Recent studies have shown the importance of subtropical forests as terrestrial carbon sinks and also their vulnerability to human disturbances and climate change. The Semi-deciduous Atlantic Forest presents large extensions replaced by productive uses, such as tree plantations, and forest remnants showing high levels of structural heterogeneity. No studies have performed carbon stock densities estimations in different pools in the region. We wonder how changes in forest structure and forest replacement by pine plantations affect ecosystem carbon stock densities in different pools and fluxes. We performed carbon estimates based on field data and compared closed (CF) and open (OF) canopy natural forest patches and Pinus taeda plantations at harvest age (PP). Structural changes in the natural forest had a profound effect on the ecosystem by halving the forest carbon stock while pulp-intended pine plantations reached the carbon stock of closed forest at harvest age. Main changes from CF to OF were a 55% decrease in the carbon of biomass and a 42% decrease in SOC. Instead, carbon stock density in biomass of PP was similar to CF but the carbon in fallen deadwood was 78% lower while in the litter layer was double; the SOC at 0-5 cm depth was 31% lower in PP than CF. Our study shows that structural changes in the natural forest halve the forest carbon stock while pulp-intended pine plantations can reach the closed forest carbon stock at harvest age. However, PP do not seem to be effective for carbon storage in the long term because of regular harvesting and clearing and their short-life products. Therefore, to effectively store the forest carbon, arresting deforestation, replacement and degradation of the original forest is crucial.

This study dealt with the influence of the temperature on the bacterial dynamics of two spontaneously fermented wheat sourdoughs, propagated at 21 ±â€¯1 °C (SD1) and 30 ±â€¯1 °C (SD2), during nine backslopping steps (BS1 to BS9). Proteobacteria was the only phylum found in flour. Escherichia hermannii was predominant, followed by Kosakonia cowanii, besides species belonging to the genera Pantoea and Pseudomonas. After one step of propagation, Clostridium and Bacillus cereus group became predominant. Lactobacillus curvatus was found at low relative abundance. For the second backslopping step, Clostridium was flanked by L. curvatus and Lactobacillus farciminis. From BS4 (6th day) onward, lactic acid bacteria (LAB) became predominant. L. farciminis overcame L. curvatus and remained dominant until the end of propagations for both sourdoughs. At 21 °C, Bacillus, Clostridium, Pseudomonas, and Enterobacteriaceae were gradually inhibited. At the end of propagation, SD1 harbored only LAB. Otherwise, the temperature of 30 °C favored the persistence of atypical bacteria in SD2, as Pseudomonas and Enterobacteriaceae. Therefore, the temperature of 21 °C was more suitable for sourdough propagation in Brazil. This study enhanced the knowledge of temperature's influence on microbial assembly and contributed to the elucidation of sourdough microbial communities in Brazil.

AIMS: To use the phage display technique to develop peptides with the capability to neutralize the cytotoxicity induced by Stx1 and Stx2 toxins produced by Shiga toxin-producing Escherichia coli (STEC). METHODS AND RESULTS: The phage display technique permitted the development of three peptides, named PC7-12, P12-26 and PC7-30, which bind to the globotriaosylceramide (Gb3) receptor for Shiga toxins produced by STEC. Moreover, these peptides were capable of competing efficiently with the Shiga toxins for binding to Gb3. The peptides described herein partially inhibited the Stx-induced cytotoxicity of cell-free filtrates of STEC O157 : H7 and purified Stx toxins in Vero cells. The inhibition of lethality induced by Stx toxins in mice indicated that peptide PC7-30 inhibited the lethality caused by Stx1 (2LD50) in mice. CONCLUSIONS: The phage display technique permitted the development of peptides that inhibited the cytotoxicity induced by Stx toxins in vitro. Peptide PC7-30 inhibited the lethality of Stx1 in vivo; this molecule would be a promising candidate for the development of therapeutic agents for STEC-related diseases in humans. SIGNIFICANCE AND IMPACT OF THE STUDY: The selection of Gb3, the common receptor for Stx1 and Stx2, may contribute to the development of efficient neutralizers for both toxins, and our approach would be an interesting alternative for the development of therapeutic molecules for the treatment of diseases caused by STEC strains.

