RESUMO

The main feature of Fanconi anemia (FA) is the high sensitivity of the cells to the clastogenic agent, diepoxybutane (DEB). Thus, differential diagnosis of this syndrome can be made by cytogenetic analysis; adding DEB to lymphocytes in culture (DEB test) increases the number of chromosome breaks. Fanconi anemia cells have an abnormal cell cycle, with an increased frequency of cells arrested at G2. In order to determine if flow cytometry can be utilized for FA diagnosis, we cultivated lymphocytes with DEB and analyzed them for G2 accumulation. Lymphocytes cultivated for 72 h were labeled with CD3 antibody and propidium iodide for analysis of the cells in the G2 phase. Cultures of lymphocytes from two FA patients who were diagnosed by the DEB test and six control individuals with a negative DEB test had 55.26% (SD +/- 6.97) and 2.81% (SD +/- 0.22) cells arrested at G2, respectively. We conclude that flow-cytometry analysis of cells exposed to DEB can be useful for FA diagnosis.

RESUMO

Effects of heat processing and storage time (up to 70 days) on migration of bisphenol A (BPA) and bisphenol A-diglycidyl ether (BADGE) from can coatings into an aqueous food simulant were determined. Distilled water was canned in two types of Mexican cans: for tuna and for jalapeño peppers. Results showed that there is an effect of heat treatment on migration of both compounds. Storage time did not show any effect in BPA migration from tuna cans. There was an effect of storage time on BPA migration from jalapeño pepper cans. Results for BADGE migration were affected by its susceptibility to hydrolyze in aqueous simulants. BADGE concentration decreased, or was not detected, during storage in both types of cans. Migration levels for BPA and BADGE were within 0.6-83.4 and <0.25-4.3 microg/kg, respectively. Both were below European and Mercosur legislation limits. Other migrating compounds were detected, although no identification was performed.

There is a paucity of information regarding excretion of contaminants in human milk, due to experimental difficulties and until recently a general lack of interest. Because of the high fat content of milk and as its acidity is higher than that of plasma, nearly all liposoluble and basic agents consumed by the mother will be excreted in the milk. Distinction must be made between, on the one hand drugs and social toxicants such as smoking and alcohol, whose intake can be stopped or limited during pregnancy and lactation, and ecological toxicants present in a polluted environment to which the mother is exposed. Cases have occurred of heavy prenatal and postnatal intoxication of infants with hexachlorobenzene in Turkey and methylmercury in Iraq due to consumption of fungicide-treated seed wheat by pregnant and lactating mothers. Recent attention has been concentrated on contamination of milk with organochlorine compounds such as DDT and PCB's, that are found in many parts of the world. The heaviest contamination with DDT has been found in Guatemala, resulting in suckling infants consuming many times the Acceptable Daily Intake of this compound proposed by WHO, with unknown future effects.
