin; and groups 7 and 8, AIN93G + total protein concentrate and water ad libitum for seven days. On day zero, animals of groups 2, 4, 6, and 8 were inoculated by gavage tube with 0.5mL of E. coli O157: H7 at a concentration of 7 x 10(10)CFU/mL. The animals were clinically followed and sacrificed on the eighth day. It was verified by histological examination and morphometry that the beta-lactoglobulin and alpha-lactalbumin exerted a protective effect on the villi of the distal jejunum and ileum (P<0.05), respectively. The total protein concentrate showed no protective effect on the villi.(AU)