Vaginal secretions are an important source of chemical signals, which affect ewes' attractiveness. Moreover, alterations of vaginal flora reduce sexual attractiveness of estrous ewes. As intravaginal sponges containing progestagens (widely used for estrous synchronization) affect vaginal flora, our aims were to determine if estrous ewes pretreated with intravaginal sponges were less attractive than ewes displaying spontaneous estrus, and if the addition of antibiotic to the sponge mitigated the decreased sexual attractiveness. Seventy-two estrous ewes were used in experiment 1: in 36, estrus was synchronized with commercial intravaginal sponges (50 mg medroxyprogesterone acetate for 14 days, group MAP1), whereas the other 36 were given a PGF2α analogue 19 to 20 days earlier and displayed spontaneous estrus (group C1). In experiment 2, 72 ewes were treated with intravaginal sponges for 14 days; for 36 ewes, the sponges contained 0.02 mg oxytetracycline (group Ox), whereas there was no antibiotic in the sponges for the remaining 36 ewes (group MAP2). In both experiments, sexual attractiveness was determined in 12 groups of six estrous ewes (three MAP1 vs. three C1, and three MAP2 vs. three Ox for Experiments 1 and 2, respectively) located in a 4 × 4 m pen. Courting and mating time that each ram spent with each ewe was recorded. After 5 min, the ewe with which the ram spent more time (most attractive ewe, ranked one, scale one to six) was taken out from the pen. The procedure was repeated until the ram ranked all six ewes, and repeated in the 12 groups in both experiments. In experiment 1, C1 ewes were more attractive than MAP1 ewes (ranks: 2.9 ± 0.3 vs. 4.1 ± 0.3, mean ± SEM, respectively; P < 0.002). In experiment 2, sexual attractiveness of MAP2 and Ox ewes was similar (3.5 ± 0.3 vs. 3.4 ± 0.3, respectively). We concluded that the use of intravaginal sponges impregnated with medroxyprogesterone acetate negatively affected ewes' sexual attractiveness, but this decrease was not mitigated by inclusion of a local antibiotic.

The aim of this work was to test 101 strains of E. coli for virulence factors associated with enterotoxigenic and enterohemorrhagic pathotypes of E. coli isolated from diarrheic and non-diarrheic calves. The virulence factors of E. coli Stx1 (Shiga toxin), Stx2, Ehly (Enterohemolysin), the eae gene, LT-II (heat-labile enterotoxin), STa (heat-stable toxin), and adhesins K99 and F41 were detected by PCR. Serogroups were determined by serological methods and Stx production was observed by biological assays in Vero cells. The frequency of the eae gene was higher in isolates from diarrheic calves (35/58, 60.3%) than in non-diarrheic calves (8/43, 18.6%; P 0.001). The gene for Stx1 occurred at high frequencies in the diarrheic strains (24/58, 41.3%) as well as in non-diarrheic (19/43, 44.2%) ones and all strains that were Stx positive by PCR showed cytotoxicity in Vero cells. Stx2 was found in ten strains, Ehly in eight strains, and LT-II in only two strains. Twenty-eight strains were negative for all of the PCR assays, including for F41 and K99 adhesins. The serogroups O7, O23, O4, O8, O153 and O156 were observed most frequently. Our results show that strains of E. coli isolated from cattle have similar virulence factors genes to strains isolated from cases of diseases in humans and may be a source of potentially pathogenic STEC for humans.

Intravaginal sponges (IS) impregnated with progestagens are widely used for oestrous synchronization in ewes. As progestogens depress the immuno response, the first aim was to determine whether medroxiprogesterone acetate (MAP) content affects the vaginal bacteria number (VBN) in IS-treated anoestrous ewes. The second aim was to compare the effectiveness of different antibiotic treatments to control the VBN increase caused by IS. In both experiments, IS were inserted during 14 days in anoestrous ewes. In the first, 11 ewes received commercial sponges (50 mg MAP), and 10 ewes received placebo sponges. For the second experiment, IS were inserted in three groups (n = 12/group), containing oxytetracycline im (20 mg/kg); injected into the sponge (0.02 mg), or control (no antibiotic). At sponge withdrawal, all ewes received 300 UI eCG. Mucous samples were collected from the vagina before sponge insertion, at sponge withdrawal, 24, 48 and 72 h later, and the VBN (colony-forming units per ml; CFU/ml) was counted after 48-h incubation. Medroxiprogesterone content did not affect VBN (log CFU/ml: 4.3 ± 0.2 vs 4.4 ± 0.2 with and without MAP, respectively). Bacterial number increased from 3.5 ± 0.2 at sponge insertion to 6.9 ± 0.1 at sponge withdrawal (p < 0.0001) and decreased the following day to 4.3 ± 0.2 (p < 0.0001). In the second experiment, VBN increased at sponge withdrawal (p < 0.0001) in all groups and decreased the following day (p < 0.0001). The CFU/ml at sponge withdrawal was lower in ewes treated with antibiotics (p < 0.0001), being even lower when local rather than systemic antibiotic was administered (log CFU/ml: 3.3 ± 1.8 vs 7.2 ± 1.8). The day of oestrous VBN was similar for all treatments and similar to that observed before sponge insertion. We concluded that MAP does not influence the increase in VBN, as the main effect is provoked by the sponge device itself, and local antibiotic treatment resulted in a lower bacterial growth than systemic treatments.

Enteric infections caused by the ingestion of contaminated water, especially by Escherichia coli, are important to define the virulence properties of these bacteria. Due to frequent infantile diarrhea in the city of Ouro Preto, Minas Gerais state, Brazil, the phenotypic and genotypic diarrheagenic properties of E. coli isolated from drinking water were studied. The culture supernatants of 39 (40 percent) among a total of 97 E. coli isolates from drinking water were positive by suckling mouse assay and induced cytotoxic effects on Vero cells. The enterotoxic and cytotoxic activities were present in the fraction with less than 10 kDa and were not lost when heated up to 60¨¬C and 100¨¬C for 30 minutes. PCR assays showed that among these 39 Vero cytotoxigenic E. coli, four (10.2 percent) were positive for ST II (estB) and two (5 percent) positive for ¥áHly (hlyA). Gene amplification of SLT (stx 1, stx 2), ST I (estA), LT (eltI, eltII), EAST1 (astA), EHly (enhly) and plasmid-encoded enterotoxin (pet) were not observed. This heat-stable cytotoxic enterotoxin of E. coli is probably a new putative diarrheagenic virulence factor, as a toxin presenting these characteristics has not yet been described.

The guidelines of the Brazilian National Health System (SUS) state that the municipal authorities are responsible for the vaccination of the population. The present study examined the types of immunobiologicals, amounts and reasons for their destruction and disposal by the Municipal Health Secretariat in the city of Bauru, São Paulo state, Brazil, between 2008 and 2009. This study comprises a descriptive, exploratory and retrospective work that employed search of bibliographic data, collection of secondary data from forms of immunobiologicals disposal and interview of the agent responsible for the disposal of these products in the municipality. It was observed that the total numbers of unused vaccines in 2008 and 2009 were similar (4523 and 4395, respectively), being the most discarded: the diphtheria, tetanus and pertussis (DTP) vaccine in 2008 and DTP, BCG and influenza in 2009. It was found that the amount of discarded vaccines could be reduced since the reasons for that were predictable. Moreover, the current study emphasized that although there is a municipal regulation for the disposal of immunobiologicals, the city still requires a better structure to handle such problem.
